In order to evaluate the properties of the tungsten alloys under transient high heat flues, four tungsten samples with different grain sizes were tested by high-intensity pulsed ion beam with a heat flux as high as 160 MW/(m^2·s^-1/2). Compared with the commercial tungsten, the surface modification of the oxide dispersion strengthened tungsten by high-intensity pulsed ion beam is completely different. The oxide dispersion strengthened tungsten shows inferior thermal shock response due to the low melting point second phase of Ti and Y2O3, which results in the surface melting, boiling bubbles and cracking. While the carbide dispersion strengthened tungsten shows better thermal shock response than the commercial tungsten.展开更多

Lectin and leghemoglobin in legumes play the important roles, respectively, in recognition of host plants to their rhizobial bacteria, and lowering the oxygen partial pressure around bacteroids and protecting nitrogen...Lectin and leghemoglobin in legumes play the important roles, respectively, in recognition of host plants to their rhizobial bacteria, and lowering the oxygen partial pressure around bacteroids and protecting nitrogenase from oxygen in symbiotic nitrogen-fixing nodules. In order to extend the host range of the rhizobial bacteria and to make them fix nitrogen in non-legumes, pea lectin gene (pl) and Parasponia hemoglobin gene ( phl,) have been constructed into a plant expression vector (pCBHUL) and the vector pCBHUL was introduced into rice calli from immature young embryos by particle bombardment. After the calli were regenerated into plantlets on the resistant-selecting media containing hygromycin, they were identified by PCR and Southern blot hybridization. It was indicated that the pi and phb genes were integrated into nucleic genome of the transformed rice plants. GUS activity and the product of the pi gene were determined by GUS staining, Western blot and in situ hybridization at translational level. Eighteen out of 40 plants resistant to hygromycin were positively identified by PCR analysis with the rate of 45%. The pi gene was expressed in 3 out of 18 plants with 17% and 7.5% in 40 plants. The results may provide a clue for exploring whether Rhizobium leguminosarum by. viceae could extend its host range and make the transgenic rice plants have the possibility of being symbiotic, or associative to nitrogen fixation.展开更多

The general procedure in designing a grand style timber structure is to start with the grade that defines the basic module (dou kou); next comes with the number of bracket set (cuan), the number of longitudinal bays and the number of purlins which affect its plan and cross section; thirdly choose a roof type that determines its longitudinal section and the facade as well. The complete open reading frame and the fragment encoding the putative mature protein were inserted into the prokaryotic expression vector pET_32b(+), respectively. Subsequent expression showed that the Rs_AFP1 was produced in E. coli as a 27 kD fusion protein only when the N_terminal signal peptide was removed. After treatment with thrombin to remove part of the N_terminal His.tag sequence, the bacterially expressed Rs_AFP1 was used for fungal growth inhibition assay which was conducted on Verticillium dahliae Kleb., a soil_born fungus causing the cotton wilt disease. Results showed that, in the liquid medium, the Rs_AFP1 fusion protein at a concentration of 0.3 g/L clearly inhibited the growth of V. dahliae and the germination of spores. Thus the bacterially expressed fusion protein had the antifungal activity against V. dahliae.展开更多

Methods: Expression of PPAR-γ in 15 cases of non-cancerous lung tissuesand 64 cases of lung cancer tissues was detected by using immunohistochemistry. The average A valueswere measured by using image analysis. The expression of PPAR-γ in lung cancer cells was detectedby using RT-PCR. After being treated with PPAR-γ activators, apoptosis was detected by using flowcytometry, and meanwhile, the change in caspase-3 activity was detected by caspase-3 kits. Results:Expression levels of PPAR-γ in cancerous tissues were higher than those in non-cancerous lungtissues. In four types of lung cancer, the sequence of PPAR-γ expression from high to low levelswas small-cell lung cancer, squamous carcinom, large-cell lung cancer, adenocarcinoma in turns. Theexpression of PPAR-γ was correlated with differentiation and postoperative TNM staging of lungcancer tissues, but not with lymph node metastasis. PPAR-γ expressed in two lung cancer cell lines,which could induce apoptosis of lung cancer cells after being treated with PPAR-γ activators, andcaspase-3 activity in cells treated with PPAR-γ activators was significantly increased. Conclusion:PPAR-γ is correlated with clinical pathology and apoptosis of lung cancer closely, and activatedPPAR-γ can increase caspase-3 activity to induce cells apoptosis. Two reducing agents, trisodium citrate dehydrate (TSC) and potassium tartrate, were used to reduce the AgNO3 solution. In this redox system, polyvinylpyrrolidone (PVP), sodium dodecyl sulfate (SDS) and arabic gum were served as surfactants. The results showed that reducing agents and surfactants both act as the capping agent adhering to the certain facets of silver seeds to block this surface to grow. The relative intensity of reducing agents also takes an active part in influencing the growth rate and direction of silver seeds. It was also found that halides can accelerate the speed of Ostwald ripening by adding Cl?, Br? and I?into the aqueous and have some effects on the morphology of the nanoplates.展开更多

