METHODS: Ten patients (eight males) wit...AIM: To evaluate the safety and feasibility of bone marrow cell (BMC) transplantation in patients with chronic liver disease on the waiting list for liver transplantation. METHODS: Ten patients (eight males) with chronic liver disease were enrolled to receive infusion of autologous bone marrow-derived cells. Seven patients were classified as Child-Pugh B and three as Child-Pugh C. Baseline assessment included complete clinical and laboratory evaluation and abdominal MRI. Approximately 50 mL of bone marrow aspirate was prepared by centrifugation in a ficoll-hypaque gradient. At least of 100 millions of mononuclear-enriched BMCs were infused into the hepatic artery using the routine technique for arterial chemoembolization for liver tumors. Patients were followed up for adverse events up to 4 mo. RESULTS: The median age of the patients was 52 years (range 24-70 years). All patients were discharged 48 h after BMC infusion. Two patients complained ofmild pain at the bone marrow needle puncture site. No other complications or specific side effects related to the procedure were observed. Bilirubin levels were lower at 1 (2.19 ± 0.9) and 4 mo (2.10 ± 1.0) after cell transplantation that baseline levels (238 ± 1.2). Albumin levels 4 mo after BMC infusion (3.73 ± 0.5) were higher than baseline levels (3.47 ± 0.5). International normalized ratio (INR) decreased from 1.48 (SD = 0.23) to 1.43 (SD = 0.23) one month after cell transplantation. CONCLUSION: BMC infusion into hepatic artery of patients with advanced chronic liver disease is safe and feasible. In addition, a decrease in mean serum bilirubin and INR levels and an increase in albumin levels are observed. Our data warrant further studies in order to evaluate the effect of BMC transplantation in patients with advanced chronic liver disease.展开更多

Methods Twenty-six patients with persistent systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), primary Sjogren's syndrome (pSS), or systemic sclerosis (SSc) who had been treated unsuccessfully with conventional treatment were enrolled in the trial in Peking Union Medical College Hospital from September 1999 to June 2004. The patients received HDIT with 200 mg/kg cyclophosphamide followed by an infusion of autologous stem cells that were CD34 selected. Disease acti- vity, adverse effect, hemopoietic and immune reconstitution, and time to recurrence of disease were monitored. Overall treatment related mortality was 7.7% (2/26) with 1 patient died of cytomegalovirus infection and an other of severe pneumonia. Relapse occurred in 3 SLE patients (17.6%) in 37, 26, and 19 months posttransplantation respectively, and 1 RA patient in 15 months posttransplantation. SLE Disease Activity Index (SLEDAI) scores of SLE survivors decreased significantly (P 〈 0.01). RA patients recorded a drop of Disease Activity Score 28 (DAS 28). The pSS patient remained symptoms free up to now, more than 50 months aider the transplantation. Conclusion HSCT can be performed relative safely in patients with severe autoimmune disease. Short-term effect of HSCT is promising. However treatment related mortality and relapse were observed in a subset of patients.展开更多

A case report and review of literature were written concerning autoimmune hepatitis in association with hepatitis A and other hepatotropic viruses. Soon a^er the administration of formalin-inactivated hepatitis A vaccine, a man who had recently recovered from an uncharacterized but self-limiting hepatitic illness, experienced a severe deterioration (AST 1687 U/L, INR 1.4). Anti-nuclear antibodies were detectable, and liver biopsy was compatible with autoimmune hepatitis. The observation supports the role of HAV as a trigger of autoimmune hepatitis. Studies in helper T-cell activity and antibody expression against hepatic proteins in the context of hepatitis A infection are summarized, and the concept of molecular mimicry with regard to other forms of viral hepatitis and autoimmunity is briefly explored.展开更多

This paper presents a reading of the seductive power of performance art as rooted in our theatrical nature. I will address performance art as an emancipated form of the theatrical, where by "theatrical" I mean a specific mode of presentation and a specific mode of perception: the mode of presentation of the self to the social and the mode of perception of the self through the social. Performance art, I will argue, is hardly an anomaly of our time. Rather, its source of disturbance and fascination lies in the natural, though excessive manifestation of our theatrical nature. By its appeal to the shocking, the perilous, or the mundane even, this form of art confirms what Paul Woodruff has addressed as "the necessity of watching and being watched." Performance art shows us the danger of self-presentation, the recognition of the other gaze as the self's greatest need and greatest fear. It needs no words. Transmission electron microscopy (TEM)image found that the self-lubricated carbide layer was a multi-phase material with carbides and graphite. As a result, theEDA-modified aluminum alloy had a negligible wear rate of ~2?10?4 mg/m (c. f. ~1.1?10?2 mg/m for aluminum alloy substrate).Notably, a new characteristic was found that the EDA-processed carbide layer was a soft magnet, which improved theelectromagnetic interference (EMI) shielding performance of the alloy.展开更多

