利用H2O2/过氧化物酶(POD)/愈创木酚(GA)反应体系的产物(470 nm)吸光度去定量POD活性是目前常用方法之一,但产物不稳定且对比色皿有吸附现象是该反应的主要缺点之一。为了解决此问题,本研究利用GA的荧光特性建立了一种能准确测定... 利用H2O2/过氧化物酶(POD)/愈创木酚(GA)反应体系的产物(470 nm)吸光度去定量POD活性是目前常用方法之一,但产物不稳定且对比色皿有吸附现象是该反应的主要缺点之一。为了解决此问题,本研究利用GA的荧光特性建立了一种能准确测定POD活性的方法。用辣根过氧化物酶( HRP)作为测试样品得到了以下优化条件:0.5 mmol/L GA,0.5 mmol/L H2O2,0.05 mol/L PBS缓冲液(pH 6.0),反应温度20℃,样品耗量20μL;本方法的线性范围为500-60000 U/L(r=0.9993),RSD≤2.4%(n=11),检出限为385 U/L。在优化条件下,用白萝卜提取液作为测试样品,用本方法((9714依132) U/L)与比色法((9926依352) U/L)和循环催化流动分析法((9608依456) U/L)进行了对比,结果表明,3种方法相互一致性很好。 展开更多

本研究给出了流动注射分光光度系统(FI-SP)测定酶活性浓度(Ec)的通用公式Ec=kΔA/Δt;Δt是在酶促反应线性响应区域内试样塞的平均留存时间;ΔA为平均留存时间处对应的吸光度;k=n V/(εv L),V为样品注入点到流通池入口之间的管路... 本研究给出了流动注射分光光度系统(FI-SP)测定酶活性浓度(Ec)的通用公式Ec=kΔA/Δt;Δt是在酶促反应线性响应区域内试样塞的平均留存时间;ΔA为平均留存时间处对应的吸光度;k=n V/(εv L),V为样品注入点到流通池入口之间的管路总容积,n为酶底物与产物的反应摩尔比,ε为产物的摩尔吸收系数,v为注入的酶液体积,L为流通池光程;FI-SP系统确定后k为常数。在此基础上,将H2O2/过氧化物酶(POD)/愈创木酚(GA)反应体系导入FI-SP系统,建立了一种无需标准曲线的POD活性快速准确测定的新方法。此方法优化后的条件:载流磷酸盐缓冲液浓度为0.05 mol/L(p H 5.5),进样体积为40μL,反应盘管长度200 cm,1.5 mmol/L H2O2,3.5 mmol/L GA,0.4 mol/L HCl(清洗液),检测波长470 nm,产物的摩尔吸光系数为0.0125 L(μmol·cm)。本方法的重现性RSD〈0.38%(n=11),分析速度为40样/h,测定范围为450~7260 U/L,检出限为89 U/L。在优化条件及60℃下,用本方法测定了多种白萝卜中POD活性,结果与动力学曲线法完全相同。 展开更多

For choosing suitable summer-planting adzuki bean crop, this experiment adopted the method of indoor petri dish bioassay to study the allelopathy effect of adzuki bean, radish, cabbage and wheat under different concentrations of adzuki bean root aqueous extracts. The result showed that the adzuki bean had a strong inhibition towards its germination rate and germination index. With the increasing concentration of the adzuki bean root aqueous extract, the seed germination rate and germination index of the three kinds of receptor crops presented a decreasing trend. From the root length, spire length, the number of lateral roots, plant height, fresh mass and other morphological indexes of seedlings, it was concluded that with the increase of concentration of adzuki bean root aqueous extract, only wheat pre- sented a promotion effect while the other two kinds of receptor crops showed an inhibition effect, which were not significant at low concentrations. With the increase of concentration of adzuki bean root extracts, the SOD and POD activities of radish and wheat also increased, while the two kinds of enzyme activities of cabbage showed a decreasing trend. The MDA contents in seedlings of radish and wheat decreased, while the MDA content in cabbage increased significantly, which showed that wheat could be used as a rotation crop of adzuki bean, and radish also had a great application potential in adzuki bean crop rotation. 展开更多

Effect of Steaming, Steam-Drying and Frying on the Provitamins A and Ascorbic Acid Contents of Squash ( Cucurbita Spp.)

Squashes pulp is rich in provitamins A and could help to reduce this deficiency. Unfortunately, in Cameroon, squash pulp is more often reserved for the nutrition of sick people and domestic animals. This study aimed at encouraging the consumption of squash pulp to contribute to vitamin A needs. The contents of a-carotene, all-trans-β-carotene and ascorbic acid were determined respectively by HPLC and titration with 2.6 dichlorophenol-indophenol in five landraces of raw, steamed, steam-dried and fried squash pulp from Cameroon. Moisture and total lipid content were also determined. Peeled pulp squash of 5 cm slice was steamed at 90℃ for 30 min or at 85℃ for 30 min, sliced again at 5 mm thickness and dried at 80℃ during 4 h to obtained steamed or steam-dried squashes. To have fried squashes, 40 g of 1 mm thickness slices pulp were fried in boiling refined palm oil (free of carotenoids) bath at 150℃ during 7 and 10 min. The results obtained showed that steam-drying and frying of squashes leaded to water losses (89%-95%). As a consequence of this, the a-carotene, all-trans-β-carotene and ascorbic acid contents (expressed in g (100 g)-1 fresh portion) of steam-dried and fried squashes were significantly higher (P 〈 0.05) than those of steamed and raw pulp. However, the retention rate of provitamins A and ascorbic acid was more elevated in steamed than in steam-dried and fried squashes. These results suggest that steam-dried and fried squashes could contribute to fight against vitamin A deficiency while increasing availability of squashes. 展开更多

The Protective Role of β-Carotene and Hesperidin on Some Hematological and Myocardial Measurements against Imidacloprid Toxicity in Albino Rats

Neonicotinoids including IM (Imidacloprid) are widely used as plant systemic insecticides. Several studies have indicated that pesticide toxicity may be associated with the enhanced production of ROS (reactive oxygen species). Both β-carotene (I3C) and hesperidin (H) have an antioxidant property and quench free radicals. This study aimed to clarify the protective role of β-carotene and hesperidin as natural antioxidants on IM induced toxicity in hematological parameters and markers of cardiac muscle activity in male albino rats. The treatment of rats with IM showed a significant decrease in hemoglobin (Hb %), MCH (mean corpuscular hemoglobin), MCHC (mean corpuscular hemoglobin concentration), HCT (hematocrit) values and RBCs count comparing with control group. On the other hand, MCV (mean corpuscular volume), WBCs (white blood cells) and Pits (platelets) count pronounced a significant increase in IM group comparing to control. Also, αFP (plasma alpha fetoprotein), CEA (carcinoembryonic antigen), CK (creatine kinase), CK-MB (creatine phosphokinase myocardial band) and LDH (lactate dehydrogenase) clarify a significant increase in IM group comparing to control. Both β-carotene and hesperidin mitigate the deleterious effects of IM on previous parameters. β-Carotene and hesperidin may protect hematopoietic system and heart muscle against toxicity of IM. These improvements of the results clarify the protective effect of the used antioxidants. Conclusion: β-carotene and hesperidin, natural antioxidants, have a protective effect against IM evoked hematological and biochemical changes. 展开更多
