MethodsMoclobemide sustained release tablets were given as a multiple oral dose regimen of 300 mg oncedaily for five consecutive days to 12 healthy volunteers. The concentrations of moclobemide inplasma were determined by reversed-phase high performance liquid chromatography. The partialpharmacokinetic parameters were calculated using 3p97 pharmacokinetic program. Results Theconcentration-time profile fitted an one-compartment model best. The steady-state pharmacokineticparameters of moclobemide sustained release tablets after multiple oral doses were as follows:C_(max) was (1 950 +- 156) μg· L^(-1), T_(max) was (6.00 +-1.55) h, T_(1/2(kel)) was (3.14 +-0.12)h, AUC_(ss 0-24) was (22 836 +- 1 842) μg·h· L^(-1), MRT was (7.68+-0.36) h, CL/F_((s)) was(20.2+-2.1) L·h^(-1), and V/F_((c)) was (91.4+-9.4) L, respectively. No marked adverse events werenoted during this study. Conclusion The formulation has a sustained-release effect and goodtolerance in the healthy volunteers, which provides useful information for clinical practice.展开更多

Methods: Relevant docu...Objective: Excavate the medication rule of traditional Chinese medicine in the treatment of prostate cancer, and predicting the biomolecular level mechanism of high-frequency drug compatibility. Methods: Relevant documents in CNKI, Wanfang Medical Network and VIP Chinese Biomedical Periodical Database Pubmed, EMbase were collected and collated systematically. Frequency statistics, association rule analysis and new party mining were carried out using TCMISSV2.5. BATMAN-TCM was used to analyze the interaction relationship and related pathways between high-frequency drug targets. Results: Huangqi (Astragalus membranaceus) was the single drug most used of the 102prescriptions included in the standard. There are 6 pairs of combinations with high confidence in association rule analysis. System entropy cluster analysis resulted in 20 core drug combinations and 9 new prescriptions. Through KEGG pathway analysis of Huangqi, Fuling (Poria cocos), Gancao (Glycyrrhiza uralensis) and Dihuang (Rehmannia glutinosa), it was found that the number of potential targets of the neural active ligand receptor rented pathway and purine metabolism pathway was the largest. Conclusions: Prostate cancer is mainly treated with deficiency-tonifying drugs, which are combined with drugs for promoting blood circulation, removing blood stasis, clearing heat, promoting diuresis, detoxifying and resolving hard mass. The mechanism of action of high-frequency traditional Chinese medicine may be realized by interfering with the neuroactive ligand receptor interaction pathway and purine metabolism pathway.展开更多

Lung cancers are generally divided into two main categories: SCLC (small cell lung cancer) and NSCLC (non-small cell lung cancer). NSCLC accounts for approximately 85% of all lung cancers. ALK (anaplastic largecell kinase) gene rearrangements are identified and targeted resulting in promising response rates for NSCLC in early studies. Ceritinib is a second-generation ALK inhibitor that has demonstrated activity in crizotinib (the first-generation ALK inhibitor)-resistant patients. In this paper, the synthesis, pharmacodynamics, pharmacokinetics, therapeutic trials, adverse events and drug-drug interactions are briefly overviewed in order to make more scholars, medical workers and patients have a more clear and comprehensive recognition on this drug.展开更多
This study was concerned with developing an antiretroviral drug distribution routing system with a goal of improving fleet utilization whilst reducing delivery costs. The system would enhance ARV drug delivery satisfa...This study was concerned with developing an antiretroviral drug distribution routing system with a goal of improving fleet utilization whilst reducing delivery costs. The system would enhance ARV drug delivery satisfaction of patients staying in the Limpopo province of South Africa. A VRP mathematical programming problem was formulated and the Savings Based as well as the Sequential Insertion algorithm was used to solve the problem. A mini program was then developed in Visual Basic.Net software that speeded up the vehicle route determination heuristics. This computer based vehicle routing system gave a total travelled distance of 1302.94 km and a space utilization of 93% as compared to the pigeonhole system which had a total travelled distance of 2874.2 km and space utilization of 86% for the demand of 5384 ARV drug patients. Therefore, the mathematical programming approach is more cost effective and efficient thereby enhancing delivery satisfaction to ARV drug patients in the province.展开更多
