Studies on the diversity and distribution of bacterial populations will improve the overall understanding of the global patterns of marine bacteria and help to comprehend local biochemical processes and environments. We evaluated the composition and the dynamics of bacterial communities in the sediment of Jiaozhou Bay (China) using PCR-denaturing gradient gel electrophoresis (DGGE). Sediment samples were collected from 10 different sites in May, August, and November 2008 and in February 2009. There was significant temporal variation in bacterial community composition at all sites. However, the spatial variation was very small. The DGGE analyses of bacterial communities were used to divide the 10 stations into three types. Canonical correspondence analysis (CCA) revealed that the changes in bacterial communities were driven by sediment properties. Sequence analysis of DGGE band-derived 16S rRNA gene fragments revealed that the dominant bacterial groups in the sediment were of the classes ~,-proteobacteria and ~3-proteobacteria and phyla Bacteroidetes and Nitrospirae. Our results provide considerable insight into the bacterial community structure in Jiaozhou Bay, China. 展开更多

Water samples from chromite mine quarry of Sukinda and its adjacent areas were analyzed for their heavy metal contamination along with physico-chemical and microbial contents. The chromite mine water samples possessed high concentrations of heavy metals in the order of Cr〉Fe〉Zn〉Ni〉Co〉Mn while ground water did not show any heavy metal contamination except Fe. Physico-chemical parameters of mine water samples showed deviation from those of normal water. Mine water harboured low microbial populations of bacteria, fungi and actinomycetes in comparison with mine adjacent water samples. The correlation of data between metals with physico-chemical parameters showed both positive and negative responses while that of metal and microbial population exhibited negative correlation. Bacterial strains isolated from chromite mine water exhibited high tolerance towards chromium and other heavy metals as well as antibiotics which could be used as an indicator of heavy metal pollution. 展开更多

Aerobic, heterotrophic bacteria, recovered from two sites located on the west coast of Scotland, were compared to cultures obtained in a similar way from industrial, aquacultural and clean sites in the vicinity of Qingdao, Shandong, P.R. China. Gram-negative bacterial cultures were examined by BIOLOG-GN, and the data analysed by the simple matching (S SM) and Jaccard coefficients (S J) and unweighted average linkage clustering using NTSys. The output revealed that 20% of the bacteria, namely, Acinetobacter johnsonii, Aquaspirillum dispar, Pseudomonas spp. (two groups), Sphingobacterium sp., Vibrio sp., V. campbellii, V. mimicus and V. hollisae, were common between the two geographical locations. However, the study revealed shortcomings with the BIOLOG-GN system for the study of coastal Gram-negative bacteria. 展开更多

Biocidal Effect of Gamma Radiation on the Ecology of Filamentous Fungal Populations Associated with Stone Deterioration

This study focused on the identification of the filamentous fungi associated with soapstone samples exposed to outdoor conditions and the biocidal effect of gamma radiations on isolated fungal populations in Minas Gerais State in Brazil. Two soapstone blocks were placed outdoors under tropical environmental conditions for 12 months. A total of 9 filamentous fungal populations were identified on their surfaces, namely Acremomium (cf.) alternatum, Alternaria alternata, Aspergillusfumigatus, Calcarisporium (cf.) arbuscula, Cladosporium cladosporioides, Curvularia lunata, Epicoccum nigrum, Fusarium equiseti and Penicillium citrinum. The gamma radiation assay was then carried out as a test of biocidal action by exposing all fungal populations to the ionizing radiation. The results showed that only the C. cladosporioides species was resistant to this biocidal agent, since it was able to increase its population post exposure. Scanning electron microscopy images identified the microbial colonization on the soapstone blocks and the stone elementar composition was analyzed by energy dispersive X-ray spectrometry. After treatment, there was no structural and aesthetic alteration in the soapstone samples, and evidencing that gamma radiation can be used as a biocidal agent. However, the resistance of the black fungal population indicates caution in the choice of gamma irradiation as biocidal treatment. 展开更多

