Methods The expressions of MAP1B protein and MAP1B mRNA in...Objective To explore the regulatory effect of fragile X mental retardation protein (FMRP) on the translation of microtubule associated protein 1B (MAP1B). Methods The expressions of MAP1B protein and MAP1B mRNA in the brains of 1-week and 6-week old fragile X mental retardation-1 (FmrI) knockout (KO) mice were investigated by immunohistochemistry, Western blot, and in situ hybridization, with the age-matched wild type mice (WT) as controls. Results The mean optical density (MOD) of MAP1B was significantly decreased in each brain region in KO6W compared with WT6W, whereas in KO1W, this decrease was only found in the hippocampus and cerebellum. MAP1B in 6-week mice was much less than that in 1-week mice of the same genotype. The results of Western blot and in situ hybridization showed that MAP1B protein and MAP1B mRNA were significantly decreased in the hippocampus of both KO1W and KO6W. Conclusion The decreased MAP1B protein and MAP1B mRNA in the Fmrl knockout mice indicate that FMRP may positively regulate the expression of MAP1B.展开更多

[Method] By difference method with different amount of alternative feeds (20% -50%), two4 x4 Latin- square-designed trials were taken on eight castrated male pigs [ Yorkshire x Landrace x Neijiang pig, initial body-weight: (46 ±2) kg ] to deter- mine the apparent digestible energy (ADE) of the eight kinds of plant protein supplement commonly used in China, that is, corn gluten meal (sol.), soybean meal ( sol. ), fababean, pea, rapeseed meal ( sol. ), sesame meal ( sol. ), rapeseed meal ( exp. ) and cotton seed meal (sol.). [Resultl (1) Fiber was the most important factor to estimate the ADE of plant protein supplement in pigs, and ADF was the best one. (2) The most effective equations were as below: ( 1 ) OE (kJ/kg DM) = 14 741.86 - 185.01ADF+54.01SCHO+22.45CP ( R =0.988,RSD= 67.9,P〈0.01 ) ; (2) DE (kJ/kg DM) =22 223.26 -209.58ADF+26.79SCHO-1.09GE ( Ff =0.989,RSD=66.9, P〈0.01 ) . Elastic behaviors of protein-like chains are investigated by Pruned-Enriched-Rosenbluth method and modified orientation-dependent monomer-monomer interactions model. The protein-like chain is pulled away from the attr...Elastic behaviors of protein-like chains are investigated by Pruned-Enriched-Rosenbluth method and modified orientation-dependent monomer-monomer interactions model. The protein-like chain is pulled away from the attractive surface slowly with elastic force acting on it. Strong adsorption interaction and no adsorption interaction are both considered. We calculate the characteristic ratio and shape factor of protein-like chains in the process of elongation. The conformation change of the protein-like chain is well depicted. The shape of chain changes from “rod” to “sphere” at the beginning of elongation. Then, the shape changes from “sphere” to “rod”. In the end, the shape becomes a “sphere” as the chain leaves away from the surface. In the meantime, we discuss average Helmoholtz free energy per bond, average energy per bond, average adsorbed energy per bond, average α-helical energy per bond, average β-sheet energy per bond and average contact energy per bond. On the other hand, elastic force is also studied. It is found that elastic force has a long plateau during the tensile elongation when there exists adsorption interaction. This result is consistent with SMFS experiment of general polymers. Energy contribution to elastic force and contact energy contribution to elastic force are both discussed. These investigations can provide some insights into the elastic behaviors of adsorbed protein chains.展开更多