METHODS: SAP model of r...AIM: To investigate the effect of IL-4 on the altered expression of complement activation regulators in pancreas and pancreatic necrosis during experimental severe acute pancreatitis (SAP). METHODS: SAP model of rats was established by retrograde injection of 5% sodium taurocholate (1 mL/kg) into the pancreatic duct. We immunohistochemically assayed the expression of three complement activation regulators: decay accelerating factor (DAF; CD55), 20 ku homologous restriction factor (HRF20; CD59) and membrane cofactor protein (MCP; CEH6), in the pancreatic acinar cells of rats at 0, 3, 6, 12, and 24 h after the induction of SAP model. Meanwhile the levels of amylase and lipase were determined, and morphological examination was performed. Then, 61 rats were randomly divided into three groups. Group A (n = 21) received no treatment after the SAP model was established; group B (n = 20) was given IL-4 (8 IJg/animal) intraperitoneally 0.5 h before the SAP model was established; group C (n = 20) was given IL-4 (8 μg/animal) intraperitoneaUy 0.5 h after the SAP model was established. Plasma amylase and lipase, extent of pancreatic necrosis and expression of complement activation regulators were investigated 6 h after the induction of SAP model. RESULTS: Three complement activation regulators were all expressed in pancreatic acinar cells. MCP was not found on the basolateral surface as reported. Contrary to the gradually increasing plasma level of amylase and lipase, expression of complement activation regulators decreased after SAP model was set up. At the same time, the severity of pancreatic necrosis was enhanced. A strong negative correlation was found between the expression of MCP, DAF, CD59 in pancreatic acinar cells and the severity of pancreatic necrosis (r = -0.748, -0.827, -0.723; P〈0.01). In the second series of experiments, no matter when the treatment of IL-4 was given (before or after the induction of SAP model), the serum level of amylase or lipase was decreased and the extent of pancreatic necrosis was ameliorated significantly. Compared to SAP control group, the expression of DAF and CD59 in pancreas was reinforced when IL-4 was given before the induction of SAP model (P〈0.01, P〈0.05), but the expression of MCP was not influenced (P〉0.05). The expression of DAF was enhanced, when IL-4 was given after the induction of SAP model (P〈0.05), but the expression of CD59 and MCP did not change (P〉0.05). CONCLUSION: Complement activation regulators may participate in the pathogenesis of pancreatic inflammation. Downregulation of complement activation regulators expression may be one of the causes of pancreatic necrosis. IL-4 treatment may control SAP aggravation by enhancing expression of DAF and CD59 in pancreas and decreasing pancreatic necrosis. Moreover, DAF and CD59 may play an important role in the regulation of complement activation regulators during SAP.展开更多

METHODS: Three siRNA-expressing vectors targeting different sites of the Bcl-XL gene were constructed from pTZ-U6+I vector. Cultured esophageal cancer cells were transfected with the siRNA-expressing vector (or the control vector) using lipofectamine 2000. BcI-XL gene expression was determined with semiquantitative RT- PCR assay and Western blotting. Among the three siRNA- expressing vectors, the most highly functional vector and its effect on cell growth and apoptosis in esophageal cancer ceils was further analyzed. RESULTS: Of the three siRNA-expressing vectors, siRNA- expressing vector No.1 was the most potent one which suppressed Bcl-XL mRNA production to 32.5% of that in the untreated esophageal cancer cells. Western blotting analysis showed that siRNA-expressing vector No.1 markedly down-regulated the expression of Bcl-XL in human esophageal cancer cells. Treatment of esophageal cancer cells with siRNA-expressing vector No.1 resulted in inhibition of cell growth and induction of apoptosis. CONCLUSION: Down-regulation of BcI-XL by vectorgenerated small interfering RNAs can suppress cell growth and induce apoptosis in human esophageal cancer cells.展开更多

This means that, with respect to the on\|line problem, some further information about the tasks is available, which allows the improvement of the performance of the best possible algorithms. Problems of this class are called semi on\|line ones. The authors studied two semi on\|line multiprocessor scheduling problems, in which, the total processing time of all tasks is known in advance, or all processing times lie in a given interval. They proposed approximation algorithms for minimizing the makespan and analyzed their performance guarantee. While research on pedogenesis mainly focuses on long-term soil formation and most often neglects recent soil evolution in response to human practices or climate changes, this article reviews the impact of artificial s...While research on pedogenesis mainly focuses on long-term soil formation and most often neglects recent soil evolution in response to human practices or climate changes, this article reviews the impact of artificial subsurface drainage on soil evolution. Artificial drainage is considered as an example of the impact of recent changes in water fluxes on soil evolution over time scales of decades to a century. Results from various classical studies on artificial drainage including hydrological and environmental studies are reviewed and collated with rare studies dealing explicitly with soil morphology changes, in response to artificial drainage. We deduce that soil should react to the perturbations associated with subsurface drainage over time scales that do not exceeding a few decades. Subsurface drainage decreases the intensity of erosion and must i) increase the intensity of the lixiviation and eluviation processes, ii) affect iron and manganese dynamics, and iii) induce heterogeneities in soil evolution at the ten meter scale. Such recent soil evolutions can no longer be neglected as they are mostly irreversible and will probably have unknown, but expectable, feedbacks on crucial soil functions such as the sequestration of soil organic matter or the water available capacity.展开更多

