根据K5蛋白(Pro451—Ala541)的结构特征和二硫键分布特点,设计K5的两个缺失突变体K5 mut1(Cys461—Cys540,保留K5 kringle环3个完整二硫键但去除N端和C端多余氨基酸)和K5mut2(Cys482—Cys535,打开kringle环,只保留2个二硫键).以野生型... 根据K5蛋白(Pro451—Ala541)的结构特征和二硫键分布特点,设计K5的两个缺失突变体K5 mut1(Cys461—Cys540,保留K5 kringle环3个完整二硫键但去除N端和C端多余氨基酸)和K5mut2(Cys482—Cys535,打开kringle环,只保留2个二硫键).以野生型人纤溶酶原K5 cDNA为模板,用PCR方法得到编码缺失突变体的DNA片段,定向克隆入pET22b(+)质粒载体,重组体转化进大肠杆菌BL21(DE3),诱导表达,产物经亲和层析和高浓度甘油透析纯化后进行鉴定和生物活性测定.K5 mut1蛋白特异性抑制人视网膜微血管内皮细胞增殖,且活性强度是完整的K5蛋白2倍;K5mut2对人视网膜微血管内皮细胞无显著抑制作用.结果提示,完整的Kringle结构(包含3个二硫键)是维持人纤溶酶原K5抗血管增生活性的必需结构域,而K5分子中Kringle结构域外的N端和C端氨基酸臂则并非其活性所必需. 展开更多

扶阳化阴方加减治疗原发性肝癌临床研究

目的:观察扶阳化阴方加减治疗原发性肝癌(HCC)的临床疗效。方法:采用掷币法将91例HCC患者分为对照组(45例)与观察组(46例)。对照组给予肝动脉化疗栓塞术(TACE)治疗,观察组在对照组基础上加用扶阳化阴方加减治疗。比较2组治疗前后肝功能... 目的:观察扶阳化阴方加减治疗原发性肝癌(HCC)的临床疗效。方法:采用掷币法将91例HCC患者分为对照组(45例)与观察组(46例)。对照组给予肝动脉化疗栓塞术(TACE)治疗,观察组在对照组基础上加用扶阳化阴方加减治疗。比较2组治疗前后肝功能指标[谷丙转氨酶(ALT)、谷草转氨酶(AST)、总胆红素(TB)]、血管增生因子指标[血管内皮生长因子(VEGF)、转化生长因子-β1(TGF-β1)]、肿瘤标志物指标[甲胎蛋白(AFP)、甲胎蛋白异质体(AFP-L3)比率、高尔基体蛋白73(GP73)]及中医证候积分,并评估2组临床疗效及不良反应发生情况。结果:治疗后,观察组总缓解率80.43%,高于对照组60.00%(P<0.05)。治疗后,2组GP73、AFP及AFP-L3比率均较治疗前降低(P<0.05),且观察组治疗后GP73、AFP水平及AFP-L3比率低于对照组(P<0.05)。治疗后,2组AST、ALT及TB均较治疗前降低(P<0.05),且观察组治疗后肝功能指标低于对照组(P<0.05)。治疗后,2组VEGF、TGF-β1及中医证候积分均较治疗前明显降低(P<0.05),且观察组治疗后血管增生因子指标及中医证候积分均低于对照组(P<0.05)。2组不良反应发生率比较,差异无统计学意义(P>0.05)。结论:扶阳化阴方加减治疗HCC效果显著,能有效改善患者肝功能,降低血管增生因子及肿瘤标志物水平,安全性较高。 展开更多