The proportions of LCSCs in A549 cells and the cells from the third passage (A549-3rd) were compared. The expression of Bmil in LCSCs was silenced by intratumoral injection with lentivirus-delivered Broil small hairpin RNA (shRNA). RT-PCR and Western blot were used to test the mRNA and protein expressions of Broil in LCSCs. The protein level of p16INK4A was analyzed by Western blotting. The self- renewal and tumorigenicity ability of LCSCs were evaluated by counting the sphere formation rate in serum-free medium and the tumor formation rate in NOD/SCID mice. Results: In vivo passaging ofA549 cells under chemotherapy pressure enriched for LCSCs. The expression of Broil in LCSCs increased. Down-regulation of Bmil by RNA interference resulted in reduced self-renewal and tumorigenicity ability of LCSCs and paralleled the increased expression of p16INK4A, a Bmil target. Alcoholic liver diseases (ALDs) range from mild to more severe conditions, such as steatosis, steatohepatitis, fibrosis, cirrhosis, and hepatocellular carcinoma. The liver is enriched with innate immune cells (e.g. natural killer cells and Kupffer cells) and hepatic stellate cells (HSCs), and interestingly, emerging evidence suggests that innate immunity contributes to the development of ALDs (e.g. steatohepatitis and liver fibrosis). Indeed, HSCs play a crucial role in alcoholic steatosis via production of endocannabinoid and retinol metabolites. This review describes the roles of the innate immunity and HSCs in the pathogenesis of ALDs, and suggests therapeutic targets and strategies to assist in the reduction of ALD.展开更多

The excavator was developed to aut...Consider the design and implementation of an electro-hydraulic control system for a robotic excavator, namely the Lancaster University computerized and intelligent excavator (LUCIE). The excavator was developed to autonomously dig trenches without human intervention. One stumbling block is the achievement of adequate, accurate, quick and smooth movement under automatic control, which is difficult for traditional control algorithm, e.g. PI/PID. A gain scheduling design, based on the true digital proportional-integral-plus (PIP) control methodology, was utilized to regulate the nonlinear joint dynamics. Simulation and initial field tests both demonstrated the feasibility and robustness of proposed technique to the uncertainties of parameters, time delay and load disturbances, with the excavator arm directed along specified trajectories in a smooth, fast and accurate manner. The tracking error magnitudes for oblique straight line and horizontal straight line are less than 20 mm and 50 mm, respectively, while the velocity reaches 9 m/min.展开更多

(Sapotaceae family), endemic to Morocco, is a multipurpose tree with an important ecological and socio-economical interest. Nevertheless, the sustainability of the agro-forestry system is now threatened by overgrazing and over-exploitation leading to an alarming decline in the number of trees. In the present study, inter-simple sequence repeats (ISSR) markers were used to evaluate the genetic variation and to assess the genetic diversity distribution within and among 17 argan populations growing naturally under semi-arid and arid climate. Thus, a total of seven primers generated 260 well-defined bands, with an average of 37.14 bands per primer. Studied populations showed a relatively high level of genetic diversity at species level and low level of genetic diversity at population level. The percentage of polymorphic bands, Nei's gene diversity and Shannon's information index at population and species level were 30.15%, 0.164, 0.217 and 98.8%, 0.172, 0.293, respectively. A relative low level of genetic differentiation (Gst = 0.39) was in agreement with the results obtained from the analysis of molecular variance (AMOVA). Estimated gene flow was Nm = 0.781 among populations. Mantel test revealed a non-significant correlation between genetic and geographic distance (r = 0.065, P 〈 0.05), suggesting that the geographic distribution is not the major factor that shaped the present population genetic structure. The results can help preserve A. spinosa L. tree as an endangered species, however, refining these finding using co-dominant markers is recommended in order to establish conservation strategies.展开更多

