近十年来,硼替佐米(bortezomib,BTZ)在血液恶性肿瘤尤其是多发性骨髓瘤(multiple myeloma,MM)和套细胞淋巴瘤(mantle cell lymphoma,MCL)的治疗中起到了非常重要的作用,尽管如此,继发性耐药的出现为该药物的临床使用带来了很大的挑战。... 近十年来,硼替佐米(bortezomib,BTZ)在血液恶性肿瘤尤其是多发性骨髓瘤(multiple myeloma,MM)和套细胞淋巴瘤(mantle cell lymphoma,MCL)的治疗中起到了非常重要的作用,尽管如此,继发性耐药的出现为该药物的临床使用带来了很大的挑战。本文根据近年来对硼替佐米耐药机制的研究现状,从泛素蛋白酶体系统(ubiquitin-proteasome system,UPS)的作用、内质网应激通路、自噬作用、细胞促存活信号通路和骨髓微环境等方面作一综述,并探讨耐药机制中可能存在的新的治疗策略。近年来随着对耐药机制的不断研究,新的组蛋白去乙酰化酶抑制剂(histone deacetylase inhibitors,HDACi)已经被用于MM的治疗,AKT抑制剂也处于临床试验阶段,这些新的联合治疗方法有望增强硼替佐米的疗效,改善患者的预后。 展开更多

目的:探讨乳酸脱氢酶A(lactate dehydrogenase A,LDHA)乙酰化调节促进多发性骨髓瘤(multiple myeloma,MM)对硼替佐米(bortezomib,BTZ)耐药的可能机制。方法:在骨髓瘤患者中检测LDHA表达与BTZ敏感性的关系。构建对BTZ耐药的骨髓瘤细胞株... 目的:探讨乳酸脱氢酶A(lactate dehydrogenase A,LDHA)乙酰化调节促进多发性骨髓瘤(multiple myeloma,MM)对硼替佐米(bortezomib,BTZ)耐药的可能机制。方法:在骨髓瘤患者中检测LDHA表达与BTZ敏感性的关系。构建对BTZ耐药的骨髓瘤细胞株,检测耐药株与敏感株LDHA的表达情况。用CCK8检测BTZ处理后骨髓瘤耐药和敏感株的细胞增殖,用流式细胞仪检测BTZ处理后耐药和敏感株的凋亡率。在iHypoxia数据库中对LDHA可能的翻译后修饰位点进行预测,并对常见的翻译后修饰进行检测。为了探索LDHA上游调控机制,使用不同的抑制剂处理骨髓瘤细胞后检测LDHA的表达情况。结果:MM患者肿瘤组织的LDHA表达明显高于对照组,且对BTZ耐药患者LDHA的蛋白和mRNA表达明显高于对BTZ敏感的患者。成功构建BTZ诱导耐药的MM1.s-_(BR)和NCI-H929-_(BR)耐药细胞株。CCK8检测细胞活性证明BTZ耐药细胞株在BTZ处理后存活率高于敏感细胞株,而流式细胞仪结果显示BTZ敏感细胞株在BTZ处理后凋亡率高于耐药细胞株,以上结果证实BTZ耐药细胞株的构建成功。Western blot结果显示LDHA在BTZ耐药细胞株中的表达高于野生型细胞株。通过数据库预测,LDHA常见的翻译后修饰位点为磷酸化、乙酰化和泛素化。BTZ耐药细胞株中乙酰化水平较敏感株明显下降,而其他翻译后修饰程度耐药株和敏感株无明显差异。组蛋白脱乙酰酶类Ⅰ和Ⅱ的共同抑制剂trichostatin处理BTZ耐药细胞株后LDHA蛋白含量下降,乙酰化水平上调,且可以克服耐药株MM1.s-_(BR)对BTZ的耐药,但不能克服高表达LDHA的MM1.sLDH high对BTZ的耐药。结论:组蛋白脱乙酰酶类Ⅰ/Ⅱ可能通过促进LDHA的去乙酰化提高LDHA水平,促进MM对BTZ耐药。 展开更多

Aim To establish a new and sensitive HPLC-MS method for the determination ofzolmitriptan in human plasma and study the pharmacokinetics of zolmitriptan in healthy volunteers.Methods A single oral dose of 5 mg of zolmitriptan tablet was given to 20 healthy male volunteers.After dosing, blood samples were collected for a period of 24 h, and zolmitriptan concentration inplasma was analyzed by HPLC-MS. Results The plasma concentration-time course fitted well atwo-compartment open model with a lag time, giving the following pharmacokinetic parameters: T_(max)1.60 ± 0.24 h, C_(max) 9.73 ± 1.43 ng·mL^(-1). T_(1/2α)1.72±0.46 h, T_(1/2β) 4.52 + 0.97 h,and AUC_(0-t) 55.59 ± 5.12 ng·mL^(-1)·h. Conclusion The improved analytical method forzolmitriptan is rapid, sensitive and suitable for application to pharmacokinetic studies and routinedetermination of numerous samples. 展开更多

