蒲金口服液对宫内感染/炎症致早产脑损伤大鼠脑组织OL、OPC细胞凋亡和Nogo-A、OMgp表达的影响

目的探讨蒲金口服液对宫内感染/炎症致早产脑损伤大鼠神经保护作用的机制。方法将80只胎龄<22 d的早产仔鼠随机分为8组(每组10只):早期药物干预组(蒲金口服液高剂量组、蒲金口服液低剂量组、银杏叶提取物组、模型组)和晚期药物干预... 目的探讨蒲金口服液对宫内感染/炎症致早产脑损伤大鼠神经保护作用的机制。方法将80只胎龄<22 d的早产仔鼠随机分为8组(每组10只):早期药物干预组(蒲金口服液高剂量组、蒲金口服液低剂量组、银杏叶提取物组、模型组)和晚期药物干预组。生后21 d龄进行神经行为学检测,并采用qRT-PCR和Western blot方法检测勿动蛋白-A(Nogo-A)和少突胶质细胞髓磷脂糖蛋白(OMgp)的表达,TUNEL法和A2B5、CNPase双标免疫荧光染色法检测少突胶质细胞(OL)、前体细胞(OPC)凋亡情况。结果LPS组早产仔鼠脑组织Nogo-A和OMgpmRNA和蛋白的表达均明显高于正常对照组(P<0.05),蒲金口服液高剂量、低剂量和银杏叶提取物干预组大鼠的神经行为学评分均优于模型组(P<0.05),且早期干预组高于晚期干预组(P<0.05)。蒲金口服液高剂量、低剂量和银杏叶提取物均能降低早产脑损伤大鼠脑组织Nogo-A、OMgp mRNA和蛋白的表达,减少OL和OPC的凋亡(P<0.05),其中蒲金口服液高剂量早期干预效果更显著(P<0.05)。结论蒲金口服液可以改善早产脑损伤大鼠的神经行为学表现,减少脑组织Nogo-A、OMgp的表达和OL、OPC的凋亡,具有神经保护的作用。 展开更多

目的分析蒲地蓝消炎口服液的有效活性成分及其治疗急性咽炎的分子机制。方法分别通过中药系统药理学数据库分析平台(TCMSP)数据库、Genecards数据库查找蒲地蓝消炎口服液中4味中药的有效活性成分及其靶点蛋白和人类急性咽炎相关的作用靶... 目的分析蒲地蓝消炎口服液的有效活性成分及其治疗急性咽炎的分子机制。方法分别通过中药系统药理学数据库分析平台(TCMSP)数据库、Genecards数据库查找蒲地蓝消炎口服液中4味中药的有效活性成分及其靶点蛋白和人类急性咽炎相关的作用靶点,构建有效活性成分-疾病靶点的网络图;蛋白互作网络通过String数据库进行构建,通过Metascape数据库进行GO和KEGG富集分析;最后使用分子对接技术对有效活性成分和靶点进行验证。结果经过筛选,蒲地蓝消炎口服液中的槲皮素、β-谷甾醇、汉黄芩素、刺槐黄素以及豆甾醇等多个关键活性成分,可能通过作用在细胞肿瘤抗原p53(cellular tumor antigen p53,TP53)、白细胞介素6(interleukin-6,IL-6)、白细胞介素8(interleukin-8,CXCL8)、过氧化物合酶2(prostaglandin G/H synthase 2,PTGS2)、白细胞介素10(interleukin-10,IL-10)、肿瘤坏死因子(tumor necrosis factor,TNF)等核心基因表达的蛋白上参与急性咽炎的治疗。KEGG富集分析所涉及的通路包括NF-kappaB信号通路(NF-kappa B signaling pathway)、癌症通路(Pathways in cancer)、HIF-1信号通路(HIF-1 signaling pathway)、TRP通道中炎症介质调节(inflammatory mediator regulation of TRP channels)等。结论蒲地蓝消炎口服液可通过多成分、多靶点、多通路的复杂作用机制发挥抗炎作用,进而对急性咽炎起到治疗作用。 展开更多

