In this study, we explored the relationship between MPs procoagulant activity and characteristics of atherosclerotic plaque detected by 64-slice computed tomography angiography (CTA). Methods In 127 consecutive patients with CAD but without acute coronary syndrome and who under went 64-slice CTA, MPs procoagulant activity in plasma Coy a thrombin generation test), soluble form of lectin-like oxidized low-density lipoprotein receptor-1 (sLOX-1) and N(epsilon)-(carboxymethyl) lysine (CML) circulating levels (by ELISA) were measured. A quantitative volumetric analysis of the lumen and plaque burden of the vessel wall (soft and calcific components), for the three major coronary vessels, was performed. The patients were classified in three groups according to the presence of calcium volume: non-calcified plaque (NCP) group (calcium volume (%) = 0), moderate calcified plaque (MCP) group (0 〈 calcium volume (%) 〈 1), and calcified plaque (CP) group (calcium volume (%) 〉 1). Results MPs procoagulant activity and CML levels were higher in MCP group than in CP or NCP group (P = 0.009 and P = 0.027, respectively). MPs procoagulant activity was positively associated with CML (r = 0.317, P 〈 0.0001) and sLOX-1 levels (r = 0.216, P = 0.0025). Conclusions MPs procoagulant activity was higher in the MCP patient group and correlated positively with sLOX-1 and CML levels, suggesting that it may characterize a state of blood vulnerability that may locally precipitate plaque instability and increase the risk of subsequent major cardiovascular events.展开更多

The molecular mechanism by which Tenl participates in telomere length regulation and chromosome end protection remains elusive. In this work, we observed a weak interaction of Cdc13p and Tenlp in a gelfiltration analysis using purified recombinant Cdc13p and Tenlp. Tenlp itself exhibits a weak DNA-binding activity, but enhances the telomeric TG1-3 DNA-binding ability of Cdc13p. Cdc13p is communoprecipitated with Tenlp. In the mutant ten1-55 or ten1-66 cells, the impaired interaction between Tenlp and Cdc13p results in much longer telomeres, as well as a decreased association of Cdc13p with telomeric DNA. Consistently, the Ten1-55 and Ten1-66 mutant proteins fail to stimulate the telomeric DNA-binding activity of Cdc13p in vitro. Disturbances in the metabolism of lipoprotein profiles and oxidative stress in hemodialyzed (HD) and post-renal transplant (Tx) patients are proatherogenic, but elevated concentrations of plasma high-density lipop...Disturbances in the metabolism of lipoprotein profiles and oxidative stress in hemodialyzed (HD) and post-renal transplant (Tx) patients are proatherogenic, but elevated concentrations of plasma high-density lipoprotein (HDL) reduce the risk of cardiovascular disease. We investigated the concentrations of lipid, lipoprotein, HDL particle, oxidized low-density lipoprotein (ox-LDL) and anti-ox-LDL, and paraoxonase-1 (PON-1) activity in HD (n=33) and Tx (n=71) patients who were non-smokers without active inflammatory disease, liver disease, diabetes, or malignancy. HD patients had moderate hypertriglyceridemia, normocholesterolemia, low HDL-C, apolipoprotein A-I (apoA-I) and HDL particle concentrations as well as PON-1 activity, and increased ox-LDL and anti-ox-LDL levels. Tx patients had hypertriglyceridemia, hypercholesterolemia, moderately decreased HDL-C and HDL particle concentrations and PON-1 activity, and moderately increased ox-LDL and anti-ox-LDL levels as compared to the reference, but ox-LDL and anti-ox-LDL levels and PON-1 activity were more disturbed in HD patients. However, in both patient groups, lipid and lipoprotein ratios (total cholesterol (TC)/HDL-C, LDL-C/HDL-C, triglyceride (TG)/HDL-C, HDL-C/non-HDL-C, apoA-I/apoB, HDL-C/apoA-I, TG/HDL) were atherogenic. The Spearman's rank coefficient test showed that the concentration of ox-LDL correlated positively with HDL particle level (R=0.363, P=0.004), and negatively with TC (R=-0.306, P=0.012), LDL-C (R=-0.283, P=0.020), and non-HDL-C (R=-0.263, P=0.030) levels in Tx patients. Multiple stepwise forward regression analysis in Tx patients demonstrated that ox-LDL concentration, as an independent variable, was associated significantly positively with HDL particle level. The results indicated that ox-LDL and de- creased PON-1 activity in Tx patients may give rise to more mildly-oxidized HDLs, which are less stable, easily undergo metabolic remodeling, generate a greater number of smaller pre-13-HDL particles, and thus accelerate reverse cholesterol transport, which may be beneficial for Tx patients. The effects of the concentration of Ag+ ions, BSA and ammonia, reactive temperature on the silver morphology and particle sizes were investigated. SEM, TEM and XRD were employed to characterize the morphology and structures of the prepared silver mirco/nano-particles. The results show that the spherical silver particle with smooth surface and narrow size distributions can be obtained by controlling the concentration of Ag+ ions, BSA, reaction temperature, etc. By controlling the above parameters, the silver spherical mirco/nano-particles with particle sizes ranging from 0.2 to 2.3 m can be well prepared, which is expected to be used in manufacturing high performance electronic pastes.展开更多

METHODS: Thirty-two SD rats were divided ran- domly into control group A (given regular feed), model group B (food high in sugar and fat), inter- vention group C (food high in sugar and fat as well as regular doses of Danqidihuang Granules), and in- tervention group D (food high in sugar and fat as well as double doses of Danqidihuang Granules). The interventions were for 8 weeks. Motion, change in color, body weight, and food intake, as well as plasma lipids (including low-density lipopro- tein-cholesterol (LDL-C), high-density lipopro- tein-cholesterol (HDL-C), total cholesterol (TC) and triglyceride (TG), fasting blood glucose (FBG), fast-ing insulin (FINs) levels, insulin sensitivity index (ISI), and insulin resistance index (HOMO-IR) were observed. RESULTS: At the end of the second week of the ex- periment, the appetite and activities of rats in groups B, C and D decreased significantly com- pared with group A. The fur of the rats in those three groups was curly. After the fourth week, the activities, food intake and color of rats in group B were worse than those in groups C and D, but there were no significant differences in weight (P〉0.05). Compared with group A, LDL-C, TC, FBG and HO- MO-IR in model group 13 were increased significant- ly (P〈0.05), whereas the FINs and ISI increased obvi- ously (P〈0.05). The levels of LDL-C and TC in group D was decreased obviously compared with those in group C, and HOMO-IR in group D was less than that in group B CP〈0.05). CONCLUSION: Danqidihuang Granules helped to prevent and improved the insulin resistance of rats.展开更多