Infrared spectra (IR) revealed that there was a substitution reaction mainly on the amine groups of chitosan (CS). By using paracetamol (PCTM) as a model drug, the balanced release concentration of PCTM in phosphate buffer solution (pH=7.4) can be decreased with the increase of degree of substitution alkyl and can be reduced effectively even under a lower PCTM loading. 展开更多

Comparative genetic studies have shown that there are widespread synteny and colinearity of the genes among different species within grass family. Rice ( Oryza sativa L.) is a model plant, and analysis of its genome allows us to reveal the common features and the evolutionary rules of the gramineous genomes and accumulate the data for establishment of a common genetic system in the Poaceae. In this study, a rice gene Pib ( 10.3 kb), a map-based cloned gene, and RFLP markers linked with it are used as the tested probes to investigate their homology and physical location among the tested species. Southern blotting analysis showed that there were sequences homologous to Pib in maize genome. Further, Pib was localized onto the chromosomes of O. sativa ssp. indica cv. Guangluai 4, O. officinalis Wall ex Watt and the inbred line of Zea mays cv. Huangzao 4. The results of fluorescence in situ hybridization (FISH) and double-color FISH indicated that a synteny of Pib and RFLP markers linked with Pib existed among the genomes of the three tested species. 展开更多

Using splenic cells of BALB/c mice infected with Friend Leukemia Virus (FVA) as a model of erythroid cells, we investigated the action of dexamethasone (Dex) to induce apoptosis of the cells in the presence of erythropoietin (EPO)——an apoptotic inhibitor in developing erythroid cells. The result indicated that after treatment with a certain range of Dex concentrations and prolonged incubation, the cells were characterized by the occurrence of DNA ladders which appeared on agarose electrophoresis. Transmission electron microscopy showed that karyopyknosis, chromatin condensation, dilatation of the perinuclear space, karyorrhexis, cytoplasmic vacuolization and cell fragmentation appeared in the cells depending on the dose of Dex and the treatment time. These results suggest that Dex could induce apoptosis in the developing erythroid cells as in other cells so far reported. 展开更多

Novel hollow Fe3O4 nanoparticles for drug delivery were synthesized via a one-step template- free approach. These nanoparticles were obtained by modifing the Fe3O4 nanoparticles with 3-aminopropyltrimethoxy silane, and then grafting alginate onto the surface of amine magnetic. The hollow structure of Fe3O4 spheres was characterized by TEM, XRD, and XPS. The M-H hysteresis loop indicated that the magnetic spheres exhibit snperparamagnetic characteristics at room temperature. Daunorubicin acting as a model drug was loaded into the carrier, and the maximum percent of envelop and load were 28.4% and 14.2% respectively. The drug controlled releasing behaviors of the carriers were compared in different pH media. 展开更多

In this paper, glasshouse experiments were conducted to determine the accumulation, distribution and transformation of DDTs and HCHs by maize under pot culture conditions. The culture soil was spiked with the organic pollutants and samples were collected in different durations. Analysis of the plants at harvest showed that the selected plant varied widely in their ability to remove and translo- cate DDTs and HCHs from the soil, the bioconcentration factor ranged from 0.004 to 0.027 for the shoot and from 0.036 to 0.097 for the roots, and the translocation factors were lower than 0.1 with variation between DDTs and HCHs, but no signifi- cant differences were observed. DDTs appeared to have accumulated by both pas- sive adsorption and active absorption, p,p'-DDT and p,p'-DDE were the major metabolite and the transformation was mediated by reductive dehalogenation, the affinity of the OCPs for lipids is one of the major factors affecting their uptake and translocation within the plants. 展开更多

目的:探讨自噬抑制剂ROC-325对人多发性骨髓瘤细胞株增殖、凋亡、自噬的影响及其与硼替佐米联合用药的细胞学效应。方法:不同浓度的ROC-325作用于骨髓瘤细胞,用CCK-8法检测药物对细胞增殖的作用,用Caspase-3/7、Caspase-9活性测定法检... 目的:探讨自噬抑制剂ROC-325对人多发性骨髓瘤细胞株增殖、凋亡、自噬的影响及其与硼替佐米联合用药的细胞学效应。方法:不同浓度的ROC-325作用于骨髓瘤细胞,用CCK-8法检测药物对细胞增殖的作用,用Caspase-3/7、Caspase-9活性测定法检测药物对细胞凋亡的影响,用单丹磺酰戊二胺染色观察自噬小体数量,Western blot检测各组细胞凋亡相关蛋白(PARP和Caspase-3)和自噬相关蛋白(P62、Beclin-1和LC3A/B)的表达情况,用CCK-8法检测ROC-325联合硼替佐米对硼替佐米耐药细胞株的抑制作用。结果:ROC-325可呈剂量依赖性抑制RPMI 8226、RPMI 8226-BTZ100、U266和IM9细胞增殖(r=-0.8275,r=-0.9079,r=-0.9422,r=-0.9305),72 h IC50分别为2.795、4.020、5.432和4.755μmol/L。Caspase-3/7、Caspase-9活性结果显示,随着药物浓度的增加,Caspase-3/7和Caspase-9相对活性逐渐升高(r=0.9648,r=0.9377,r=0.9318;r=0.9087,r=0.9431,r=0.8914)。单丹磺酰戊二胺染色结果显示,细胞内自噬泡数量随着ROC-325浓度的提高逐渐增多(r=0.9565,r=0.9373,r=0.9233)。Western blot结果显示,ROC-325可诱导凋亡相关蛋白PARP、Caspase-3表达上调以及自噬相关蛋白P62、LC3-Ⅱ/LC3-Ⅰ表达上调,Beclin-1表达下调。ROC-325联合硼替佐米的CCK-8法检测结果显示,ROC-325与硼替佐米对耐药细胞株RPMI 8226-BTZ100的抑制具有协同作用。结论:ROC-325可以抑制骨髓瘤细胞增殖,通过线粒体途径诱导骨髓瘤细胞的凋亡,通过影响自噬体和溶酶体的融合抑制骨髓瘤细胞的自噬,ROC-325联合硼替佐米后可以克服硼替佐米耐药。 展开更多