A strong negative correlation was found between the expression of MCP, DAF, CD59 in pancreatic acinar cells and the severity of pancreatic necrosis (r = -0.748, -0.827, -0.723; P〈0.01). In the second series of experiments, no matter when the treatment of IL-4 was given (before or after the induction of SAP model), the serum level of amylase or lipase was decreased and the extent of pancreatic necrosis was ameliorated significantly. Compared to SAP control group, the expression of DAF and CD59 in pancreas was reinforced when IL-4 was given before the induction of SAP model (P〈0.01, P〈0.05), but the expression of MCP was not influenced (P〉0.05). The expression of DAF was enhanced, when IL-4 was given after the induction of SAP model (P〈0.05), but the expression of CD59 and MCP did not change (P〉0.05). CONCLUSION: Complement activation regulators may participate in the pathogenesis of pancreatic inflammation. Downregulation of complement activation regulators expression may be one of the causes of pancreatic necrosis. IL-4 treatment may control SAP aggravation by enhancing expression of DAF and CD59 in pancreas and decreasing pancreatic necrosis. Moreover, DAF and CD59 may play an important role in the regulation of complement activation regulators during SAP.

文摘AIM: To determine the inhibitory effect of the vectorgenerated small interfering RNAs (siRNAs) on the expression of the BcI-XL gene in established human esophageal cancer cells, and to investigate the effect of the BcI-XL siRNAs on cell growth and apoptosis in esophageal cancer cells. METHODS: Three siRNA-expressing vectors targeting different sites of the Bcl-XL gene were constructed from pTZ-U6+I vector. Cultured esophageal cancer cells were transfected with the siRNA-expressing vector (or the control vector) using lipofectamine 2000. BcI-XL gene expression was determined with semiquantitative RT- PCR assay and Western blotting. Among the three siRNA- expressing vectors, the most highly functional vector and its effect on cell growth and apoptosis in esophageal cancer ceils was further analyzed. RESULTS: Of the three siRNA-expressing vectors, siRNA- expressing vector No.1 was the most potent one which suppressed Bcl-XL mRNA production to 32.5% of that in the untreated esophageal cancer cells. Western blotting analysis showed that siRNA-expressing vector No.1 markedly down-regulated the expression of Bcl-XL in human esophageal cancer cells. Treatment of esophageal cancer cells with siRNA-expressing vector No.1 resulted in inhibition of cell growth and induction of apoptosis. CONCLUSION: Down-regulation of BcI-XL by vectorgenerated small interfering RNAs can suppress cell growth and induce apoptosis in human esophageal cancer cells.

This paper has collected some of students' compositions of TEM-4, and according to the score evaluation of the syllabus as well as TEM-4, the paper aims to analyze students' errors and find out their reasons behind in order to improve students' writing capacity in TEM-4.
文摘In the classical multiprocessor scheduling problems, it is assumed that the problems are considered in off\|line or on\|line environment. But in practice, problems are often not really off\|line or on\|line but somehow in between. This means that, with respect to the on\|line problem, some further information about the tasks is available, which allows the improvement of the performance of the best possible algorithms. Problems of this class are called semi on\|line ones. The authors studied two semi on\|line multiprocessor scheduling problems, in which, the total processing time of all tasks is known in advance, or all processing times lie in a given interval. They proposed approximation algorithms for minimizing the makespan and analyzed their performance guarantee. The algorithms improve the known results for 3 or more processor cases in the literature.
文摘While research on pedogenesis mainly focuses on long-term soil formation and most often neglects recent soil evolution in response to human practices or climate changes, this article reviews the impact of artificial subsurface drainage on soil evolution. Artificial drainage is considered as an example of the impact of recent changes in water fluxes on soil evolution over time scales of decades to a century. Results from various classical studies on artificial drainage including hydrological and environmental studies are reviewed and collated with rare studies dealing explicitly with soil morphology changes, in response to artificial drainage. We deduce that soil should react to the perturbations associated with subsurface drainage over time scales that do not exceeding a few decades. Subsurface drainage decreases the intensity of erosion and must i) increase the intensity of the lixiviation and eluviation processes, ii) affect iron and manganese dynamics, and iii) induce heterogeneities in soil evolution at the ten meter scale. Such recent soil evolutions can no longer be neglected as they are mostly irreversible and will probably have unknown, but expectable, feedbacks on crucial soil functions such as the sequestration of soil organic matter or the water available capacity.
