矿质元素和有机肥对萝卜产量和“蓝心”率的影响 被引量:1

为保证萝卜产量的同时,又避免萝卜"蓝心"的发生,采用大田试验和不同的储藏方法,研究矿质元素和有机肥不同配施对萝卜产量和"蓝心"问题的影响。结果表明:NPK2(尿素+钙镁磷肥+硫酸钾)+鸡粪处理产量最高,达151.67t... 为保证萝卜产量的同时,又避免萝卜"蓝心"的发生,采用大田试验和不同的储藏方法,研究矿质元素和有机肥不同配施对萝卜产量和"蓝心"问题的影响。结果表明:NPK2(尿素+钙镁磷肥+硫酸钾)+鸡粪处理产量最高,达151.67t·hm-2,显著高于NP(CK)处理;NPK1(尿素+钙镁磷肥+氯化钾)和NPK1+鸡粪处理的萝卜出现"蓝心"率分别为20.0%和16.7%,明显高于未施用氯化钾的处理,且冷藏易诱发萝卜提早出现"蓝心"现象。经土壤样品和植株样品检验萝卜心泛蓝现象属Cu离子和Cl离子含量较高,为此,提出萝卜高产优质栽培模式,即萝卜种植地农家有机肥与氯化钾不要混合施用,钾肥应使用硫酸钾为佳,可采用尿素+钙镁磷肥+硫酸钾+鸡粪的施肥方式。 展开更多

目的建立蓝心草颗粒的定性定量方法。方法对蓝心草颗粒中板蓝根、穿心莲、鱼腥草进行薄层色谱鉴别。用HPLC法测定穿心莲内酯、脱水穿心莲内酯的含量,采用Dikma DiamonsiL C18柱,以甲醇-水(48∶52)为流动相,流速为1.0 mL·min-1,检... 目的建立蓝心草颗粒的定性定量方法。方法对蓝心草颗粒中板蓝根、穿心莲、鱼腥草进行薄层色谱鉴别。用HPLC法测定穿心莲内酯、脱水穿心莲内酯的含量,采用Dikma DiamonsiL C18柱,以甲醇-水(48∶52)为流动相,流速为1.0 mL·min-1,检测波长为240 nm。结果薄层色谱斑点清晰,阴性样品无干扰。穿心莲内酯进样量在0.036 6~1.464μg与峰面积线性关系良好,r=0.999 2,平均回收率为97.6%,RSD为1.3%(n=9);脱水穿心莲内酯进样量在0.041 9~1.677μg与峰面积线性关系良好,r=0.999 9,平均回收率为98.9%,RSD为1.7%(n=9)。结论本法专属性强,准确度高,重现性好,可作为该产品质量控制方法。 展开更多

A monoclonal antibody (MAb) specific for the bluetongue virus (BTV) group specific antigen (VP7) was characterized for its reactivity with purified virus and recombinant BTV VP7 (rVP7) protein and its suitability for use in the sandwich ELISA.The MAb,designated as 5B5 was specific to VP7 and belongs to IgG2a subclass and was selected for the development of the sELISA in this study.The MAb had a titer of 1:25 with BTV and 1:2 with the rVP7 protein.The sELISA is based on capturing of BTV antigen with VP7 specific MAb followed by detection using BTV polyclonal antiserum raised in rabbits.The assay was evaluated with six cell culture adapted serotypes of BTV that have been isolated from India,1,2,15,17,18 and 23.The assay could detect BTV antigen as early as day 8 in blood.It was also successfully applied for the detection of BTV group specific antigen in clinical samples of blood,washed RBCs,buffy coat and plasma.A total of 102 field samples from animals,suspected of being infected with BTV,were tested and 29.42% were positive.The blood samples were also amplified in cell culture which improved the sensitivity of the assay.Results confirmed that the sELISA is rapid and specific. 展开更多

Interstitial Cajal-like cells are a distinct type of interstitial cell with a wide distribution in mammalian organs and tissues,and have been given the name"telocytes".Recent studies have demonstrated the potential roles of telocytes in heart development,renewal,and repair.However,further research on the functions of telocytes is limited by the complicated in vivo environment.This study was designed to construct engineered heart tissue(EHT)as a three-dimensional model in vitro to better understand the role of telocytes in the architectural organization of the myocardium.EHTs were constructed by seeding neonatal cardiomyocytes in collagen/Matrigel scaffolds followed by culture under persistent static stretch.Telocytes in EHTs were identified by histology,toluidine blue staining,immunofluorescence,and transmission electron microscopy.The results from histology and toluidine blue staining demonstrated widespread putative telocytes with compact toluidine blue-stained nuclei,which were located around cardiomyocytes.Prolongations from the cell bodies showed a characteristic dichotomous branching pattern and formed networks in EHTs.Immunofluorescence revealed positive staining of telocytes for CD34 and vimentin with typical moniliform prolongations.A series of electron microscopy images further showed that typical telocytes embraced the cardiomyocytes with their long prolongations and exhibited a marked appearance of nursing cardiomyocytes during the construction of EHTs.This finding highlights the great importance of telocytes in the architectural organization of EHTs.It also suggests that EHT is an appropriate physical and pathological model system in vitro to study the roles of telocytes during heart development and regeneration. 展开更多