An example of this behavior is the perennially of the so-called Inca wall in Peru, which despite having suffered several earthquakes over time has remained stable without collapsing. This article presents the research carried out on stone masonry wails with dry joint, without mortar, subject to a seismic action. In order to understand the behavior of the masonry without mortar, it designs a Grid mode/ of Finite Elements. From the results, it is concluded that these walls with a certain thickness have ductility that allows them to withstand high displacement and rotation values, thus accommodating the movement of the earth subject to an earthquake. The individual stone blocks move relative to each other through rotations and displacements, which are processed in the free joints of any mortar. The joints work as energy sinks. The free movements in the joints dissipate the energy transmitted by the earthquake, not causing in this way the rupture of the stone blocks. The goal of this article is to understand the p importance of lack of mortar in the seismic behavior of the mansonry.展开更多

Gay, an endemic Saharan species. The antioxidant assays revealed that crude extract showed strong DPPH scavenging activity (IC50 = 49.357 μg/mL) but in reducing power and hydrogen peroxide scavenging activity, butanolic and ethyl acetate fractions have a potent ferrous ion-chelating ability in particularly the butanolic fraction (63.18% reduced power at 50 μg/mL) and a powerful scavenging activity on hydrogen peroxide in particularly ethyl acetate fraction (IC50 = 0.205 μg/mL). The phenolic compounds of Tamarix pauciovulata leaves were analyzed by HPLC-UV. The major phenolic of leaf extracts are syringic acid (1.07 g/100g), quercetin (34.1 mg/100g), kaempferol (5.77 mg/100g), isorhamnetin (5 mg/100g). Others phenols were identified such as isoquercetin, catechin, epicatechin and its derivatives. Results indicated that the leaves extracts from Tamarix pauciovulata have great capacities to prevent diseases caused by the overproduction of radicals and can become important source of dietary compounds with health protective potential.展开更多

Down-regulation of Bmil by RNA interference resulted in reduced self-renewal and tumorigenicity ability of LCSCs and paralleled the increased expression of p16INK4A, a Bmil target. Conclu- sion: Broil regulates self-renewal and tumorigenicity of LCSCs by silencing some target genes, including p16INK4A.

文摘Classical interferon-alpha has been shown to be correlated with the development of a variety of autoimmune disorders. A 38 year-old female patient developed simultaneously diabetic ketoacidosis and hyperthyroidism 5 mo following initiation of treatment with pegylated interferon-α and ribavirin for chronic hepatitis C. High titers of glutamic acid decarboxylase, antinuclear and thyroid (thyroid peroxidase and thyroglobulin) antibodies were detected. Antiviral treatment was withdrawn and the patient was treated with insulin for insulin-dependent diabetes mellitus and propranolol for hyperthyroidism. Twelve months after cessation of pegylated interferon-α therapy the patient was euthyroid without any medication but remained insulin-dependent.

文摘Constant alcohol consumption is a major cause of chronic liver disease, and there has been a growing concern regarding the increased mortality rates worldwide. Alcoholic liver diseases (ALDs) range from mild to more severe conditions, such as steatosis, steatohepatitis, fibrosis, cirrhosis, and hepatocellular carcinoma. The liver is enriched with innate immune cells (e.g. natural killer cells and Kupffer cells) and hepatic stellate cells (HSCs), and interestingly, emerging evidence suggests that innate immunity contributes to the development of ALDs (e.g. steatohepatitis and liver fibrosis). Indeed, HSCs play a crucial role in alcoholic steatosis via production of endocannabinoid and retinol metabolites. This review describes the roles of the innate immunity and HSCs in the pathogenesis of ALDs, and suggests therapeutic targets and strategies to assist in the reduction of ALD.

Macroscopic quantum coherence of the domain walls is demonstrated in terms of the instantons.