DNA fingerprinting among members of the Chinese drug Pu Gong Ying(Taraxacum mongolicum Hand,-Mazz.)and six adulterants of Tu Gong Ying were demonstrated with random-primed polymerase chain reaction(PCR)including arbitrarily primed polymerase chain reaction(AP-PCR)and random amplified polymorphic DNA(RAPD).Distinctive,reproducible genomic fingerprints from DNA from 7 species belonged to Compositae were generated with two long(20 and 24 mer)and one short(10 mer)randomly chosen primers.The Pu Gong Ying can be differentiated from six species of Tu Gong Ying according to the banding pattems of their amplified DNA on agarose gels.The results showed that AP-PCR and RAPD methods can be used for identifying Chinese drugs.Moreover,the Similarity Indexes of the genomic DNA fingerprints showed that Pu Gong Ying and its adulterants are unrelated.Therefore,AP-PCR and RAPD methods can be used for identifying Chinese drugs. 展开更多

[Objective] To study the preparation of Zipu Capsule and its clinical effect on the treatment of chronic periodontitis. [Method] The extraction, concentration, drying and preparation process of the raw herbal material... [Objective] To study the preparation of Zipu Capsule and its clinical effect on the treatment of chronic periodontitis. [Method] The extraction, concentration, drying and preparation process of the raw herbal materials for Zipu Capsule were selected out. And 60 cases of chronic periodontitis patients were randomly assigned to two groups and were treated with conventional therapy and nursing care, while the treatment group was added with taking the Zipu capsule, to compare the cura- tive effects of the two groups. [Result] Both of two groups showed decreases in gingival index (GO, periodontal pocket depth (PD) and attachment loss values (AL) after treatment, and those of the treatment group was significantly lower than those of the control group (P〈0.05). The total curative effective rate of the treatment group was 91.17%, and 76.92% in the control group, and the differences between the two groups reached the significant level (P〈0.05). [Conclusion] Zipu Capsule had definite therapeutic effect in treatment of chronic periodontitis and less adverse re- actions, which laid the foundation for the further development into drug products. 展开更多

[Objective] This study aimed to investigate the chromosomes and karyotypes of three seagrass species of hydrocharitaceae-Enhalus acoroides, Thalassia hemprichii and Halophila minor collected from Li’an Lagoon, Hainan Province, China. [Method] The root-tips of E. acoroides, T. hemprichii and the axillary buds of H. minor were selected as the materials in this study. The conventional crushing method was used to prepare the slice specimens of chromosomes, and the karyotypes of the three species were analyzed. [Result] The chromosome numbers of E. acoroides, T. hemprichii and H. minor were 2n=18, 18, 28, respectively. The karyotype formulas were K (2n)=18= 12m+6sm, K(2n)=18=12m+6sm and K(2n)=28=16m+8sm+4st, respectively. According to the standard of Stebbins, the karyotypes of E. acoroides and T. hemprichii were 2A, and that of H. minor was 2B. There was no B chromosome and satellite in these three species. [Conclusion] According to the comparison on the karyotypes of this three species, E. acoroides and T. hemprichii are similar in karyotypes to each other, indicating that there is close, inter-relationship between E. acoroides and T. hemprichii. 展开更多

