血小板转化生长因子与移植心脏血管病和纤维化关系的研究

目的探讨血小板转化生长因子(PDGF)与移植心脏排斥反应之间的关系。方法建立大鼠颈部心脏移植的动物模型。采用供体脾细胞(SPC)和环磷酰胺(CP)以及环孢素A(CsA)治疗移植受体,在移植术后的不同时间,采集移植心脏,对移植心脏... 目的探讨血小板转化生长因子(PDGF)与移植心脏排斥反应之间的关系。方法建立大鼠颈部心脏移植的动物模型。采用供体脾细胞(SPC)和环磷酰胺(CP)以及环孢素A(CsA)治疗移植受体,在移植术后的不同时间,采集移植心脏,对移植心脏血管病和心肌纤维化以及PDGF的表达水平等进行分析。结果急性排斥反应和CsA治疗的移植组中,冠状动脉的外膜和中层有大量胶原纤维浸润,并有内膜的水肿、血管壁增厚和血管腔狭窄,PDGF在冠状血管和心脏组织中呈高水平表达。SPC和cP预处理后,移植心脏的存活时间明显延长,PDGF在冠状血管和心肌组织中呈低水平表达,移植心脏血管病和心肌纤维化明显减轻。结论PDGF的表达水平与移植心脏血管病和心肌纤维化的严重程度密切相关,测定移植受体的PDGF水平可以预测移植器官的功能状况,为移植排斥反应的早期诊断和预防提供依据。 展开更多

目的探讨雾霾颗粒物对支气管哮喘缓解期儿童外周血和痰液中血小板衍生生长因子(PDGF)、转化生长因子-β1(TGF-β1)、c-fos mRNA表达的影响。方法选取来自重度雾霾污染、轻度雾霾污染和没有雾霾污染3个地区的支气管哮喘缓解期的儿童为样... 目的探讨雾霾颗粒物对支气管哮喘缓解期儿童外周血和痰液中血小板衍生生长因子(PDGF)、转化生长因子-β1(TGF-β1)、c-fos mRNA表达的影响。方法选取来自重度雾霾污染、轻度雾霾污染和没有雾霾污染3个地区的支气管哮喘缓解期的儿童为样本,并将它们分为3组。检测3组患儿PDGF、TGF-β1、c-fos mRNA的相对表达水平。同时,比较受到污染时间不同的患儿这几种RNA的相对表达水平。结果重雾霾污染组、轻雾霾污染组和无雾霾污染组3组支气管哮喘缓解期患儿的外周血PDGF、TGF-β1、c-fos mRNA的相对表达水平分别为(3.47±0.2)、(2.77±0.3)和(1.67±0.1),(3.71±0.4)、(2.91±0.4)和(1.91±0.2),(4.23±0.5)、(3.33±0.4)和(2.13±0.3),痰液中的PDGF、TGF-β1、c-fos mRNA的相对表达水平分别为(5.38±0.6)、(4.18±0.5)和(2.98±0.3),(5.62±0.4)、(4.42±0.4)和(3.12±0.3),(5.94±0.7)、(4.84±0.6)和(3.64±0.4),3组比较差异有统计学意义(均P<0.05)。受到雾霾颗粒物污染长达3年及以上患儿的外周血PDGF、TGF-β1、c-fos mRNA相对表达水平分别为(4.35±0.3)、(5.15±0.4)、(5.26±0.5),痰液中的PDGF、TGF-β1、c-fos mRNA相对表达水平分别为(5.24±0.4)、(5.78±0.3)、(5.87±0.4);而受到雾霾颗粒物污染3年以下的患儿外周血PDGF、TGF-β1、c-fos mRNA相对表达水平分别为(2.65±0.4)、(2.76±0.3)、(2.86±0.3),痰液中的PDGF、TGF-β1、c-fos mRNA相对表达水平分别为(4.32±0.3)、(4.18±0.2)、(3.75±0.2),两组比较差异有统计学意义(均P<0.05)。结论雾霾颗粒物显著增加支气管哮喘缓解期儿童外周血和痰液中的PDGF、TGF-β1、c-fos mRNA的表达水平;并且污染时间越长,表达水平越高。 展开更多

