药物对小鼠胃片GABA摄取的影响

本文应用同位素示踪技术和离体孵育小鼠胃片的方法,探讨了与调节胃功能活动的GABA相关的药物对胃片摄取GABA的影响。实验显示,GABA-T抑制剂AOAA在低剂量时(1~5mmol·L^(-1))可抑制胃片摄取GABA(P<0.01),而高剂量时(10mmol·... 本文应用同位素示踪技术和离体孵育小鼠胃片的方法,探讨了与调节胃功能活动的GABA相关的药物对胃片摄取GABA的影响。实验显示,GABA-T抑制剂AOAA在低剂量时(1~5mmol·L^(-1))可抑制胃片摄取GABA(P<0.01),而高剂量时(10mmol·L^(-1))能促进GABA摄取(P<0.05)。蝇(?)醇(Muscimol),是一种GABA_A受体激动剂,0.5μg·ml^(-1)时能促进胃片摄取GABA(P<0.01)。胆碱能神经M型受体阻断剂阿托品(0.01~0.25mg·ml^(-1))能抑制胃片摄取GABA(P<0.01)。盐酸(0.01~0.05N)和胃蛋白酶(0.05~1mg·ml^(-1))二者都能促进胃片摄取GABA(P<0.05或0.01)。这些结果提示:胃的GABA摄取系统可能有一种负反馈式自身调节机制。即当GABA调节胃功能活动增强时,它会加速GABA摄取,结果是尽快地终止GABA对胃的效应;相反当GABA调节胃功能活动减弱时,可抑制GABA摄取,结果是保持GABA对胃的作用,从而维持胃功能的稳态。 展开更多

[Objective] This study aims to conduct cloning and sequence analysis of ADH gene in D. Antiqua. [Method] Full-length cDNA of ADH gene in D. antiqua was cloned by using RACE technology (GenBank access number: JQ66600... [Objective] This study aims to conduct cloning and sequence analysis of ADH gene in D. Antiqua. [Method] Full-length cDNA of ADH gene in D. antiqua was cloned by using RACE technology (GenBank access number: JQ666006). Analysis of the homology, characteristics and functional domains of ADH sequence and the phy- Iogenetic relationship to other dipteran ADH were conducted. [Result] The full length of ADH cDNA is 1 088 bp containing a 771 bp of ORF, encoding 256 amino acids, with a calculated relative molecular weight of 30.80 kDa and a theoretical isoelectric point of 8.22. The deduced amino acid sequence shares the highest homology with Glossina morsitans morsitans based on homological analysis and phylogenetic analysis. [Conclusion] This study provides basis for further research of ADH gene. 展开更多

Phenol oxidase in Drosophila melanogaster occurs as folded phase precursors designated as prophenol oxidase A1 and A3, and prophenol oxidase is activated with alcohol, especially 2-propanol, within a few minutes as unfolded-phase in vitro. To clarify a common effect of alcohols on proteins and peptides, the extract containing prophenol oxidase protein was prepared. Phenol oxidase activity activated with 2-propanol has been maintained stable at least 24 hours remains as it is. Protein of prophenol oxidase was not denatured opposite hypnoses known as the instability of protein with alcohol. Activated prophenol oxidase with 2-propanol remain enzyme activity with no aggregation, stable, renaturation, and the refolding phenomena occurred around the active phase within the catalytic active center of prophenol oxidase protein in Drosophila melanogaster. This study is important to induce the wide range applications of the effect in many fields for rational drag design. 展开更多

The developmental functi... Lipids are essential membrane structural components and important signal carriers. The major enzymatic metabolisms of various lipids (phospholipid, sphingolipid, cholesterol) are well studied. The developmental function of lipid metabolism has remained, for the most part, elusive. With the help of new techniques and model organisms, the important roles of lipid metabolism in development just start to emerge. Drosophila spermatogenesis is an ideal system for in vivo studies of cytokinesis and membrane remodeling during development. The metabolic regulators of many lipids, including phosphatidylinositol (PI) lipids, fatty acids and cholesterol, are reported to play critical roles in various steps during Drosophila spermatogenesis. In this mini-review, we summarized recent findings supporting a tight link between lipids metabolism and Drosophila sperm development. 展开更多