背景目前,烟雾病(MMD)的病因及发病机制尚不完全明确,此类患者通常在发生脑血管事件后才被确诊,因此探讨MMD的预测因子及评估其严重程度的指标具有重要的临床意义。目的探讨成年MMD患者血清血管细胞黏附分子1(VCAM-1)、基质金属蛋白酶9(... 背景目前,烟雾病(MMD)的病因及发病机制尚不完全明确,此类患者通常在发生脑血管事件后才被确诊,因此探讨MMD的预测因子及评估其严重程度的指标具有重要的临床意义。目的探讨成年MMD患者血清血管细胞黏附分子1(VCAM-1)、基质金属蛋白酶9(MMP-9)、转化生长因子β(TGF-β)、血管内皮生长因子(VEGF)水平及其临床意义。方法选取2016年3月-2019年1月四川大学华西广安医院收治的成年MMD患者114例作为病例组,其中缺血型68例,出血型46例;另选取同期在本院门诊健康体检中心体检健康者30例作为对照组。比较对照组受试者体检当日与病例组患者入院第1天血清VCAM-1、MMP-9、TGF-β、VEGF水平,比较出血型与缺血型患者入院第1、7、14天血清VCAM-1、MMP-9、TGF-β、VEGF水平,并比较不同Suzuki分级成年MMD患者入院第1天血清VCAM-1、MMP-9、TGF-β、VEGF水平;成年MMD患者入院第1天血清VCAM-1、MMP-9水平与血清TGF-β、VEGF水平的相关性分析采用Pearson相关分析。结果病例组患者入院第1天血清VCAM-1、MMP-9、TGF-β、VEGF水平高于对照组(体检当日)(P<0.05)。时间与方法在血清VCAM-1、MMP-9水平上不存在交互作用(P>0.05),在血清TGF-β、VEGF水平上存在交互作用(P<0.05);时间在血清VCAM-1、MMP-9、TGF-β、VEGF水平上主效应显著(P<0.05);方法在血清VCAM-1、MMP-9水平上主效应不显著(P>0.05),在血清TGF-β、VEGF水平上主效应显著(P<0.05);出血型患者入院第1、7、14天血清TGF-β、VEGF水平高于缺血型患者(P<0.05)。Suzuki分级3级缺血型患者血清入院第1天MMP-9、VEGF水平高于Suzuki分级2级患者,Suzuki分级3级出血型患者入院第1天血清VEGF水平高于Suzuki分级2级患者(P<0.05);Suzuki分级4级缺血型、出血型患者入院第1天血清VCAM-1、MMP-9、TGF-β、VEGF水平分别高于Suzuki分级2、3级缺血型、出血型患者(P<0.05);Suzuki分级5、6级缺血型、出血型患者入院第1天血清VCAM-1、MMP-9、TGF-β、VEGF水平分别高于Suzuki分级2、3、4级缺血型、出血型患者(P<0.05);Suzuki分级6级缺血型、出血型患者入院第1天血清VEGF水平分别高于Suzuki分级5级缺血型、出血型患者(P<0.05)。Pearson相关分析结果显示,缺血型、出血型MMD患者入院第1天血清VCAM-1、MMP-9水平分别与血清TGF-β、VEGF水平呈正相关(P<0.05)。结论缺血型和出血型成年MMD患者血清VCAM-1、MMP-9、TGF-β、VEGF水平均异常升高,且均与患者病情严重程度相关;血清VCAM-1、MMP-9水平分别与缺血型、出血型MMD患者血清TGF-β、VEGF水平呈正相关,VCAM-1、MMP-9可能参与了成年MMD患者颅内出血过程。 展开更多

