氨基酸平衡低蛋白质日粮对育肥猪肠道黏膜抗菌肽与微生物区系的影响 被引量:7

旨在研究氨基酸平衡低蛋白质日粮对育肥猪肠道抗菌肽与微生物区系的影响。将20头(60±2.5)kg的育肥猪(杜×长×大)随机分为4个处理:14.0%CP日粮(14.0%CP),添加Glu、Lys、Met、Thr和Trp的12.5%CP日粮(12.5%CP),添加Glu、Lys... 旨在研究氨基酸平衡低蛋白质日粮对育肥猪肠道抗菌肽与微生物区系的影响。将20头(60±2.5)kg的育肥猪(杜×长×大)随机分为4个处理:14.0%CP日粮(14.0%CP),添加Glu、Lys、Met、Thr和Trp的12.5%CP日粮(12.5%CP),添加Glu、Lys、Met、Thr和Trp的11.0%CP日粮(11.0%CP),添加Glu、Lys、Met、Thr、Trp和BCAA(Val、Ile、Leu)的11.0%CP日粮(11.0%CP+BCAA)。每个处理5个重复,每个重复1头猪。试验分为5d预试期和30d正试期。结果表明:(1)尿液尿素氮和尿酸含量随日粮蛋白质水平降低而显著降低(P<0.05);(2)低蛋白质水平日粮显著降低空肠黏膜、回肠黏膜β-defensin-2水平和ANG1、ANG4 mRNA表达水平(P<0.05);(3)不同蛋白质水平日粮对部分回肠、直肠食糜微生物氨基酸组成有显著影响(P<0.05);(4)与14.0%CP组相比,12.5%CP组Observed species显著提高(P<0.05),11.0%CP组Shannon、PD显著降低(P<0.05),11.0%CP+BCAA组Shannon显著降低(P<0.05)。4组之间拟杆菌门、厚壁菌门的相对丰度差异不显著(P>0.05)。与14.0%CP组相比,12.5%CP组和11.0%CP组酸杆菌门、芽单胞菌门相对丰度显著降低(P<0.05),11.0%CP+BCAA组芽单胞菌门相对丰度显著降低(P<0.05)。14.0%CP组变形菌门相对丰度显著高于12.5%CP组和11.0%CP+BCAA组(P<0.05),与11.0%CP组差异不显著(P>0.05),14.0%CP组弯曲菌科相对丰度显著高于其他组(P<0.05)。14.0%CP组粪肠球菌属、狭义梭菌属的相对丰度显著高于其他组(P<0.05),11.0%CP组和11.0%CP+BCAA组毛螺菌属FCS020的相对丰度显著高于12.5%CP组和14.0%CP组(P<0.05)。低蛋白质水平氨基酸平衡日粮能提高氮的利用率,影响肠道微生物区系组成,在低蛋白质日粮中添加BCAA有助于增加氨基酸的平衡性、维持肠道免疫机能。 展开更多

基于13C标记的代谢通量分析(13CMFA)由于具有准确性和应用性已成为国际上代谢工程研究的热点,其中一个至关重要的问题就是分析菌体蛋白氨基酸的13C标记丰度信息。用含20%全标记葡萄糖([U-13C])和80%天然葡萄糖的合成培养基喂养维生素B1... 基于13C标记的代谢通量分析(13CMFA)由于具有准确性和应用性已成为国际上代谢工程研究的热点,其中一个至关重要的问题就是分析菌体蛋白氨基酸的13C标记丰度信息。用含20%全标记葡萄糖([U-13C])和80%天然葡萄糖的合成培养基喂养维生素B12生产菌Pseudomonas denitrifican,然后在稳态条件下取20mg带13C标记菌体用1ml6mol/L盐酸95℃水解24h得到带13C标记的菌体蛋白氨基酸;氨基酸经分离、浓缩、真空干燥、MBDSTFA衍生化后得到的TBDMS衍生物可以用气相色谱-质谱(GC-MS)进行分析,最后分析质谱图得到15种菌体蛋白氨基酸的13C标记丰度信息。成功获得菌体蛋白氨基酸13C标记丰度信息的实验方法和样品处理技术,为13CMFA在国内进一步发展提供了参考意义。 展开更多

The gene was comprised of an open reading frame of 1224bp, encoding 407 amino acid residues... A new κ-carrageenase gene cgkS was cloned from marine bacterium Shewanella sp. Kz7 by using degenerate and site-finding PCR. The gene was comprised of an open reading frame of 1224bp, encoding 407 amino acid residues, with a signal peptide of 24 residues. Based on the deduced amino acid sequence, the κ-carrageenase CgkS was classified into the Glycoside Hy- drolase family 16. The cgkS gene was expressed in Escherichia coli, and the recombinant enzyme was purified to homogeneity with a specific activity of 716.8 U mg-1 and a yield of 69%. Recombinant CgkS was most active at 45 ℃ and pH 8.0. It was stable at pH 6.0-9.0 and below 30℃. The enzyme did not require NaCl for activity, although its activity was enhanced by NaCI. CgkS degraded κ-carrageenan in an endo-fashion releasing tetrasaccharides and disaccharides as main hydrolysis products. 展开更多

Bactericidal activity of some arginine based biodegradable polymers-PEURs (poly (ester urethane)s) and PEUs (poly (ester urea)s) with low cytotoxicity was studied in in vitro experiments. Various bacterial strains both Gram-positive and Gram-negative were used to explore the bactericidal activity of the cationic polymers. As the test objects, the following microorganisms were used: Bacillus subtilis, Staphylococcus aureus, Mycobacterium album, Pseudomonas fluorescens, Escherichia coli, Actinomyces griseus and Aspergillus niger. The obtained results showed that the new cationic polymers suppressed the growth of the studied microorganisms and the bactericidal activity of the tested cationic polymers strongly depending on their chemical structure. 展开更多

THE ROLE OF BACTERIAL SYMBIONTS IN AMINO ACID COMPOSITION OF BLACK BEAN APHIDS 被引量:1

To evaluate the role of bacterial symbionts ( Buchnera spp. ) in the black bean aphids ( Aphis craccivora Koch), the aphids were treated with the antibiotic, rifampicin, to eliminate their intracellular symbiotic bacteria. Analysis of protein and amino acid concentration in 7-day-old of aposymbiotic aphids showed that the total protein content per mg fresh weight was significantly reduced by 29 %, but free amino acid titers were increased by 17% . The ratio of the essential amino acids was in general only around 20% essential amino acids in phloem sap of broad bean, whereas it was 44% and 37% in symbiotic and aposymbiotic aphids, respectively,suggesting that the composition of the free amino acids was unbalanced. For example, the essential amino acid,threonine represented 21. 6% of essential amino acids in symbiotic aphids, but it was only 16.7% in aposymbiotic aphids. Likewise, two nonessential amino acids, tyrosine and serine, represented 8.9% and 5.6% of total amino acids in symbiontic aphids, respectively, but they enhanced to 21.1% and 13.6% in aposymbiotic aphids. It seems likely that the elevated free amino acid concentration in aposymbiotic aphids was caused by the limited protein anabolism as the result of the unbalanced amino acid composition. 展开更多