文摘Consider the design and implementation of an electro-hydraulic control system for a robotic excavator, namely the Lancaster University computerized and intelligent excavator (LUCIE). The excavator was developed to autonomously dig trenches without human intervention. One stumbling block is the achievement of adequate, accurate, quick and smooth movement under automatic control, which is difficult for traditional control algorithm, e.g. PI/PID. A gain scheduling design, based on the true digital proportional-integral-plus (PIP) control methodology, was utilized to regulate the nonlinear joint dynamics. Simulation and initial field tests both demonstrated the feasibility and robustness of proposed technique to the uncertainties of parameters, time delay and load disturbances, with the excavator arm directed along specified trajectories in a smooth, fast and accurate manner. The tracking error magnitudes for oblique straight line and horizontal straight line are less than 20 mm and 50 mm, respectively, while the velocity reaches 9 m/min.
文摘Argania spinosa L. (Sapotaceae family), endemic to Morocco, is a multipurpose tree with an important ecological and socio-economical interest. Nevertheless, the sustainability of the agro-forestry system is now threatened by overgrazing and over-exploitation leading to an alarming decline in the number of trees. In the present study, inter-simple sequence repeats (ISSR) markers were used to evaluate the genetic variation and to assess the genetic diversity distribution within and among 17 argan populations growing naturally under semi-arid and arid climate. Thus, a total of seven primers generated 260 well-defined bands, with an average of 37.14 bands per primer. Studied populations showed a relatively high level of genetic diversity at species level and low level of genetic diversity at population level. The percentage of polymorphic bands, Nei's gene diversity and Shannon's information index at population and species level were 30.15%, 0.164, 0.217 and 98.8%, 0.172, 0.293, respectively. A relative low level of genetic differentiation (Gst = 0.39) was in agreement with the results obtained from the analysis of molecular variance (AMOVA). Estimated gene flow was Nm = 0.781 among populations. Mantel test revealed a non-significant correlation between genetic and geographic distance (r = 0.065, P 〈 0.05), suggesting that the geographic distribution is not the major factor that shaped the present population genetic structure. The results can help preserve A. spinosa L. tree as an endangered species, however, refining these finding using co-dominant markers is recommended in order to establish conservation strategies.

文摘An adaptive output feedback control was proposed to deal with a class of nonholonomic systems in chained form with strong nonlinear disturbances and drift terms. The objective was to design adaptive nonlinear output feedback laws such that the closed-loop systems were globally asymptotically stable, while the estimated parameters remained bounded. The proposed systematic strategy combined input-state-scaling with backstepping technique. The adaptive output feedback controller was designed for a general case of uncertain chained system. Furthermore, one special case was considered. Simulation results demonstrate the effectiveness of the proposed controllers.
文摘Stone structures with dry joints, that is, without mortar, have shown a surprising behavior when earthquakes occur. An example of this behavior is the perennially of the so-called Inca wall in Peru, which despite having suffered several earthquakes over time has remained stable without collapsing. This article presents the research carried out on stone masonry wails with dry joint, without mortar, subject to a seismic action. In order to understand the behavior of the masonry without mortar, it designs a Grid mode/ of Finite Elements. From the results, it is concluded that these walls with a certain thickness have ductility that allows them to withstand high displacement and rotation values, thus accommodating the movement of the earth subject to an earthquake. The individual stone blocks move relative to each other through rotations and displacements, which are processed in the free joints of any mortar. The joints work as energy sinks. The free movements in the joints dissipate the energy transmitted by the earthquake, not causing in this way the rupture of the stone blocks. The goal of this article is to understand the p importance of lack of mortar in the seismic behavior of the mansonry.
文摘This research presents complete phenolic compounds and biological activity of Tamarix pauciovulata J. Gay, an endemic Saharan species. The antioxidant assays revealed that crude extract showed strong DPPH scavenging activity (IC50 = 49.357 μg/mL) but in reducing power and hydrogen peroxide scavenging activity, butanolic and ethyl acetate fractions have a potent ferrous ion-chelating ability in particularly the butanolic fraction (63.18% reduced power at 50 μg/mL) and a powerful scavenging activity on hydrogen peroxide in particularly ethyl acetate fraction (IC50 = 0.205 μg/mL). The phenolic compounds of Tamarix pauciovulata leaves were analyzed by HPLC-UV. The major phenolic of leaf extracts are syringic acid (1.07 g/100g), quercetin (34.1 mg/100g), kaempferol (5.77 mg/100g), isorhamnetin (5 mg/100g). Others phenols were identified such as isoquercetin, catechin, epicatechin and its derivatives. Results indicated that the leaves extracts from Tamarix pauciovulata have great capacities to prevent diseases caused by the overproduction of radicals and can become important source of dietary compounds with health protective potential.
