RT_PCR and Southern blotting using 3′_untranslated region (3′_UTR) as specific probe revealed that pea isoactin genes were expressed in roots, stems, leaves, tendrils, pollen and juvenile siliques, but displayed different patterns of transcript accumulation. Two_fold serial dilution electrophoresis showed PEAcⅠ mRNA preferentially accumulated in rapidly developing tissues: it peaked in seven days' stem; remained at a rather high level in leaves within a month but decreased significantly later; varied a little in tendrils and reached a median and a very low level respectively in juvenile siliques and in pollen. PEAcⅡ displayed somewhat similar expression pattern to PEAcⅠ. The observed differences in sequences and transcript accumulation patterns suggest that the individual pea actin genes may differ in their transcriptional regulation and cellular function. Phylogenetic tree of actins showed that pea actin isoforms are as diverged from each other as they are from other plant actins, and pea actins might have originated from a common ancestor before the divergence of the dicot and monocot plants.展开更多

Neurons, flagellated cells, and pigment cells have been traditionally used as model systems to study the cellular functions of kinesins. Here, we report silkworm posterior silkgland (PSG), specialized cells with an extensive endomembrane system for intracellular transport and efficient secretion of fibroin, as a novel model for kinesin study. To investigate kinesindriven intracellular transport in PSG cells, we cloned five silkworm kinesin-like proteins (KLPs), BmKinesin-1, BmKinesin-6, BmKinesin-7, BmKinesin-13, and BmKinesin-14A. We determined their expression patterns by relative real-time PCR and western blotting. Immunofluorescence microscopy verified their colocalization with microtubules. By combining pull-down assays, LC-MS/MS, and western blotting analysis, we identified many potential cargoes of BmKinesin-1 in PSG, including fibroin-containing granules and exuperantia-associated ribonucleoprotein (RNP) complexes. Moreover, BmKinesin-13 overexpression disrupted the microtubule network in BmN cells, which is consistent with a role of Kinesin-13 in regulating microtubule dynamics in other organisms. On the basis of these results, we concluded that PSG might have advantages in elucidating mechanisms of intracellular transport in secretory tissues and could serve as a potential model for kinesin studies.展开更多

Alarm root-cause analysis and alarm prioritization are good for alarm flood reduction. This paper proposes a systematic rationalization method for multivariate correlated alarms to realize the root cause analysis and alarm prioritization. An information fusion based interpretive structural model is constructed according to the data-driven partial correlation coefficient calculation and process knowledge modification. This hierarchical multi-layer model is helpful in abnormality propagation path identification and root-cause analysis. Revised Likert scale method is adopted to determine the alarm priority and reduce the blindness of alarm handling. As a case study, the Tennessee Eastman process is utilized to show the effectiveness and validity of proposed approach. Alarm system performance comparison shows that our rationalization methodology can reduce the alarm flood to some extent and improve the performance.展开更多

UML is a widely used modeling standard, yet it is very difficult to apply it to system verification and testing directly for its imprecise semantics. After analyzing the expression ability of the two models, several transforma- tion rules are defined and each step of transformation is described in detail, after that, the approach is illustrated with an example. The influences of interannual variability of vegetation LAI on surface temperature are investigated via two ensemble simulations, applying the Community Earth System Model. The interannual LAI, derived from Global Inv...The influences of interannual variability of vegetation LAI on surface temperature are investigated via two ensemble simulations, applying the Community Earth System Model. The interannual LAI, derived from Global Inventory Modeling and Mapping Studies NDVI for the period 1982-2011, and its associated climatological LAI, are used in the two ensemble simulations, respectively.The results show that the signals of the influences, represented as ensemble-mean differences, are generally weaker than the noises of the atmospheric variability, represented as one standard deviation of the ensemble differences. Spatially, the signals are stronger over the tropics compared with the mid-high latitudes. Such stronger signals are contributed by the significant linearity between LAI and surface temperature, which is mainly caused via the influences of LAI on evapotranspiration.The maximum amplitudes of the influences on the interannual variability of surface temperature are high and thus deserve full consideration. However, the mean magnitudes of influences are small because of the small changes in the amplitudes of LAI. This work only investigates the influences of the interannual variability of LAI and does not consider interannual changes in other vegetation characteristics, such as canopy height and fractional cover. Further work involving dynamic vegetation models may be needed to investigate the influences of vegetation variability.展开更多
To investigate the existence of the major outer membrane protein (MOMP) gene LipL32 in 15 dominant Chinese strains of 15 serogroups of Leptospira interrogans and 2 international strains of 2 serogroups of Leptospira b...To investigate the existence of the major outer membrane protein (MOMP) gene LipL32 in 15 dominant Chinese strains of 15 serogroups of Leptospira interrogans and 2 international strains of 2 serogroups of Leptospira biflexa, and to clone and construct the expression system as well as to identify the recombinant proteins, genomic DNAs from strains of leptospira were prepared by routine phenol-chloroform method, and the fragments of the LipL32 gene with the whole length from the strains were amplified with high fidelity PCR. The target amplification products were sequenced after T-A cloning, and the expression system for the genes were thereby constructed. Expression of the recombinant proteins was identified by using SDS-PAGE after induction with IPTG at different dosages. Western blot assays with rabbit antiserum against the whole cell of TR/PatocⅠ of Leptospira and immunized serum with rMOMPs were used to determine the immunoreactivity and immunogenicity of the recombinant proteins. Microscopic agglutination test was used to determine the cross- agglutination titres in rabbit sera immunized with rMOMPs, and the cell adherence model of Leptospira was used to examine the blocking effects of rabbit antisera against these rMOMPs. It was found that the LipL32 gene could be found in all the 17 strains of Leptospira mentioned above with two different genotypes, i.e. LipL32/1 and LipL32/2. Amounts of expressions of rMOMP1 and rMOMP2 after IPTG accounted for 40% and 10% of the total bacterial proteins respectively. Both rMOMP1 and rMOMP2 could combine with the rabbit antiserum against leptospiral TR/PatocⅠ, and could induce the production of agglutination antibodies to these 17 strains of Leptospira with 1∶2 to 1∶64 MAT titres. The rabbit anti-rMOMP1 and anti-MOMP2 antibodies at 1∶2 to 1∶16 dilutions could efficiently block adherence of Leptospira. It concludes that all the Leptospira tested in the present study possess LipL32/1 or LipL32/2 genes, and the constructed expression system can express the rMOMP1 and rMOMP2. These recombinant proteins are showed to have good immunogenicity and satisfactory immunoreactivity.展开更多
