PP_(333)调控黄瓜子叶节培养物的花芽形态分化

应用离体黄瓜子叶节花芽分化实验系统,研究了PP333在黄瓜花芽分化进程中的作用.组织培养结果显示,0.3mg·L-1PP333处理显著促进离体黄瓜子叶节培养物的花芽分化,直接花形成率由对照的16.86%升高至36.02%,直接花芽/总芽的比值达到最... 应用离体黄瓜子叶节花芽分化实验系统,研究了PP333在黄瓜花芽分化进程中的作用.组织培养结果显示,0.3mg·L-1PP333处理显著促进离体黄瓜子叶节培养物的花芽分化,直接花形成率由对照的16.86%升高至36.02%,直接花芽/总芽的比值达到最高,约为对照的1.7倍;营养芽形成率和营养芽/总芽的比值均降至最低,分别为对照的72%和59%.组织切片结果显示,0.3mg·L-1PP333处理明显提高黄瓜子叶节培养物的花芽原基百分率和花芽原基/总原基的百分率,增幅分别为42%和50%,而营养芽原基百分率则较对照降低约47%.实验结果发现,适宜浓度的PP333处理可以促进培养物所生成原基向花属性方向发育,是PP333处理促进黄瓜子叶节培养物花芽形成的主要原因. 展开更多

连续多年观察青钱柳开花,分析了温度、光照、降水、树高、胸径和花龄对青钱柳幼树花性别分化的影响。结果表明,青钱柳的雌、雄花出现时间与花前积温有关,雄花始终比雌花早出现5-7 d,雌、雄花期能相遇;青钱柳幼树有雌花株、雄花株、雌雄... 连续多年观察青钱柳开花,分析了温度、光照、降水、树高、胸径和花龄对青钱柳幼树花性别分化的影响。结果表明,青钱柳的雌、雄花出现时间与花前积温有关,雄花始终比雌花早出现5-7 d,雌、雄花期能相遇;青钱柳幼树有雌花株、雄花株、雌雄花同株多种花性别类型,其中雌花株出现的概率与前一年的降水量和开花前6个月的光照存在显著的正相关关系,其他花性别类型个体与温度、光照和降水关系暂不明显;不同花性别类型植株胸径和树高均值大小为:两性花株>雌雄花同株>雌花株>雄花株>未开花株;青钱柳幼树花性别转变现象普遍,大部分青钱柳首次只开雌花,第2、3年开花时变成雌雄花同株类型,雄花株比例随花龄增长呈下降趋势,雌雄花同株比例增大;青钱柳开花后会出现不同程度的停开花现象。 展开更多

【目的】鉴定向日葵中花发育相关的HaYABBY基因,有助于探究其在向日葵花发育过程中的生物学功能和调控机制,为向日葵品种选育提供重要的分子线索。【方法】基于向日葵基因组和转录组数据,使用生物信息学方法,分析向日葵YABBY基因家族成... 【目的】鉴定向日葵中花发育相关的HaYABBY基因,有助于探究其在向日葵花发育过程中的生物学功能和调控机制,为向日葵品种选育提供重要的分子线索。【方法】基于向日葵基因组和转录组数据,使用生物信息学方法,分析向日葵YABBY基因家族成员的染色体定位、系统进化关系、基因结构、保守基序、蛋白结构及理化性质、启动子顺式作用元件和组织表达模式等,并通过RT-qPCR分析HaYABBY基因在WT向日葵和花分生组织决定和花器官分化缺陷突变体cb1中st2-st8时期的表达差异。【结果】在向日葵基因组中鉴定到12个YABBY基因家族成员,分属于4个亚家族,分布在8条染色体上,并且同一亚家族中成员在基因结构和保守基序组成上高度相似。其中8个基因HaYABBY1a、HaYABBY1c、HaYABBY3、HaYABBY4b、HaYABBY5a、HaYABBY5b、HaCRABS CLAWa、HaCRABS CLAWb在向日葵花发育时期特异性高表达。在这8个基因中,HaYABBY3、HaYABBY5a和HaYABBY5d在突变体cb1花发育后期表达上调,且与WT花发育早期水平相当;而HaYABBY4b和HaCRABS CLAWa在cb1中的表达被强烈抑制。顺式作用元件分析显示,HaYABBY均含有多个启动子顺式作用元件,部分元件仅存在于特定基因中。其中,胚乳表达元件存在于HaCRABS CLAWa和HaYABBY4a中;生长素响应元件存在于HaCRABS CLAWa、HaCRABS CLAWb、HaYABBY4a、HaYABBY4b和HaYABBY5c中;分生组织表达元件存在于HaYABBY1a、HaYABBY1c、HaYABBY4b、HaYABBY5a和HaYABBY5d中;赤霉素响应元件存在于HaCRABS CLAWa、HaYABBY5b、HaYABBY5c和HaYABBY5d中;种子特异性调控元件存在于HaYABBY5a、HaYABBY5c和HaYABBY5d中。【结论】HaYABBY3、HaYABBY4b、HaYABBY5a、HaYABBY5d和HaCRABS CLAWa等5个HaYABBY基因在花分生组织的转变和花器官发生过程中可能发挥重要作用,它们在花分生组织转变和花器官发生调控中可能受到生长素响应因子ARF、赤霉素响应因子MYB以及MADS-box等关键因子调节。 展开更多

