宿主口服益生菌的定性和局部检测具有重要意义。本研究通过设计株水平特异性荧光原位杂交(FISH)探针来研究鼠李糖乳杆菌在宿主肠道中的定殖。根据鼠李糖乳杆菌Probio-M9和其它相关物种的基因组序列,分别设计了鼠李糖乳杆菌种水平特异性... 宿主口服益生菌的定性和局部检测具有重要意义。本研究通过设计株水平特异性荧光原位杂交(FISH)探针来研究鼠李糖乳杆菌在宿主肠道中的定殖。根据鼠李糖乳杆菌Probio-M9和其它相关物种的基因组序列,分别设计了鼠李糖乳杆菌种水平特异性探针(Probe-16S)和鼠李糖杆菌Probio-M9菌株水平特异性探针(Probe-SP)。在使用15株鼠李糖乳杆菌菌株和15株其它益生菌验证这些特异性探针后,通过荧光原位杂交(FISH)验证鼠李糖乳杆菌Probio-M9菌株的特异性探针在体外和体内的适用性。结合荧光D-氨基酸(FDAA)探针,在大鼠的肠道中检测到鼠李糖乳杆菌Probio-M9的活细胞。本研究设计的FISH探针可从肠道菌群中特异性鉴定鼠李糖乳杆菌Probio-M9,该方法可与FDAA探针结合使用,以进一步探索该益生菌菌株在宿主肠道中的定位。 展开更多

Our objective is to investigate the subcellular transport mechanism of the UL4 protein. In this study, fluorescence microscopy was employed to investigate the subcellular localization of UL4 and characterize the transport mechanism in living cells. By constructing a series of deletion mutants fused with enhanced yellow fluorescent protein (EYFP), the nuclear export signals (NES) of UL4 were for the first time mapped to amino acid residues 178 to 186. In addition, the N-terminal 19 amino acids are identified to be required for the granule-like cytoplasmic pattem of UL4. Furthermore, the UL4 protein was demonstrated to be exported to the cytoplasm through the NES in a chromosomal region maintenance 1 (CRM1)-dependent manner involving RanGTP hydrolysis 展开更多

有效补充益生菌可有益肠道健康,但是益生菌在肠道原位活性的分析却鲜有报道。本研究通过将异硫氰酸荧光素(FITC)和5(6)-羧基四甲基罗丹明琥珀酰亚胺酯[5(6)-TAMRA-SE]分别与D-赖氨酸进行偶联,获得了两种颜色的荧光D-氨基酸(fluorescent ... 有效补充益生菌可有益肠道健康,但是益生菌在肠道原位活性的分析却鲜有报道。本研究通过将异硫氰酸荧光素(FITC)和5(6)-羧基四甲基罗丹明琥珀酰亚胺酯[5(6)-TAMRA-SE]分别与D-赖氨酸进行偶联,获得了两种颜色的荧光D-氨基酸(fluorescent D-amino acids, FDAAs)探针:绿色探针(fluorescein-D-lysine, FDL)和红色探针(TAMRA-D-lysine, TDL),并尝试对嗜酸乳杆菌(Lactobacillus acidophilus, LA)、干酪乳杆菌(Lactobacillus casei, LC)和韦荣球菌(Veillonella atypica, VA) 3种益生菌进行了体外标记。随后将FDAAs应用到小鼠肠道菌群标记,建立方法考察了3种常用益生菌口服定植存活情况。所有动物实验方案经由广州中医药大学动物伦理委员会审查通过。结果表明,合成的两种FDAAs可以无毒且完全对3种益生菌进行体外标记;通过FDAAs两步标记口服益生菌可知,LA存活率较高为92.30%±1.67%, LC和VA的存活率相近,分别为84.13%±4.06%、82.27%±2.43%。本研究可通过快速比较不同益生菌在体内定植存活率的变化,为益生菌在肠道原位定植活性提供理论支撑,同时对临床益生菌合理用药具有指导作用。 展开更多

Amino acid neurotransmitters facilitate the transmission of nerve messages across the synapses and play essential roles in the control and regulation of a variety of functions in the central and peripheral nervous system. In this study, we developed a sensitive and efficient method using high-performance liquid chromatography (HPLC) with fluorescence detection for the assay of five important amino acid n After derivatization with o-phthaldialdehyde (OPA), aspartate (Asp), glutamic acid (Glu), glycine (Gly), taurine (Tau) and ),-aminobutyric acid (GABA) were simultaneously detected in the presence of the internal standard homoserine (Hse). Precise separation of these five amino acids was achieved using isocratic elution within 24 min. Good linearity was found over the concentration range with correlation coefficients (r2) not less than 0.9998. The limit of detection (LOD) values were no more than 10 nmol/L. The intra- and inter-day reproducibility was adequate with the relative standard deviation (RSD) of 10.5% or below. This method has also been applied to the analysis of amino acids in the substantia nigra and striatum samples obtained from C57BL/6 mice. 展开更多