目的探讨新蒲饮联合奥美拉唑治疗幽门螺杆菌相关性胃炎疗效的基础研究。方法将50只C57BL/6(B6)小鼠采用细菌灌胃的方法建立小鼠幽门螺杆菌(helicobacter pylori,Hp)相关性胃炎模型,造模成功后随机分为对照组、模型组、单纯新蒲饮组(新... 目的探讨新蒲饮联合奥美拉唑治疗幽门螺杆菌相关性胃炎疗效的基础研究。方法将50只C57BL/6(B6)小鼠采用细菌灌胃的方法建立小鼠幽门螺杆菌(helicobacter pylori,Hp)相关性胃炎模型,造模成功后随机分为对照组、模型组、单纯新蒲饮组(新蒲饮组)、新蒲饮联合奥美拉唑组(新奥组)和阿莫西林+克拉霉素+奥美拉唑肠溶片组(胃三联组),后各给药组连续给药14 d。通过快速尿素酶法及吉姆萨染色观察Hp的分布及定植情况,采用酶联免疫吸附测定(enzyme linked immunosorbent assay,ELISA)检测血清白细胞介素-8(interleukin-8,IL-8)的表达,免疫组化以及蛋白质印迹法检测IL-8、核因子-κB(nuclear factor-k-gene binding,NF-κB)的蛋白表达,实时荧光定量PCR(quantificational real-time PCR,qRT-PCR)检测IL-8、NF-κB的信使RNA(message RNA,mRNA)表达。结果通过尿素酶检测及吉姆萨染色可以发现,与模型组比较,各给药组小鼠胃黏膜Hp明显减少,胃三联组与新奥组较为明显,新蒲饮组次之;治疗前,模型组IL-8、NF-κB蛋白和mRNA表达水平及血清IL-8含量均高于对照组(P<0.05);治疗后,与模型组比较,新蒲饮组、新奥组及胃三联组IL-8、NF-κB蛋白和mRNA表达水平及血清IL-8含量均明显降低(P<0.05);进一步分析发现,新奥组和胃三联组IL-8、NF-κB及血清IL-8含量明显低于新蒲饮组。结论动物实验表明新蒲饮能够治疗Hp相关性胃炎,联合奥美拉唑效果更优,与胃三联效果相似。由此可见,新蒲饮联合奥美拉唑有望成为治疗Hp相关性胃炎的新方法。 展开更多

This study is intended to explore the chemical differences of Acori Tatarinowii Rhizoma (ATR) samples collected from two habitats, Sichuan and Anhui provinces, China. Gas chromatography-mass spectrometry (GC-MS) was applied to establishing the quantitative chemical fingerprints of ATRs. A total of 104 volatile compounds were identified and quantified with the information of mass spectra and retention index (RI). Furthermore, least absolute shrinkage and selection operator (LASSO), a sparse regularization method, combined with subsampling was employed to improve the classification ability of partial least squares-discriminant analysis (PLS-DA). After variable selection by LASSO, three chemical markers,β-elemene, α-selinene and α-asarone, were identified for the discrimination of ATRs from two habitats, and the total classification correct rate was increased from 82.76% to 96.55%. The proposed LASSO-PLS-DA method can serve as an efficient strategy for screening marked chemical components and geo-herbalism research of traditional Chinese medicines. 展开更多

To evaluate the effect of tectonic deformation on coal reservoir properties, we provide an analysis of the types of tectonically deformed coal, macroand microscopic deformation and discuss pore structural characteristics and connectivity based on samples from the Puhe and Shanchahe coal mines. Our research shows that the tectonically deformed coal mostly includes cataclastic structural coal, mortar structural coal and schistose structural coal of a brittle deformation series. The major pore structures of different types of tectonically deformed coal are transitional pores and micropores. The pore volumes of macropores and visible fracture pores produced by structural deformations vary over a large range and increase with the intensity of tectonic deformation. Mesopores as connecting passages develop well in schistose structural coal. According to the shapes of intrusive mercury curves, tectonically deformed coal can be divided into parallel, open and occluded types. The parallel type has poor connectivity and is relatively closed; the open type reflects uniformly developed open pores with good connectivity while the occluded type is good for coalbed methane enrichment, but has poor connectivity between pores. 展开更多

