This study was designed to investigate the effects on pharmaceutical characteristics and cell uptake PEI after a long-circulation modification with poly(ethylene glycol) (PEG). DFA or ODN reacted with PEI or PEI-PEG to form polyelectrolyte complexes. Surface characters of these complexes and the retardation of ODN by PEI and PEI-PEG were evaluated. The uptake rates of DFA/PEI and DFA/PEI-PEG complexes by MCF-7 cells were evaluated by flow cytometry. Confocal laser scanning microscopy was utilized to visualize the internalization of these complexes. ODN/PEI complex showed the dependence of their size and ξ potential on the N/P ratio. ODN/PEI-PEG complex were much less affected by N/P ratio and their size was around 30 100 nm. PEI and PEI-PEG retarded ODN even at N/P ratio as low as 4, and complete retardation was found at N/P ratio of 8. The uptake rate by MCF-7 cells was direct correlated to the DFA concentration and incubation time, and the uptake rate could exceed 99% under the selected condition. The results in this study showed that PEI self-assembly polyelectrolyte complex after stealth or long circulation modification may increase the ability as a gene vector to delivery genes into cells.展开更多

Approximately one third of its flora is endemic. The Muscari muscarimi which belongs to the Liliaceae family is one of its most prominent endemic geophyte plants which originates in southwestern of Turkey where it is found in rocky places like Ermenek, Ba?yayla (Büyükkarap?nar village), Sar?veliler. This species is the most sweetly scented member of the Liliaceae family, having something of the musk scent. Also, this plant locally is used in traditional medicine as antirheumatic, stomachic, diuretic and expectorant. In addition to this, it has also been used as food for humans and animals, ornamental plants in gardens. Morphologically, it has 3-6 linear-lanceolate, grayish-green leaves per plant and bulbs of 2-4 cm in diameter with thick fleshy perennial roots which delve down into the rocky ground of their natural habitats. The chemical is composed of polysaccharides, homoisoflavanons, glycosides etc. Main components of M. muscarimi were identified as (E)-β-ocimene (t-36%), methyl salicylate (1-21%), E-methyl isoeugenol (4-22%) and benzyl benzoate (7-56%). The picking up of the plant bulbs from their natural habitats is prohibited, in accordance with international agreements for the protection of endangered geophytes. The overly habitat destruction in native growing area threatened the existence of the species categories. It is known that in vitro bulblet production in Muscari muscarimi is low. This is difficult for its cultivation in native Ermenek area districts. Recent works show that the culturing immature Musari muscarimi embryos on different growth media could be considered as a useful method for in vitro propagation. As a result, the geophytes, such as M. muscarimi, are eligible widely for perfume production and in the pharmaceutical industry as well as for ornamental flowers. Because of this plant economic importance, bulbs growing and cultivation in their natural habitats could be discussed in this study.展开更多

IR and 1^H-NMR were used to characterize the products. Then Fc-GSH was immobilized on the surface of gold electrode. Cyclic votammetry (CV) was adopted to investigate the electrochemical properties of this Fc-GSH modified electrode in the absence and presence of Cd^2+ aqueous solutions. The peak oxidation potential (Ea) and reduction potential (Ec) of Fc-GSH modified electrode were observed at Ea= 0.74 V and Ec= 0.64 V (vs Ag/AgCl) before the accumulation of Cd^2+. This redox process is a monoelectron chemical reaction. The anodic shift is about 80 mV in the presence of 20 nmol/L of Cd^2+ aqueous solution. Moreover, this shift is in proportion to the concentration of Cd^2+ when the concentration of Cd^2+ is lower than 20 nmol/L. So the modified electrode can be used as probes to detect cadmium ions with the limit of 0.1 nmol/L by cyclic voltammetry.展开更多