It can increase the stability of the drug and enhance drug bioavailability. The selective targeting of nanoparticles can be achieved through enhanced permeability and retention effect and a conjugated specific ligand or through the effects of physiological conditions, such as pH and temperature. Nanoparticles can be prepared by using a wide range of materials and can be used to encapsulate chemotherapeutic agents to reduce toxicity, which can be used for imaging, therapy, and diagnosis. In this research, recent progress on nanoparticles as a targeted drug delivery system will be reviewed, including positive-targeting, negative-targeting, and physicochemical-targeting used as anticancer drug carriers. 展开更多

Fe3O4 magnetic nanoparticles were prepared by the aqueous co-precipitation of FeCl3-6H2O and FeCl2-4H2O with addition of ammonium hydroxide. The conditions for the preparation of Fe3O4 magnetic nanoparticles were optimized, and Fe3O4 magnetic nanoparticles obtained were characterized systematically by means of transmission electron microscope (TEM), dynamic laser scattering analyzer (DLS) and X-ray diffraction (XRD). The results revealed that the magnetic nanoparticles were cubic shaped and dispersive, with narrow size distribution and average diameter of 11.4 nm. It was found that the homogeneous variation of pH value in the solution via the control on the dropping rate of aqueous ammonia played a critical role in size distribution. The magnetic response of the product in the magnetic field was also analyzed and evaluated carefully. A 32.6 mT magnetic field which is produced by four ferromagnets was found to be sufficient to excite the dipole moments of 0.05 g Fe3O4 powder 2 cm far away from the ferromagnets. In conclusion, the Fe3O4 magnetic nanoparticles with excellent properties were competent for the magnetic carders of targeted-drug in future application. 展开更多

Platinum-based anticancer agents are widely used as first-line drugs in cancer chemotherapy for various solid tumors. However, great side effects and occurrence of resistance remain as the major drawbacks for almost all the platinum drugs developed. To conquer these problems, new strategies should be adopted for platinum drug based chemotherapy. Modern nanotechnology has been widely employed in the delivery of various therapeutics and diagnostic. It provides the possibility of targeted delivery of a certain anticancer drug to the tumor site, which could minimize toxicity and optimize the drug efficacy. AIM: Predictive value of serum b2-microglobulin (b2m)levels for virological breakthrough (VB) in HBeAg-negative chronic hepatitis B (CHB) patients under long-term treatment schedules including lamivudine (LAM).METHODS: Serum b2m levels were calculated during treatment in 25 CHB patients under long-term LAM monotherapy (group A) and 12 patients under initial interferon plus LAM treatment followed by LAM monotherapy (group B), using the MEIA technology. We used Cox proportional hazard models in order to investigate the association between serum b2m levels and VB.RESULTS: Seven of 25 patients (28%), 9/25 (36%) and 14/25 (56%) from group A and 0/12, 2/12 (16.6%) and 3/12 (25%) from group B exhibited VB at months 12, 24 and 36 of treatment, respectively. All patients, from both groups, who did not show VB exhibited b2m elevation in mo 3. The duration of b2m elevation was significantly longer in the virological responder's subgroup from group A than the non-responder's one (7.3±2.6 vs 3.8±3.4 mo,P = 0.02). In comparison to group A patients whose b2m levels were increased at 3 mo, patients whose b2m levels were decreased had 4.6 times higher risk of experiencing VB (RR = 4.6, P = 0.024). When baseline variables were simultaneously included in the same Cox model, decreased b2m status was still associated with increased risk of VB (RR = 12.2, P = 0.03).CONCLUSION: In HBeAg-negative CHB patients under either long-term LAM monotherapy or initial combination treatment, serum b2m levels at 3 mo of treatment,compared to baseline ones, might be a predictor of risk for VB. 展开更多
