补肾降浊饮及其拆方改善非酒精性脂肪肝大鼠胰岛素抵抗的机制研究

目的:研究补肾降浊饮及其拆方对非酒精性脂肪性肝炎(Nonalcoholic Steatohepatitis,NASH)大鼠胰岛素抵抗(Insulin Resistance,IR)的影响,探讨补肾降浊饮及其拆方对胰岛素信号通路相关细胞因子及激酶的影响。方法:将60只SD大鼠随机分为... 目的:研究补肾降浊饮及其拆方对非酒精性脂肪性肝炎(Nonalcoholic Steatohepatitis,NASH)大鼠胰岛素抵抗(Insulin Resistance,IR)的影响,探讨补肾降浊饮及其拆方对胰岛素信号通路相关细胞因子及激酶的影响。方法:将60只SD大鼠随机分为正常组、模型组、补肾组、降浊组、补肾降浊饮组及西药组,正常组予普通饲料喂养,其余各组予高脂饲料喂养并分别灌胃生理盐水或相应剂量药物干预。12周后检测对比各组大鼠血清胆固醇(Total Cholesterol,TC)、三酰甘油(Triglyceride,TG)、丙氨酸氨基转移酶(Alanine Aminotransferase,ALT)、天门冬氨酸氨基转移酶(Aspartate Aminotransferase,AST)、空腹血糖(Glucose,GLU)、胰岛素(Insulin,INS)水平、胰岛素抵抗指数(Insulin Resistance Index,IRI)、胰岛素敏感指数(Insulin Sensitivity Index,ISI)水平及血清脂联素(Adiponectin,ADP)、瘦素(Leptin,LEP)、肿瘤坏死因子(Tumor Necrosis Factor,TNF)-α、白细胞介素(Interleukin,IL)-6水平;油红O染色检测肝组织脂肪变性程度,糖原染色检测肝组织糖原含量;Western-blot法检测肝组织中c-Jun氨基端激酶1(c-Jun N-terminal Kinase 1,JNK1)、磷酸化c-Jun氨基末端激酶(Phospho-c-Jun N-terminal Kinase,p-JNK)及胰岛素受体α(Insulin Receptor alpha,IRα)、磷酸化胰岛素受体底物1(Phospho-insulin Receptor Substrate-1,p-IRS-1)表达水平。结果:中药各组均可不同程度改善大鼠血清中TC、TG、ALT、AST及GLU、INS、IRI、ISI水平(P<0.05,P<0.01),亦能不同程度影响血清ADP、LEP、TNF-α、IL-6含量及肝组织JNK1、p-JNK、IRα及p-IRS-1表达水平(P<0.05,P<0.01)。中药各组在改善GLU、INS、IRI、ISI及p-IRS-1方面均优于西药组(P<0.05,P<0.01);其中补肾组在改善TC、TG、ADP、LEP方面优于西药组(P<0.05,P<0.01);降浊组在改善ALT、AST、TNF-α、IL-6方面优于西药组(P<0.05,P<0.01);补肾降浊饮组在各方面均优于西药组(P<0.01)。结论:补肾降浊饮可明显改善NASH大鼠IR,其中补肾组分别通过调节脂肪细胞因子ADP、LEP水平,降浊组分别通过调整炎性因子TNF-α、IL-6、JNK1、p-JNK水平,共同增强胰岛素受体底物活性,进而改善胰岛素信号转导。 展开更多

OBJECTIVE:To observe the effects of Bushen Huoxue Yin(补肾活血饮,BSHXY) on nuclear transcription factor kappa B(NF-κB) and nitric oxide(NO) in the brain of the Parkinson's disease(PD) model mouse.METHODS:Forty-five C57BL/6 mice were randomly divided into three groups;normal,model and BSHXY treatment groups.Concentrations of NF-κB and NO in mouse brain tissue were determined by ELISA and spectrophotometry,respectively.RESULTS:NF-κB concentration in brain tissue in the model group was 14.04±4.38 μg· L-1,which was higher than that in normal(P<0.01) and BSHXY(P< 0.05) groups.NO content in brain tissue in the model group was 5.93±0.79 μmol · gprot-1,which was also higher than that in model(P<0.01) and BSHXY(P<0.01) groups.However,there were no significant differences in the content of NF-κB and NO between BSHXY and normal groups(P>0.05).CONCLUSION:The mechanism of BSHXY for treatment of PD is possibly related to inhibition ofNF-κB activation and decreased NO content in the brain. 展开更多