AIM: To investigate the implication of the hypoxia inducible factor HIF-1α mRNA in gastric carcinoma and its relation to the expression of vascular endothelial growth factor (VEGF) protein, tumor angiogenesis invasion/metastasis and the patient's survival. METHODS: In situ hybridization was used to examine expression of HIF-1α mRNA, and immunohistochemical staining was used to examine expression of VEGF protein and CD34 in 118 specimens from patients with gastric carcinoma. RESULTS: The positive rates of HIF-1α mRNA and VEGF protein were 49.15% and 55.92%, respectively. Positive expressions of HIF-1α and VEGF in stage T3-T4 tumors and those with vessel invasion, lymph node metastasis and distant metastasis were dramatically stronger than stage T1-T2 cases and those without vessel invasion, lymph node metastasis and distant metastasis. The mean microvascular density (MVD) in stage T3-T4 tumors and those with vessel invasion, lymph node metastasis and distant metastasis was significantly higher than stage T1-T2 tumors and those without vessel invasion, lymph node metastasis and distant metastasis. The mean MVD in tumors with positive HIF-1α and VEGF expression was significantly higher than that in tumors with negative HIF-1α and VEGF expression. The expression of HIF- 1α was positively correlated with VEGF protein. There were positive correlations between MVD and expression of HIF-1α and VEGF. The mean survival time and the S-year survival rate in cases with positive expression HIF-1α and VEGF and MVD value ≥ 41.5/0.72 mm^2 were significantly lower than those with negative expression of HIF-1α and VEGF and MVD value 〈 41.5/0.72 mm^2. CONCLUSION: Overexpression of HIF-1α is found in gastric carcinoma. HIF-1α may induce the angiogenesis in gastric carcinoma by upregulating the transcription of VEGF gene, and take part in tumor invasion and metastasis. They can be used as prognostic markers of gastric cancer in clinical practice. 展开更多

The predictive value of vascular endothelial growth factor and Ki-67 expression on neoadjuvant therapy in rectal cancer 被引量:1

Objective: To investigate the expressions of vascular endothelial growth factor (VEGF) and proliferating cell nuclear antigen (Ki-67) in patients with rectal adenocarcinoma and their associations with neoadjuvant therapy. Methods: The expressions of Ki-67 and VEGF in 32 cases of rectal adenocarcinoma, including both pretreatment tumor biopsies and postoperative specimen, were detected by immunohistochemistry using specific antibodies, and were correlated with clinicopathological factors. Results: The intensity of VEGF staining was significantly correlated with lymph nodal metastasis (P =0.033), depth of tumor invasion (P =0.007) and tumor stage (P= 0.016), but not with histological types, tumor sizes, patients' ages and genders (P 〉 0.05). Low level of VEGF expression had significant correlation with the high sensitivity of response to neoadjuvant therapy (P = 0.016). The transient increase of VEGF expression could be seen after neoadjuvant therapy (P = 0.035). Ki-67 labeling index (Ki-67-LI) was significantly correlated with lymph node metastasis (P = 0.028), but not correlated to tumor sizes, patients' ages and genders (P 〉 0.05). Tumors with lower Ki-67-LI were more sensitive to neoadjuvant therapy (P = 0.032). The Ki-67 level decreased after neoadjuvant therapy, but no statistical significance was found (P 〉 0.05). Conclusion: Our results demonstrate that the expression of VEGF and Ki-67 in pretreatment rectal adenocarcinoma biopsies may be predictive of tumor response to neoadjuvant therapy. 展开更多

AIM: To determine whether insulin could promote sinusoidal endothelial cell (SEC) proliferation mediated by upregulation of vascular endothelial growth factor (VEGF) in regenerating rat liver after partial hepatectomy (PHx). METHODS: Adult male Sprague-Dawley rats undergoing 70% PHx were injected with insulin (300 MU/kg) or saline via the tail veins every 8 h after surgery for 7 d and killed at 0, 24, 48, 72, 96, 120, 144, and 168 h after surgery. Proliferation of both hepatocytes and SECs was monitored by evaluating the proliferating cell nuclear antigen (PCNA) labeling index (LI). The expression of VEGF protein was evaluated by immunohistochemistJv. The mRNA expressions of VEGF and its receptors FIt-1 and FIk-1 were evaluated by semi-quantitative reverse transcription-PCR. RESULTS: Insulin markedly increased the expression of VEGF mRNA between 24 and 120 h after hepatectomy compared to controls. Similarly, insulin significantly increased the expression of Fit-1 between 24 and 96 h. However, insulin had no significant effect on FIk-1. Furthermore, the immunohistochemical staining revealed that expression of VEGF protein increased in the insulin groups. Insulin significantly increased the PCNA LI of hepatocytes and SECs compared to controls. CONCLUSION: Exogenous insulin may promote SEC proliferation with an enhanced expression of VEGF and its receptor Fit-1 in regenerating rat liver after PHx. 展开更多