To probe the influence and the adverse-resistance characteristics of wetland plants in presence of silver nanoparticles (AgNPs), the changes in the physiological and biochemical characteristics (including the superoxide dismutase (SOD) activity, catalase (CAT) activity, peroxidase (POD) activity, soluble protein content, and chlorophyll content) of Typha orientalis exposed to different concentrations of AgNPs solutions (0, 0.1, 1, 20 and 40 mg/L) were explored. Meantime, the accumulation of silver content in these plants was revealed. The results show that under low concentrations of AgNPs, the SOD and POD activities in the leaves of Typha orientalis are strengthened to different degrees. However, high concentrations of AgNPs inhibit the activities of SOD and POD. Under the stress of different concentrations of AgNPs, the CAT activities are inhibited initially and later recovered to some extent. Under the stress of low concentrations of AgNPs, the soluble protein content in the leaves of Typha orientalis increases significantly, but decreases more significantly with increasing concentrations of AgNPs. Low concentrations of AgNPs promote chlorophyll synthesis in the leaves of Typha orientalis , but the chlorophyll content subsequently falls to pre-stress levels. In contrast, high concentrations of AgNPs cause a certain inhibition to generate chlorophyll. Meanwhile, the results show that the silver concentrations of plant tissues increase with the exposure of concentrations of AgNPs and they have a positive relationship with the exposure of concentrations of AgNPs. 展开更多

This whole procedure was repeated for 5 d. We determined the pancreatic weight/body weight ratio, the levels of pancreatic HSP60 and HSP72, and the secretion of pro-inflammatory cytokines. Repeated CCK octapeptide treatment resulted in typical laboratory and morphological changes of experimentally-induced pancreatitis.RESULTS: TO significantly decreased the pancreatic weight/body weight ratio in CCK octapeptide-induced acute pancreatitis. TO also increased the pancreatic levels of HSP60 and HSP72. Additionally, the secretion of IL-6 and TNF-α decreased in the animals treated with TO.CONCLUSION: TO may have a protective effect against CCK octapeptide-induced acute pancreatitis. 展开更多

In this study,the fifth-generation Pennsylvania State University-National Center for Atmospheric Research (PSU-NCAR) Mesoscale Model (MM5) is used to simulate Typhoon Mindulle (2004) at high resolution (3-km grid size).The data from measurements show that in the upper atmosphere the existence of an upper jet is important to the transition cyclone.When Mindulle moved to the area of the upper jet entrance,where high-altitude divergence existed, the pumping of the high-altitude divergence would enhance the vertical motion and low-level cyclone convergence. The enhanced vertical motion was confirmed by the simulation results and indicated that the existence of upper divergence enhanced the vertical motion which was favorable for the maintenance of Typhoon Mindulle.The process of extratropical transition (ET) and re-intensification always accompanies the process of cold air invasion. This process enhances the baroclinicity of the atmosphere and the formation of front at high altitudes, which converts baroclinic potential energy into kinetic energy and strengthens the cyclone vortex.The distributions of equivalent potential temperature (θe) and temperature anomalies show that the warm-core of the typhoon at the tropopause aids the re-intensification of the system. As the typhoon reenters the ocean, latent heat flux (LHF) increases in the north and west and the strong reflectivity and vertical motion occur in the east and southeast,and the west.With the re-intensification of the typhoon the wind field evolves from an oval to a circle at the lower atmosphere, the area coverage by high winds increases, and the distribution of the tangential wind shows an asymmetric pattern. 展开更多

The resultant oil contained eugenol (48.92%), caryophyllene (18.55%), α-... Essential oil from clove buds Syzygium aromaticum L. Merr. & Perry (Myrtaceae) was extracted using petroleum ether in Soxhlet apparatus. The resultant oil contained eugenol (48.92%), caryophyllene (18.55%), α-caryophyllene (3.25%), eugenol acetate (23%), cis-13-docosenamide (3.2 1%), presenting more than 96% of the oil. This oil was examined as a fumigant and repellent agent against adults of the red flour beetle, Tribolium castaneum. Data showed that various concentrations of the oil have revealed dramatic repellent activity against the tested insect, where it gave 100% repellency by 1.0, 0.8, and 0.2% clove oil at 4, 8, 10 min, respectively. The RTs0 and RT95 were 1.1 and 8.0 min at 0.2% and 0.4, 2.6 min at 0.8%, respectively. In addition, fumigation assay has also exhibited strong fumigant activity toward the adults of T. castaneum. At 100 μL oil/L air, mortality was 75, 80 and 100% after 6, 7, and 8 days exposure period with LCs0 and LC95 17 and 70 μL/L air, respectively. The utilization of clove oil for its potential effects against stored product insect is discussed. 展开更多
