Methods Themodel of PD was induced by intraperitoneal injection of MPTP into C57BL/6J mice for 4 d. Modafinil(ip, 50 or 100 mg·kg^(-1)·d^(-1)) was administered following MPTP for 4 d and for another 10 dconsecatirely. The effects of modafinil on the locomotor activity, and the incubation, maintenanceperiod and grade of the tremor, the duration of the climbing rod of mouse, and the distribution ofpositive cells of ty-rosine hydroxylase (TH) and Nissl bodies in the striatum and substantia nigra(SN) were observed. The contents of dopam-ine (DA) , noradrenaline (NA) and 5-hydroxytryptamine(5-HT) in the striatum were determined. Results Modafinil (50 and 100 mg·kg^(-1)) significantlyprevented the locomotor, the tremor and climbing rod defect behavior in a dose-dependent manner (P <0.05 and P < 0.01, n = 10), prevented the decrease in the number of TH-positive cells and Nisslbodies (P<0.05, n=10), and reduced the decrease of DA, NA, and 5-HT in the striatum (P < 0.05, n =10) induced by MPTP. Conclusion Modafinil improves the behavioral deficits and prevents themonoaminergic neuron lesion in seriously impaired MPTP mouse model.展开更多

Famotidine plasma concentration was determined by HPLC method, and the plasma concentration time data were processed with the method provided by USP XXIII. For single dose administration the peak plasma concentration occurring at 8 13±0 34 h was 69 52±3 00 ng/ml and the relative bioavailability was 112 4±8 6%. For multiple dose administration the peak plasma concentration of SR tablet was 86 14±2 95 ng/ml and the degree of fluctuation (DF) was 140 6±13 5% at steady state. Two one sided tests were performed in bioequivalence assessment. The results showed that the sustained release tablets were basically bioequivalent to the immediate release (IR) tablets on sale.展开更多

The prescription automatic screening system was used to screen the prescriptions in children with ADHD. Kaplan-Meier regression analysis was used to compare prescription retention rates between the two regimens after adjusting for gender, age, body weight, and prescription cost. The mean age of the methylphenidate hydrochloride group was 8.75 ± 2.16 years, and the monthly prescription cost was 327.37 ± 146.64 RMB. The average age of the atomoxetine group was 8.33 ± 1.73 years, and the monthly prescription cost was 363.15 ± 154.90 ¥. There were some differences in the age of enrollment and the monthly prescription cost between the two regimens(all P < 0.01). Moreover, the retention rate of methylphenidate hydrochloride was higher compared with atomoxetine hydrochloride within 18 months. Kaplan-Meier regression analysis showed that this trend was significant(Tarone-ware, Chi-square value = 14.893, P < 0.001). Prescription costs might be a factor affecting drug retention. This study found that the retention rates were decreased month by month in children with ADHD, and after 5 months, the retention rates were 52.20% and 41.22%, respectively, far below the recommended levels of the guidelines.展开更多

Methods: Experimental and control rabbits were subjected to a closed head injury. In one group nimodipine was given intravenously and the effect evaluated by electron microscopy, brain water content, calcium levels, transcranial Doppler, and intracranial pressure monitoring. Results: In rabbits treated with nimodipine the level of neuronal cytosolic free calcium was markedly decreased. There were less cellular damage and less spasm of the middle cerebral artery seen on electron microscopy. No difference regarding intracranial pressure changes between the two groups was noted. Conclusions: Nimodipine has a protective action on brain injury by blocking a series of pathological reactions induced by neuronal calcium overload, and by reducing the spasm of brain vessels and improving cerebral blood flow.展开更多
