莲种胚在温度胁迫下的逆境生理研究 被引量:2

为明确莲种胚对不同温度胁迫的耐受能力,探索莲种胚细胞在逆境胁迫下的生理响应规律,该研究以成熟莲子为材料,从形态学、抗性生理和抗氧化相关基因表达定量3个层面进行了研究。结果表明:(1)莲子对于高温与超低温具有较好的胁迫耐受能力,... 为明确莲种胚对不同温度胁迫的耐受能力,探索莲种胚细胞在逆境胁迫下的生理响应规律,该研究以成熟莲子为材料,从形态学、抗性生理和抗氧化相关基因表达定量3个层面进行了研究。结果表明:(1)莲子对于高温与超低温具有较好的胁迫耐受能力,经70℃高温与-196℃超低温处理后,其发芽率和种芽长相比对照组无显著变化;80℃及以上的高温处理会使莲种胚吸水萌发迟缓,发芽率降低50%及以上,种芽萌发变慢。(2)高温处理后,莲种胚细胞中抗氧化酶活性随吸水萌发过程呈上升趋势,非酶促抗氧化剂含量下降,细胞膜脂过氧化程度逐渐减轻,质膜完整性有所恢复。(3)高温处理下,莲种胚氧化应激相关基因(DHN Rab18、Cu/Zn SOD、Mn SOD、POD41、POD73、CAT1、GR、APX)积极参与胁迫响应,出现不同程度的上调表达,100℃处理组中DHN Rab18、Cu/Zn SOD、POD41、GR与APX上调表达幅度较大。综上结果认为,莲种胚具有良好的高温与低温胁迫耐受性,在不同温度胁迫下抗氧化系统与抗逆保护类蛋白对维持莲种胚的细胞活力可能发挥重要保护作用。 展开更多

Introduction, Collection and Characterization of Lotus spp. Germplasm in Chile

The information related with results of a project on germplasm characterization of three Lotus species, important as forage plants, curried out in five environments of the central-southern zone of Chile, was reviewed. An evaluation was made of 12 L. corniculatus cultivars introduced from different parts of the world, as well as germplasm naturalized in Chile, being 11 accessions of L. tenuis and 21 accessions ofL. uliginosus collected from 33°S to 38°S and 38°S to 45°S, respectively. Phenological, morphological, and agronomical characters were evaluated. Nitrogen fixation in field conditions and concentrations of condensed tannins in herbage were also included. It is concluded that there are cultivars of high value in L. corniculatus introduced germplasm which can be recommended for different environments of the central zone of Chile, as well as for local species breeding programmes. Information obtained from L. tenuis and L. uliginosus accessions shows genetic variability within both species. L. tenuis germplasm is adapted to clay, medium-textured, or sandy soils with water restrictions and phosphorous deficiency. Therefore, characterized accessions could be used to breed cultivars for low input production systems in constrained environments. L. uliginosus accessions include genotypes collected in sites with acid soils and variable tannin content in plant tissues that may be promissory genetic materials for breeding programmes with the objective of producing cultivars for animal production systems for the wet and acid soils in the southern zone of the country. 展开更多

With rutin standard sample as the comparison, flavonoid extraction rate as an indicator, UV-2450 ultraviolet visible light spectrophotometer to measure flavonoid content in planted Trollius chinensis through orthogonal experiment, this experiment optimizes process condition of flavonoid in Trollius chinensis through water extraction and ethanol extraction. The result shows that flavonoid extraction rate of Trollius chinensis through ethanol extraction method is obviously higher than water extraction, and the optimal extraction process condition is: ethanol concentration is 75%, extraction time is 90min, extraction times are 3, and fluid material ratio is 20:1. 展开更多

The physicochemical analysis was carried out to quantify the total soluble solids, acidity, pH, ascorbic acid contents and pulp colour of eight selected durian hybrid clones. The aim of this study was to select the most promising durian hybrids for commercialization. The durian hybrid fruits were collected in an established durian research plot. All tested characteristics with the exception for total soluble solids (TSS) showed significant differences (P 〈 0.05) amongst the eight durian hybrids with four check cultivars. F1 hybrid 15-3 had significantly higher pH (7.25) and lower titratable acidity (0.09%) than other hybrids. While for ascorbic acid content, durian hybrid 7-3 (26.13 rag/100 g) showed significantly higher than other hybrids. As for pulp colour, hybrids 1-15, 1-17 and 7-20 were yellowish orange/deep yellow. Even though there was no significant differences for TSS, three durian hybrids showed value above 39.00 °Brix and there were hybrid 14-3 (39.97 °Brix), hybrid 8-16 (39.83 °Brix) and hybrid 7-3 (39.50 °Brix). Based on the results of this study, Fl hybrids durian 1-15, 7-3, 8-16, 14-3 and 15-3 shall become successful candidates for recommendation to farmers for commercial production in Malaysia. However, further evaluations on fruit eating quality and stability of the hybrids should be completed prior to recommendation for commercialization. 展开更多