A gene sequence coding for the precursor of Galanthus nivalis agglutinin (GNA) was modified by site-directed mutagenesis to change very low usage bias codons to higher usage bias ones for improvement of the gene expression in transgenic tobacco (Nicotiana tabacum L.) plants. Results from Western blot analysis of some of the transgenic tobacco plants showed that the expression level of GNA in plants transformed with the modified gene GNA34m reached 0.25% of total soluble proteins, while that of the GNA34 gene transgenic plants was 0.17%. Since the GNA expression level increased, the aphid resistance of GNA34m transgenic plants were also enhanced significantly as judged by a 71.0% aphid population inhibition in insect bioassay of GNA34m transformed plants and 63.7% for the plants transformed with the natural GNA34 gene. 展开更多

Transcription analysis of peloric mutants of Phalaenopsis orchids derived from tissue culture 被引量:21

In this study, wild... Tissue culture has been widely used for mass propagation of Phalaenopsis. However, somaclonal variation occurred during micropropagation process posed a severe problem by affecting product quality. In this study, wild type and peloric flower buds of Phalaenopsis hybrids derived from flower stalk nodal culture were used for cDNA-RAPD and cDNA suppression subtractive hybridization analyses in order to study their genetic difference in terms of expressed sequence tags. A total of 209 ESTs from normal flower buds and 230 from mutants were sequenced. These ESTs sequences can be grouped into several functional categories involved in different cellular processes including metabolism, signal transduction, transcription, cell growth and division, protein synthesis, and protein localization, and into a subcat- egory of proteins with unknown function. Cymbidium mosaic virus transcript was surprisingly found expressed fre- quently in the peloric mutant of P. Little Mary. Real-time RT-PCR analysis on selected ESTs showed that in mutant flower buds, a bZIP transcription factor (TGA1a-like protein) was down-regulated, while up-regulated genes include auxin-regulated protein kinase, cyclophilin, and TCP-like genes. A retroelement clone was also preferentially expressed in the peloric mutant flowers. On the other hand, ESTs involved in DNA methylation, chromatin remodeling and post- transcriptional regulation, such as DNA methyltransferase, histone acetyltransferase, ERECTA, and DEAD/DEAH RNA helicase, were enriched in normal flower buds than the mutants. The enriched transcripts in the wild type indicate the down regulation of these transcripts in the mutants, and vice versa. The potential roles of the analyzed transcripts in the development of Phalaenopsis flowers are discussed. 展开更多

Emplacement and deformation of Shigujian syntectonic granite in central part of the Dabie orogen:Implications for tectonic regime transformation 被引量:5

The Shigujian pluton is a gneissic quartz monzonite located in Tiantangzhai area in central part of the Dabie orogen.Anisotropy of magnetic susceptibility(AMS) data show that most magnetic foliations dip steeply to southeast.About 85% of sampling points dip from 40° to 90°.Magnetic foliations are generally parallel to the foliations measured in the field.The pluton has NWW-SEE trending lineations in the southeast and NE-SW trending lineations in central part and north,but the lineations plunge to SW in central part and to NE in the north.All plunges are moderate.The anisotropy degree(P) is between 1.065 and 1.532 and the shape parameter(T) is between 0.005 and 0.694.A Flinn diagram of the magnetic fabrics shows that the value of K is less than 1.The analysis of AMS suggests that the pluton was emplaced and deformed under a SE-NW compressional stress regime.The analysis of quartz C-axis fabrics indicates that the pluton was deformed under compressional stress and deformation temperatures range from 400 to 500℃.Microstructures indicate that the pluton is deformed in near solidus conditions and the pluton is a synkinematic intrusion.The emplacement of the Shigujian granite is inferred to have taken place syntectonically.The zircon U-Pb dating of the granite suggests that the pluton was intruded at 141±2.3 Ma.By synthesizing all data,it seems that the Shigujian pluton was emplaced in a compressional environment and the transformation time of the Dabie orogen from compression to extension took place after 141 Ma.The structural evolution of the Dabie orogen was controlled by the Pacific tectonic domain when the Shigujian pluton was emplaced,whereas the adjacent Tiantangzhai complex massif is the result of an extensional environment. 展开更多