对糖尿病肾病患者采用补气化瘀固肾方联合ACEI进行治疗,并对患者的糖化血红蛋白与炎症因子含量进行分析。方法:设置研究起止时间为2019年8月至2020年8月,研究所选80名参研对象均为我院收治的糖尿病肾病患者,将上述参研对象依照计算机表... 对糖尿病肾病患者采用补气化瘀固肾方联合ACEI进行治疗,并对患者的糖化血红蛋白与炎症因子含量进行分析。方法:设置研究起止时间为2019年8月至2020年8月,研究所选80名参研对象均为我院收治的糖尿病肾病患者,将上述参研对象依照计算机表法分为A、B两组,两组人数均等(40人),分别给予两组患者常规治疗与补气化瘀固肾方联合ACEI治疗。对两组患者的临床指标(血清超敏C反应蛋白(hs-CRP)、糖化血红蛋白(HbAlc)、收缩压和舒张压)、炎症因子变化情况(尿蛋白(Pro)、白细胞介素-6(IL-6)和血肌酐(SCR))和临床总疗效进行记录,并对最终数据进行分析与对比。结果:经比较发现,B组患者的临床指标与炎症因子恢复情况显著优于A组,组间差异明显(P<0.05);同时B组的临床疗效整体高于A组,数据差异较大(P<0.05)。讨论:补气化瘀固肾方联合ACEI治疗糖尿病肾病效果显著,可缓解患者的炎症反应,有利于其血压等临床指标恢复,应进一步推广与应用。 展开更多

OBJECTIVE: To observe the effect of "Yang-warming and kidney essence-replenishing" herbal paste on cold-related asthma exacerbation. METHODS: One hundred and fifty one patients with moderate to severe persistent asthma were randomly divided into a treatment group (n=74) and a control group (n=69). Both groups were given basic treatment according to the Global Initiative for Asthma scheme. The treatment group was also treated with herbal paste in the winter. The frequency of asthma exacerbation, cold and cold-re-lated asthma exacerbation, scores of the asthma control test (ACT), and kidney-deficiency syndrome during the one-year follow-up in each group were recorded. RESULTS: Compared with the control group, the onset frequencies of catching cold, asthma exacerbation, cold-related asthma exacerbation, and kidney-deficiency syndrome score were significantly reduced. There was no significant difference in scores of ACT between the two groups. CONCLUSION: "Yang-warming and kidney essence-replenishing" herbal paste could reduce cold-related asthma exacerbation. 展开更多

OBJECTIVE: To investigate the effect of Bushenyisui Formula on cell apoptosis and positive B cell lym- phoma (Bcl-2) in the Brain of rat models of Alzheim- er's disease (AD) induced by beta-amyloid protein (All3) and the mechanism underlying the effect. METHODS: Total of 40 SD rats, 20 females and 20 males, were randomly assigned to 4 groups, con- trolled group (A), model group (B), conventional treatment group (C) and Bushenyisui Formula treat- ment (BYFT) group (D), 10 rats in each group. AI3 1-42 was injected into the bilateral hippocampus of the rats in group B, C and D to create the models of AD. Sham operation was performed on the rats of group A in the same way by injecting equal volume of 0.9% sodium chloride solution into their bilateral hippocampus. 5 days after operation, Bushenyisui Formula was intraperitoneally administered at a dose of 450 mg/kg to the rats of group D (QD) for 20 days. Equal volume of 0.9% sodium chloride solution was intraperitoneally injected into the rats of group B with the same procedure. C suspension (20 mg/mL) was intraperitoneally injected into the rats of group B with the same procedure. The number of apoptot- ic cells in Brain and the positive Bcl-2 were count- ed. The changes of learning and memory abilities were evaluated using Y-maze. RESULTS: Right after the establishment of the mod- els, group B, C and D compared to group A respec- tively, the outcomes of Y-maze were significantly different from that of group A, which suggested ob- vious learning and memory disorder in those groups (P〈0.01). After treatment, the times of elec- tronic shocks of group C and D were significantly less than that of group B (P〈0.05), and the numbers of apoptotic cells and positive Bcl-2 were signifi- cantly different from those of group B, apoptotic sells' number of group C and D smaller than that of group B and the number of positive Bcl-2 greater than that of group B. CONCLUSION: Bushenyisui Formula could increase the number of Bcl-2 in brain, which improved the function of nervous system pertaining to learning and memory abilities. 展开更多