Methods: Cultured lung A549 ceils were randomly divided into 6 groups: normal control group (NC), the Jinshuixian group (JSX), radiotherapy (RT), JSX for the first day and the next day followed by RT group (JSX~ RT), RT for the first day and the next day followed by JSX group (RT→JSX) and RT + JSX concomitantly group (JSX + RT). MTT was applied to measure the cell viability, RT-PCR and ELISA were used to test the expression of mRNA and protein of VEGF. Results: The proliferation of A549 cells were inhibited in JSX and combined groups and the inhibiting effects were time dependent. The expression of VEGF in RT group was increased, however, VEGF in JSX and combination groups were largely decreased over time when compared to NC group. Results in JSX→RT, RT→JSX and JSX + RT groups did not achieve significantly differences. Conclusion: JSX has the ability of anti-tumor growth in vitro accompanied down-regulating of VEGF, especially when combined with radiotherapy, and its effect is time-dependent. However, more studies in vivo and in vitro are needed to further supporting these effects. 展开更多

Evaluation of the tumor angiogenesis in benign prostate hyperplasia and prostatic cancer with MR perfusion-weighted imaging

Objective: To explore the application of MR perfusion-weighted imaging (PWI) in the benign and malignant prostate diseases, and evaluate the correlations of PWl features with vascular endothelial growth factor (VEGF) and microvessel density (MVD). Methods: Seventy-four consecutive patients who were diagnosed clinically for the prostate diseases, including forty-four cases with benign prostate hyperplasia and thirty cases with prostatic cancer proved pathologically, were examined by PWI. MVD and VEGF were stained with immunohistochemical methods. Some parameters of PWl, including the steepest slope of signal intensity-time curve (SSmax) and the change in relaxation rate (ΔR2^* peak) at lesions, were analyzed. Correlation analysis was used to determine the relationship between the results of PWl and immunohistochemistry. Results: (1) In the benign prostate hyperplasia (BPH), SSmax and ΔR2^* peak of perfusion curve were 34.2 ± 2.9 and 1.49 ± 0.11, respectively; however, in the prostatic cancer (Pca), they were 58.6± 4.8 and 3.18 ±0.49 respectively; there were statistical differences (t = 2.16 and 2.31, P 〈 0.05). (2) The VEGF and MVD expressions of thirty Pca patients were significantly higher than those of forty-four BPH patients (x2 = 28.64, P 〈 0.01; t = 21.2, P 〈 0.01). MVD expressions of Pca and BPH groups showed positive associations with VEGF expressions (P 〈 0.01). On MR perfusion-weighted imaging, SSmax and ΔR2^* peak showed associations with MVD and VEGF expressions (P 〈 0.01). Conclusion: On MR perfusion-weighted imaging, SSmax and ΔR2^* peak can reflect MVD and VEGF expression levels in the benign and malignant prostate diseases and might be implied the tumor angiogenesis so as to distinguish benign from malignant and provide the important information for the surgeon to diagnose and treat the prostatic diseases. 展开更多

In this research, we explored the effect of DADAG on angiogenesis in chick chorioallantoic membrane (CAM) model and on the proliferation and migration of human umbilical vein endothelial cells (HUVECs). We also studied the possible mechanism of the anti-angiogenesis effect of DADAG. The results showed that DADAG (100, 500 and 1000μnol/L) inhibited angiogenesis in CAM model dose-dependently. Sulforhodamine B (SRB) assay indicated that DADAG (45, 90, 135, 180 and 225 μmol/L) suppressed HUVECs proliferation in a dose-dependent and time-dependent manner. High Content Screening (HCS, Cellomics) assay, in which the influence of cell proliferation on migration could be excluded, indicated that DADAG (45, 135 and 225 ~xmol/L) directly inhibited the motility ofHUVECs. Immunofiuorescence assay suggested that DADAG inhibited angiogenesis possibly by decreasing vascular endothelial growth factor (VEGF) expression in HUVECs.