Its biological activity against Rhizoctonia solani was determined for the control efficacy to pepper seedling blight caused by R. solani in the field. The assay methods were treating R. solani sclerotia by Trichoderma conidial suspension (10^6cfu ml^-1) and scattering Thichoderma rice bran over the pepper root medium. The results showed that T02-25 was active against R.solani in both ways, and its control efficacy was 82.7% and 78.0%, respectively. In addition to comparison of the efficacy of the two application methods, the relationship of different factors in the control efficacy of Trichoderma against R. solani was discussed. 展开更多

Prevalence of Antibiotic Resistant Bacteria on Tomato Surfaces and Effectiveness of Disinfectants in Reducing the Microbial Load

This study was conducted to isolate and identify antibiotic resistant bacteria in fresh tomato sold in three different local markets of Dhaka city. On an average 〉 5.0 log CFU/g of natural microbial populations and coliform bacteria was present in the samples. Prevalence of Salmonella spp., E. coli, Listeria spp., and Yersinia spp., were recorded through cultivation dependent assay of tomato samples of different markets. Almost all of the isolates were uniformly resistant to rifampicin, erythromycin, clindamycin and oxacillin (100%), vancomycin (93%), amoxicillin (87%), whereas 60% of the isolates were found to be resistant to ampicillin and cephalexin. Antibiotic sensitivity test of the presumptive E. coli and Salmonella spp. and corresponding plasmid profiling indicated a correlation of plasmid mediated multidrug resistance (MDR) of these environmental bacteria. Washing with sanitizing agents such as acidified sodium chlorite (ASC) and 200 ppm chlorinated water was able to reduce _〈 2.0 log CFU/g of natural microflora and coliform bacteria. In contrast, ASC was able to reduce 〈 2.0 log CFU/g of other food borne pathogens and, chlorinated water was able to reduce up to undetectable level. Therefore, washing of tomato with 200 ppm chlorinated water could be useful in reducing the pathogen populations on tomatoes. 展开更多

The dynamics of a bacterial population exposed to the minimum inhibitory concentration (MIC) of an antibiotic is an important issue in pharmacological research. Therefore, a novel antibiotic susceptibility test is urgently needed that can both precisely determine the MIC and accurately select antibiotic-resistant strains from clinical bacterial populations. For this purpose, we developed a method based on Fick's laws of diffusion using agar plates containing a linear gradient of antibiotic. The gradient plate contained two layers. The bottom layer consisted of 15 mL agar containing the appropriate concentration of enrofloxacin and allowed to harden in the form of a wedge with the plate slanted such that the entire bottom was just covered. The upper layer consisted of 15 mL plain nutrient agar added with the plate held in the horizontal position. After allowing vertical diffusion of the drug from the bottom agar layer for 12 h, the enrofloxacin concentration was diluted in proportion to the ratio of the agar layer thicknesses. The uniform linear concentration gradient was verified by measuring the enrofloxacin concentration on the agar surface. When heavy bacterial suspensions were spread on the agar surface and incubated for more than 12 h, only resistant cells were able to form colonies beyond the boundary of confluent growth of susceptible cells. In this way, the true MIC of enrofloxacin was determined. The MICs obtained using this linear gradient plate were consistent with those obtained using conventional antibiotic susceptibility tests. Discrete colonies were then spread onto a gradient plate with higher antibiotic concentrations; the boundary line increased significantly, and gene mutations conferring resistance were identified. This new method enables the rapid identification of resistant strains in the bacterial population. Use of the linear gradient plate can easily identify the precise MIC and reveal the dynamic differentiation of bacteria near the MIC. This method allows the study of genetic and physiological characteristics of individual strains, and may be useful for early warning of antibiotic resistance that may occur after use of certain antirnicrobial agents, and guide clinical treatment. 展开更多
