[Method] With total RNA extracted from buckwheat as the template, CHS cD- NA sequence was cloned from buckwheat by using RACE technology and CODEHOP primer design method, the full length gene was obtained by primers which were de- signed for amplification of full-length gene sequence with buckwheat DNA template. Clustalxl.81 and MEGA4 software were used for sequence analysis and construction of phylogenetic tree; NCBI Blastn and Biastp programs were applied for homology analysis of nucleic acid and protein. [Result] Bioinformatics analysis showed that the full length of this gene is 1 906 bp, containing a 463 bp intron sequence and a 1 188 bp coding region, encoding 395 amino acids. Blastn sequence alignment revealed that the CHS gene sequence obtained in this study shared 86% homology with the CHS gene of closely related species. [Conclusion] This study laid the foundation to clarify molecular basis of the synthesis of buckwheat bioflavanoids and explore an effective way to improve the content of buckwheat bioflavanoids.展开更多

METHODS: Human gastric cancer lines SGC-7901,MKN-45, MKN-74 were treated with OM in the absenceand presence of NM-3. The inhibitory rates weredetected by MTT assay. Synergistic effect of OM andNM-3 on the growth of survivin, bcl-2, bax and p53 inSGC-7901 cells were examined by semiquantitative RT-PCR and Western blotting, and their growth inhibitoryeffects were also observed on SGC-7901 tumor xenograftin nude mice.RESULTS: OM combined with NM-3 exhibited asynergistic inhibitory effect on the growth of SGC-7901,MKN-45 and MKN-74 cells in a time-dependent manner.Twenty-four hours after treatment with OM, NM-3 aloneand their combination, mRNA expression of survivin andbcl-2 in SGC-7901 cells decreased, p53 mRNA expressionincreased. OM (4 g/L) combined with NM-3 significantlyincreased the expression of p53 mRNA and decreasedthe expression of survivin and bcl-2 compared witheither agent alone (193% ± 34% vs 129% ± 12%;44% ± 18% vs 92% ± 18%; 36 ± 17% vs 93% ± 23%,P < 0.05). Western blotting showed that the synergisticeffect of OM and NM-3 on protein translation of survivin,bcl-2 and p 53 was in accordance with their mRNAs.Furthermore, OM/NM-3 combination obviously exhibitedantitumor growth effect in xenografted human gastriccancer cells SGC-7901 compared with either agent alone.CONCLUSION: OM combined with NM-3 has synergisticinhibitory effects on human gastric cancer cells in vitro and can suppress the growth of xenografted human gastric cancer cells SGC-7901 in vivo.展开更多

The growth traits of each plant growing in 7 different matrix materials were investigated. The plant height, crown width and chlorophyl content of each plant were mea-sured. The results showed that the best substrate ratio was peat soil∶vermiculite=2∶1 for C. gueilinensis, L. pauciflorus and H. henryi; peat soil∶perlite∶vermiculite = 2∶1∶1 for S. ionantha; peat soil∶vermiculite∶yel ow mud=2∶1∶1 for S. speciosa; peat soil∶per-lite∶vermiculite∶yel ow mud=2∶1∶1∶1 for A. acuminatus.展开更多

Sterilized for 6 rain with 0.1% (w/v) mercuric chloride solution is an essential sterilization method for stems. June and July proved to be better for material taking, followed by May. The callus formation rate of balsam pears reached the highest when medium added with IBA 1.0mg/L and BA 2.0 mg/L, while the optimum medium for adventitious bud induction was MS added with IBA 0.5mg/L and BA 4.0 mg/L. And it will provide theoretical and technical basis for rapid propagation and breeding.展开更多

In order to establish a more perfect regeneration system from anther cultuer, the effects of low temperature pretreatment, 2,4-dichlorophenoxyacetic acid (2,4-D), vitamin C (Vc) and silver nitrate (AgNO3) on callus induction and browning in anther culture of balsam pear (Momordiea charantia L.) were investigated. The results showed that after pretreatment at 4 ℃ for 1 day, callus induction rate was the highest and browning rate was the lowest. Anthers on MS medium supplemented with 2,4-D 0.5 mg/L formed more and better callus. The medium supplemented with Vc or AgNO3 was advantageous to the induction of callus and reduction of browning. When cultured on medium supplemented with 50 mg/L Vc or 5 mg/L AgNO3, callus induction rate was the highest and browning rate was rather low.展开更多