目的探讨蒙古山萝卜酸(MSA)对肝纤维化大鼠血清MMP-1、TIMP-1与肝脏中TGF-β_1、PDGF表达水平的影响。方法将大鼠分为对照组、模型组、阳性对照组(秋水仙碱1.5mg·kg-1)和实验药物高剂量组、低剂量组(MSA 240和30mg·kg-1),每... 目的探讨蒙古山萝卜酸(MSA)对肝纤维化大鼠血清MMP-1、TIMP-1与肝脏中TGF-β_1、PDGF表达水平的影响。方法将大鼠分为对照组、模型组、阳性对照组(秋水仙碱1.5mg·kg-1)和实验药物高剂量组、低剂量组(MSA 240和30mg·kg-1),每组10只,除对照组外,皮下注射含1mL·kg-1250mL·L^(-1)四氯化碳的花生油,每隔4d注射1次,共计30次。第4次注射后,阳性对照组和实验药物高、低剂量组开始ig给药,每日1次。实验结束后,麻醉,采血,分离血清,ELISA法测定血清MMP-1、TIMP-1,免疫组化法测定肝脏TGF-β_1、PDGF。结果对照组MMP-1、TIMP-1低活性表达,模型组MMP-1表达水平明显降低(与对照组比较P<0.05),TIMP-1表达水平显著升高(与对照组比较P<0.01);阳性药组MMP-1无明显改变(与模型组比较P>0.05),TIMP-1表达水平显著降低(与模型组比较P<0.05);实验药物高剂量组MMP-1表达水平明显上升(与模型组比较P<0.05),TIMP-1表达水平显著降低(与模型组比较P<0.05),MMP-1/TIMP-1比值提高;实验药物低剂量组对MMP-1、TIMP-1无明显变化(与模型组比较均P>0.05)。对照组TGF-β_1、PDGF表达低,模型组TGF-β_1、PDGF表达水平明显升高(与对照组比较均P<0.01);阳性药组TGF-β_1、PDGF表达水平显著降低(与模型组比较均P<0.05);实验药物高剂量组TGF-β_1、PDGF表达水平显著降低(与模型组比较分别P<0.01,P<0.05),实验药物低剂量组无明显作用(P>0.05)。结论蒙古山萝卜酸具有防治实验性大鼠肝损伤纤维化,与MMP-1、TIMP-1酶活性及TGF-β_1、PDGF表达水平失衡的调节作用有关。 展开更多

Effects of Salvia miltiorrhiza injectional powder on the development of liver fibrosis initiated by dimethylnitrosamine in rats 被引量:5

In this study, liver fibrosis rats were indu... Salvia miltiorrhiza (Sm) is a traditional herbal medicine with multiple effects on various diseases. Its water-soluble parts have been used to produce injectional powder. In this study, liver fibrosis rats were induced by intraperitoneal injection of dimethylnitrosarnine for 3 consecutive days per week for 4 weeks. After 2 weeks, rats in the positive drug group were subcutaneously injected with 8×10^5 IU/kg IFNα2b, while the Sm treatment groups were intraperitoneally injected with 50, 100 and 200 mg/kg solution of Sm injectional powder, respectively, for 6 days per week for 4 weeks. The results showed that either IFNα2b or the Sm injectional powder significantly increased the body weight and liver to spleen ratio, and three doses of the powder brought down the spleen index. Serum analysis showed that both IFNα2b and the Sm powder reduced levels of alanine transaminase and total bilirubin, while only 100 and 200 mg/kg of the Sm powder ameliorated aspartate transaminase and albumin levels. In the collagen examination, reduced hyaluronic acid and procollagen type III levels, less fibrous hyperplasia and collagen deposits, and improved hepatocyte states were clearly observed in rats treated with either IFNα2b or Sm injectional powder. In addition, the mechanism of action of the Sm powder was also studied. Immunohistochemical staining showed that IFNα2b and Sm injectional powder significantly down-regulated the expression of transforming growth factor-β1 (TGF-β1) and platelet derived growth factor (PDGF). In conclusion, Sm injectional powder has protective effects on dimethylnitrosamine-initiated liver fibrosis in rats, and the mechanism may include the down-regulation of TGF-β1 and PDGF. 展开更多

Objective To observe the efficacy of mild moxibustion combined with arthroscopic meniscal repair in the treatment of meniscal injury and to explore its action mechanism.Methods Ninety-eight patients with meniscal injury were divided into a surgery group and a moxibustion plus surgery group by the random number table method,with 49 cases in each group.Both groups received arthroscopic meniscal repair,and the moxibustion plus surgery group was treated with add-on mild moxibustion.The Lysholm score,visual analog scale(VAS)score,and mobility of the affected knee were measured before and after treatment,and transforming growth factor(TGF)-β1 and platelet-derived growth factor(PDGF)levels were measured in the fluid of the affected knee joint.The healing of the meniscus was recorded at a follow-up visit 3 months after the surgery.Results After treatment,the Lysholm score increased,the VAS score decreased in both groups,and the intra-group differences in both groups were statistically significant(P<0.05).The Lysholm score was higher in the moxibustion plus surgery group than in the surgery group,and the VAS score was lower in the moxibustion plus surgery group than in the surgery group.The differences between groups in both scores were statistically significant(P<0.05).The mobility of the affected knee joint increased in both groups(P<0.05),and it was greater in the moxibustion plus surgery group than in the surgery group(P<0.05).When compared with pretreatment,the levels of TGF-β1 and PDGF in the fluid of the knee joint increased in both groups(P<0.05),and the levels of TGF-β1 and PDGF in the fluid of the affected knee joint were higher in the moxibustion plus surgery group than in the surgery group(P<0.05).The healing of the meniscus in the moxibustion plus surgery group was significantly better than that in the surgery group at the follow-up visit 3 months after the surgery(P<0.05).Conclusion The effect of mild moxibustion combined with arthroscopic meniscal repair is better than the surgery alone in improving knee function,relieving pain,increasing mobility of the affected knee,and promoting meniscal healing,which may be related to the up-regulation of TGF-β1 and PDGF levels in the fluid of knee joint. 展开更多
