目的获得红芪多糖纯品并且得到其单糖组成。方法通过分步醇沉法和凝胶柱色谱获得红芪多糖纯品,用气相色谱法测其组成。结果红芪多糖1和2经Sephadex G-200纯化后分别得到两个组分,红芪多糖3通过Sephadex G-25得到两个组分;红芪多糖1,2,3... 目的获得红芪多糖纯品并且得到其单糖组成。方法通过分步醇沉法和凝胶柱色谱获得红芪多糖纯品,用气相色谱法测其组成。结果红芪多糖1和2经Sephadex G-200纯化后分别得到两个组分,红芪多糖3通过Sephadex G-25得到两个组分;红芪多糖1,2,3均由鼠李糖、阿拉伯糖、木糖、葡萄糖、半乳糖五种单糖组成。结论分步醇沉法获得的三种多糖1,2,3经Sephadex G-200和Sephadex G-25纯化可获得红芪多糖纯品,GC法测其组成方法简便可行。 展开更多

AIM:To investigate the effect of severe acute pan- creatitis(SAP)on pharmacokinetics of Da-Cheng-Qi Decoction(DCQD)components in rats. METHODS:Rats were divided into SAP group and sham-operation group as a control group(n=6). Rhein,chrysophanol,rheochrysidin,magnolol,hesperidin and naringin in DCQD were quantified in rat serum by high performance liquid chromatography tandem mass spectrometry for studying their pharmacokinetics. RESULTS:Early absorption of each DCQD component was tended to degrade in SAP group after treatment with DCQD by gavage.The Cmax(chrysophanol,P= 0.0059;rheochrysidin,P=0.0288;magnolol,P= 0.0487;hesperidin,P=0.0277;naringin,P=0.0023) and AUC(rhein,P=0.0186;chrysophanol,P=0.0013; magnolol,P=0.001;hesperidin,P=0.0081;naringin, P=0.0272)of DCQD component were obviously lower in SAP group than in control group.The T1/2α of chrysophanol and rheochrysidin(P=0.0467 and 0.0005,respectively)and Tmax of chrysophanol and rheochrysidin(P=0.0101 and 0.0037,respectively) lasted longer in SAP group than in control group. CONCLUSION:SAP can significantly impact the ab-sorption of DCQD components in rats and their phar-macokinetic parameters. 展开更多

Tribological characteristics and self-repairing effect of hydroxy-magnesium silicate (HMS) dispersed in lubricant oil on steel-to-steel friction pairs with various surface roughness were analyzed.The friction-reduction,anti-wear and self-repairing performance of various surface roughness friction pairs were examined by friction testing machine.An operation comparison was made between SJ10W-40 lubricant with and without HMS.The surface morphology and elementary composition of the grinding cracks were analyzed by scanning electron microscope (SEM) and energy dispersive spectrometer (EDS).The results show that the lubrication state changes from boundary lubrication into mixed lubrication after operation in lubricant with HMS.The friction-reduction,anti-wear and self-repairing performance of the friction pairs with various surface roughness are distinctly different.There is a repairing film whose material is different from substrate material on the grinding cracks.In addition,Si,Mg,O,Al and other elements are deposited on the repairing film which contains nanocrystals of these elements.And HMS self-repairing material possesses superior performance of friction-reduction,anti-wear and self-repairing effects. 展开更多

Multiplexed immunohistochemistry/fluorescence(mIHC/IF)in combination with multispectral unmixing is a novel multitarget histopathological staining and imaging technique.By simultaneously revealing expression level and spatial information for up to eight biomarkers in situ,in addition to a nuclear stain within a single formalin-fixed paraffin-embedded(FFPE)tissue section,this technology can analyze the phenotype,abundance,morphology and intercellular relationship of cells while providing statistically significant results.In recent years,technical improvements have brought new insight into mIHC/IF and multispectral imaging approaches,which have been successfully applied in the field of cancer immunotherapy,specifically in regard to tumor microenvironment research,immunotherapy drug discovery,and prognostic and metastatic risk evaluation.This review highlights the principle,workflow,advantages and disadvantages of the technology,and discusses the latest applications of mIHC/IF-based imaging technology in the field of TME-related research and immunotherapy drug discovery. 展开更多
