(Ru-biaceae) is a medicinal herbwidely distributed in northeast Asian countries. In traditional Chinese medicine, it has the effectof 'clearing away heat and toxic material, promoting blood circulation and removing blood stasis'.It is a well known Chinese folk-medicine used for the treatment of appendicitis, sore throat, mumps, acne, sebo-rheic dermatitis and various kinds of tumors, such as tumors of digestive tract, carcinoma of liver. It was reported that the MeOH extract of H. diffusa demonstrated a significantantitumor activity and ursolic acid succeeded in being isolated from the MeOH extract as an activecomponent . Shan BN, et al suggested that the direct aqueous extract of H. diffusa hadimmuno-modulating activity and antitumor activity in vitro through stimulating the immune system tokill or engulf tumor cells. But regarding anti-tumor activity in vivo of water soluble extracts fromH. diffusa, no detail was reported. Therefore, we prepared water soluble extracts (H_1 and H_2)from H. diffusa and evaluated their anti-tumor property in vivo experiments as well as protectiveeffect on chemo-therapeutic damage.展开更多

Methods The venom was isolated by FPLC column chromatography consistingof DEAE Sepharose FF and Source 30S. The cytotoxic activity on tumor cells was detected by MITmethod. Purity and molecular weight were determined by SDS-PAGE (silver staining). Their stabilitiesto temperature and pH were also detected. Results Two pure cytotoxins named ACTX-6 and ACTX-8 wereobtained. Their molecular weights are 98 kDa and 27 kDa, respectively. ACTX-6 consists of twosubunits bonded together by disulfide bonds. Conclusion ACTX-6 and ATCX-8 have highest inhibitoryactivity on lung cancer cell A549. ACTX-6 is stable to heat while ACTX-8 not. ACTX-6 is stablebetween pH 7-9 and ACTX-8 between pH 6 - 9.展开更多

Enzyme-linked immuno-sorbent assay (ELISA) and coagulating time method were used to assess antigenic stability and coagulating stability, respectively. The change of antigenic activities and coagulating activities of Defibrase in the same buffer solutions (pH 6, 7 and 8, with the exception of pH 3.6) showed similar tendency to decline with the time. Concentrated Defi-brase was relatively stable at neutral pH 6~7, more than 95% of its initial activities (100BUmL-1) was kept after a 10-day storage at 40 oC, whereas in pH 3.6 and pH 9 buffer solutions, diluted Defibrase was very labile. Addition of Triton X-100 or bovine serum albumin could effectively prevent loss of Defibrase by minimizing adsorption of De-fibrase to plastic surface (P<0.005). Concentration of Defibrase could also affect its stability in aqueous solutions.展开更多

Venom produced by C. nozakii during mass agglomerations can contaminate seawater resulting in death of the halobios and seriously damage commercial fisheries. Swimmers and fishermen commonly suffer painful stings from this j ellyfish, resulting in local edema, tingling, breathing difficulties, depressed blood pressure and even death. Such effects arise from the complex mixture of biologically active molecules that make up jellyfish venom. In the present study, the hemolytic activity of venom from tentacles of C. nozakii and factors affecting its activity were assayed. The HUso (defined as the amount of protein required to lyse 50% of erythrocytes) of the venom against dove and chicken erythrocytes was 34 and 59 gg/mL, respectively. Carboxylmethyl chitosan and glycerol could increase hemolytic activity at concentrations greater than 0.06% and 0.2 mol/L, respectively.展开更多
