The production and utilization of high-energetic explosives often pose a range of safety hazards, with sensitivity being a key factor in evaluating these risks. To investigate how temperature, particle size, and air humidity affect the responsiveness of commonly used high-energetic explosives,a series of BAM(Bundesanstalt für Materialforschung und-prüfung)impact and friction sensitivity tests were carried out to determine the critical impact energy and critical load pressure of four representative high-energetic explosives(RDX,HMX,PETN and CL-20)under different temperatures, particle sizes, and air humidity conditions. The experimental findings facilitated an examination of temperature and particle size affecting the sensitivity of high-energetic explosives, along with an assessment of the influence of air humidity on sensitivity testing. The results clearly indicate that high-energetic explosives display a substantial decline in critical reaction energy when subjected to micrometre-sized particles and an air humidity level of 45%at a temperature of 90℃.Furthermore,it was noted that the critical reaction energy of high-energetic explosives diminishes with an increase in temperature within 25℃−90℃.In the same vein,as the particle sizes of high-energetic explosives increase,so does the critical reaction energy for micrometre-sized particles.High air humidity significantly affects the sensitivity testing of high-energetic explosives, emphasizing the importance of refraining from conducting sensitivity tests in such conditions. 展开更多

This review starts with a brief history of drug discovery & development, and the place of Asia in this worldwide effort discussed. The conditions and constraints of a successful translational R&D involving academic basic research and clinical research are discussed and the Singapore model for pursuit of open R&D described. The importance of well-characterized, validated drug targets for the search for novel targeted anti-cancer agents is emphasized, as well as a structured, high quality translational R&D. Furthermore, the characteristics of an attractive preclinical development drug candidate are discussed laying the foundation of a successful preclinical development. The most frequent sources of failures are described and risk management at every stage is highly recommended. Organizational factors are also considered to play an important role. The factors to consider before starting a new drug discovery & development project are described, and an example is given of a successful clinical project that has had its roots in local universities and was carried through preclinical development into phase I clinical trials. 展开更多

The elderly population is increasing worldwide, with subjects 〉 65 years of age constituting the fastest-growing age group. Furthermore, the elderly face the greatest risk and burden of cardiovascular disease mortality and morbidity. Although elderly patients, particularly those older 〉 75, have not been well represented in randomized clinical trials evaluating lipid-lowering therapy, the available evidence supporting the use of statin therapy in primary prevention in older individuals is derived mainly from subgroup analyses and post-hoc data. On the other hand, elderly patients often have multiple co-morbidities that require a high number of concurrent medications; this may increase the risk for drug-drug interactions, thereby reducing the potential benefits of statin therapy. The aim of this review was to present the relevant literature regarding statin use in the elderly for theft primary cardiovascular disease, with the associated risks and benefits of treatment. 展开更多

Abstract: The main purpose of the research was to study a role and place of managers in maintaining staff policy at pharmaceutical companies. It was interesting to see how Georgian pharmaceutical companies are getting new employees at work, what is the priority of the new recruits? What methods are used for selection and recruitment? There were made inquiries by the way of filling applications, which (the applications) were given to the managers ofAversi, PSP, GPC, Pharmadepot and Pharmaceutical House. 50 managers were interviewed totally. It was established that pharmaceutical company management appoints people on the position of pharmacist-managers mainly on the basis of contract (90%). At present, the pharmacist education does not comply with the competence, so 55% of interviewed managers are bachelors, 45% are masters. In order to increase the management's qualifications, pharmaceutical companies are conducting seminars (50%), trainings (30%), and testing (20%). Staff administration in the pharmacies is mainly carried out by the company administration (75%), rarely by managers (25%), but very often the administration takes into consideration the manager's opinion; employees are accepted by testing-interview (70%) or CV (30%). Professional career and experience of the newly employed staff are not highly important (50/50), and the company prefers to educate and share practice to the personnel itself. It is possible to conclude that today the pharmacist-managers of the pharmacies are working in accordance with the contractual fights and obligations signed with the employers and pharmaceutical activities regulated under non-governmental institutes. As a result, it is important to be included special features of pharmaceutical management and the recommendations offered by the World Health Organization and the International Pharmaceutical Federation in the articles of regulatory laws. 展开更多

A dynamic experimental set-up was utilized to measure ibuprofen solubility in supercritical CO2 at the pressure range of 8-13 MPa and the temperatures of 308, 313 and 318 K. Mole fraction values varied from 0.015×10^-3 to 3.261×10^-3 and correlated by using seven different semi empirical equations of state (Bartle, Modi-fied Bartle, Mendez-Teja, Modified Mendez-Teja, Kumar-Johnson, Sung-shim and Gordillo) as well as seven cubic equations of state (van der Waals, Redlich-Kwong, Soave-Redlich-Kwong, Peng-Robinson, Stryjek-Vera, Patel-Teja-Valderana and Pazuki). Single and twin-parametric van der Walls mixing rules (vdW1, vdW2) were ap-plied in order to estimate the supercritical solution properties. The physicochemical properties were also obtained using Joback, Lydersen and Ambrose methods. Absolute average relatives deviation (AARD) were calculated and compared for all the correlating systems. Results showed that among the cubic equations of state (EOSs) the Pazuki equation (AARD=19.85% using vdW1 and AARD=8.79% using vdW2) and SRK equation (AARD=19.20%using vdW1 and AARD=10.03%using vdW2) predicted the ibuprofen solubility in supercritical CO2 with the least error in comparison to the others. Among the semi-empirical EOSs the most desirable deviation (AARD〈10%) was obtained by using Modified Bartle and Modified Mendez-Teja equations in all the studied temperatures. 展开更多

Coronavirus disease 2019(COVID-19), caused by severe acute respiratory syndrome coronavirus 2(SARSCo V-2), has become a global pandemic. Clinical evidence suggests that the intestine is another high-risk organ for SARS-Co V-2 infection besides the lungs. However, a model that can accurately reflect the response of the human intestine to the virus is still lacking. Here, we created an intestinal infection model on a chip that allows the recapitulation of human relevant intestinal pathophysiology induced by SARSCo V-2 at organ level. This microengineered gut-on-chip reconstitutes the key features of the intestinal epithelium-vascular endothelium barrier through the three-dimensional(3 D) co-culture of human intestinal epithelial, mucin-secreting, and vascular endothelial cells under physiological fluid flow. The intestinal epithelium showed permissiveness for viral infection and obvious morphological changes with injury of intestinal villi, dispersed distribution of mucus-secreting cells, and reduced expression of tight junction(E-cadherin), indicating the destruction of the intestinal barrier integrity caused by virus. Moreover, the vascular endothelium exhibited abnormal cell morphology, with disrupted adherent junctions. Transcriptional analysis revealed abnormal RNA and protein metabolism, as well as activated immune responses in both epithelial and endothelial cells after viral infection(e.g., upregulated cytokine genes), which may contribute to the injury of the intestinal barrier associated with gastrointestinal symptoms. This human organ system can partially mirror intestinal barrier injury and the human response to viral infection, which is not possible in existing in vitro culture models. It provides a unique and rapid platform to accelerate COVID-19 research and develop novel therapies. 展开更多
