The results showed the treatment of magnetized water soaking for 4 h promoted water absorption rate and amylase ac- tivities of seeds significantly, which accelerated the transformation process of en- dosperm starch to soluble sugar, resulting in emergence of 36 hours in advance under low temperature condition. Germination rate and germination potential of magnetized water soaking were higher than the contrast by 6.7% and 10.0%, which helped cultivate vigorous seedling.展开更多

However, in this report, we showed that both PS II and PS I could be formed in dark-grown lotus (Nelumbo nucifera Gaertn.) seedlings. Lots of evidence were given: First I during the dark-grown period, the single fluorescence emission peak at 679 nm in lotus embryo red-shifted and transformed into the normal PS II fluorescence emission; Simultaneously, PS I fluorescence emission at 730 nm appeared and increased obviously; Second, with partial denaturing SDS-PAGE method, PS I chlorophyll-protein complex could be clearly separated from 10 days dark-grown lotus seedlings; Third, the existence of Lhca1 was also proved by Western blots. Moreover, measurements of electron transfer rate demonstrated that both PS II and PS I core in dark-grown lotus seedlings were photochemically active.展开更多

SDS PAGE analysis of chlorophyll_protein complexes showed that there was only the light harvesting chlorophyll a/b protein complex from PSⅡ (LHCⅡ) precursor in chloroplast from lotus seeds germinated for 2 to 6 days, while LHC Ⅱ 1, and the chlorophyll_protein complex of PSⅠ (CPⅠ) appeared on the 8th day of germination and PSⅡ reaction center complex appeared later. Studies on the polypeptides composition of the chloroplast revealed the following results: 1) Small amount of the 27 kD polypeptide was synthesized in invisible light; 2) The 30 kD polypeptide existed previously in the plumules of the dry seeds; 3) The amount of the 30 kD polypeptide was more than any other polypeptides before germination and decreased gradually throughout germination, while the 27 kD polypeptide changed in the opposite way; 4) In the process of germination, measurement of the electron transport rate and the fluorescence induction kinetics at room temperature showed that PSⅡ activities and efficiency of primary light energy transformation were only experimentally measurable after 7 days of germination and gradually increased afterwards. At the same time, the chl a/b ratio rose from the lower value to normal; 5) The changes of chloroplast membrane components and its functions are concomitant in concert with that of the ultrastructure of chloroplast membranes during germination, as shown in our earlier work . The results have proved again that a different developmental pathway of chloroplast is likely to exist in the lotus plumules, which might provide an important clue for N. nucifera in having an unique position in the phylogeny of the angiosperm.展开更多

The results showed that 80 kinds of protein spots in endosperms of Gingkgo biloba were clear observed in the 2-DE spectrum. Protein molecular weights were in the range of 26-52kD, and their isoelectric points were in the range of 5.8-7.8. In the course of seed germination, 13 kinds of proteins were degraded, and 13 kinds of proteins were synthesized; 7 kinds of proteins with different molecular weights and isoelectric points of 35kD/pIr.8, 31kD/pIr.8, 29kD/pIr.8, 33kD/pI6.6, 33kD/pI 6.4, 34kD/pI7.7 and 31kD/pI7.7 were identified primarily as vegetative storage proteins (VSPs).展开更多

To reveal the relationship between germination and the 3'-5'-cyclic adenosine monophosphate(c AMP) content from the dormant(black or green-black) and non-dormant(yellow)chlamydospore in Ustiloginoidea virens, this study adopts ultrasonic-bath method and high-performance liquid chromatography(HPLC) method, for extraction c AMP content of the different color chlamydospore. The results demonstrated that, as for the freshly chlamydospores collected from the false smut balls during their germination, c AMP content of yellow chlamydospore appeared a slight growth during 0-12 h, and showed a rapid declining as the germination time extended(12-48 h). Above all, in yellow chlamydospores, the germination rate and the content of c AMP presented a very notably negative correlation(|r|=0.861 9>r0.01=0.834), but the correlation between germination rate and content of c AMP was not obvious in black chlamydospores. The germination rate and the content of c AMP presented a notably positive correlation under different storage period(0, 2, 4, 6, 8,10 months) of the two color chlamydospores(the yellow of chlamydospore: |r | = 0.785 1 > r0.05= 0.707; the black of chlamydospore: |r| = 0.957 9 > r0.05= 0.707).展开更多

Seeds were collected from 55-year-old plants in March 2012 in Chengdu Wenjiang Park in China and sown on illumination incubator at 25 ℃/18℃ (day/might) under 15 h light after soaked 24 h with 0, 50, 100 and 200 mg L-1 IAA, NAA solutions. The thousand seed weight ofJ. mimosifolia was 9.039 g and seed average vigor was 32.7%. Germination rate was 13.3% without treatment, and seeds with 50, 100 and 200 mg L-1 IAA treatment showed germination rate of 20.0%, 22.3% and 25.0%, respectively, whereas the rate was 18.3%, 20.0% and 23.3% in 50, 100 and 200 mg L-1 NAA treatment. There was significant difference between 100, 200 mg L-1 and other IAA treatment, but only 200 mg L-1 NAA treatment had significant difference among others. It showed that IAA could promote seed germination more significantly than NAA in the same concentration conditions. These results indicate that soaking by 100 and 200 mg L-1 IAA or 200 mg L-1 NAA for 24 h, seed germination rate ofJ. mimosifolia can be significantly improved and robust seedlings obtained from germinated seeds.展开更多

Poit. is an invasive alien weed commonly known as "Bana tulst" in Odisha belonging to the family Lamiaceae and widely distributed in different parts of India. The leaves of the plant have been reported to be used as stimulant, carminative, sudorific and also as a cure for parasitic cutaneous diseases besides the crude leaf extract is reported to be used against colic and stomach-ache. The leaves are the source of different alkaloids, terpenes and volatile oils having allelopathic effect. In view of this the present work was carried out with an aim to investigate the allelopathic effect upon germination of rice (Oryza sativa L.). The result indicated that the higher concentration of the leaf leachate could decrease the percentage of germination besides identified as a potential bio-herbicide.展开更多

L.) were investigated. The result showed that the extracts brought about a considerable inhibition in the germination of soybean seeds, reductions in the radicle and plumule lengths of soybean seedlings. Whereas, the extracts of maize inflorescence and rice husk had more inhibitory effects on the growth of soybean radicle, all the three extracts demonstrated pronounced inhibitory effects on the growth of the plumule. The retardation of soybean germination, radicle and plumule growths were concentration dependents as the degree of retardations increases with increase in the concentration of the extracts. Statistical analysis (ANOVA, P = 0.05) revealed that there were no significant differences in the germination, radicle and plumule growths at 24 and 48 h experimental time when compared to the control. However, statistical differences abound in the growth parameters at 72 and 144 h experimental time.展开更多
