不同颜色蝴蝶兰花瓣表皮细胞形态差异比较 被引量:5

为探究不同颜色蝴蝶兰花瓣表皮细胞形态的差异性,采用扫描电子显微镜分别对8个不同颜色的纯色蝴蝶兰品种的花瓣中心部位(2个白色蝴蝶兰品种,3个黄绿色蝴蝶兰品种和3个紫红色蝴蝶兰品种)和4个有斑的蝴蝶兰品种(2个白底红斑的蝴蝶兰品种和... 为探究不同颜色蝴蝶兰花瓣表皮细胞形态的差异性,采用扫描电子显微镜分别对8个不同颜色的纯色蝴蝶兰品种的花瓣中心部位(2个白色蝴蝶兰品种,3个黄绿色蝴蝶兰品种和3个紫红色蝴蝶兰品种)和4个有斑的蝴蝶兰品种(2个白底红斑的蝴蝶兰品种和2个黄底红斑的蝴蝶兰品种)的底色部位、斑色部位及底色与斑色相交部位分别进行观察对比,同时采用鲜切片浸泡在0.25%PEG溶液中观察有斑样品斑纹交界处的纵切面表皮细胞形状变化。结果发现,除2个白色品种的表皮细胞为乳突状外,其他品种的表皮细胞均为穹顶状。另外发现部分样品花瓣表皮存在少量气孔。结果表明,相同颜色不同品种的蝴蝶兰花瓣表皮细胞形状可能不同,但同一片花瓣不同颜色区域的表皮细胞结构则是相同的,说明花瓣颜色并不是影响蝴蝶兰花瓣表皮细胞形状的决定因素。以上不同颜色蝴蝶兰花瓣显微结构的观察比较,将对蝴蝶兰花色研究提供一定的参考。 展开更多

基于秋石斛花朵颜色、色素分布及表皮细胞形态的表型分类 被引量:15

以107个秋石斛(Dendrobium hybrids)品种为试材,利用分光色差仪测定花瓣和唇瓣的颜色并进行聚类分析,通过组织切片观察花瓣和唇瓣的解剖结构,分析花色素分布模式、表皮细胞形态差异以及它们与花色表型间的相关性。结果表明:基于CIELab... 以107个秋石斛(Dendrobium hybrids)品种为试材,利用分光色差仪测定花瓣和唇瓣的颜色并进行聚类分析,通过组织切片观察花瓣和唇瓣的解剖结构,分析花色素分布模式、表皮细胞形态差异以及它们与花色表型间的相关性。结果表明:基于CIELab表色系统的聚类分析将秋石斛分为白、粉、黄绿、紫红、浅紫、紫、深紫、复色等8个色系,以紫色色调为主,颜色相对单一;唇瓣的颜色比花瓣更暗、更深。对72个代表性品种花瓣的解剖观察结果表明,花青素苷有13种组织部位分布模式,花青素苷分布部位越多花色明度会减小并颜色加深,在一品种中发现了能增强颜色的花青素苷体(anthocyanoplasts,ACP);类胡萝卜素广泛分布于花瓣的各个组织部位。秋石斛花存在5种形态的表皮细胞(扁平细胞、圆顶细胞、细长圆顶细胞、乳突细胞和表皮毛细胞),上表皮细胞的圆锥凸起程度大于下表皮,唇瓣上表皮细胞的圆锥程度比花瓣更大。在富含花青素苷的品种中,色相a*值、彩度C值与花瓣表皮细胞的高宽比正相关,在不含花青素苷的品种中,色相a*值、明度L*值与花瓣表皮细胞的高宽比正相关,而彩度C值为负相关;含类胡萝卜素的花朵表皮细胞的圆锥程度显著小于不含类胡萝卜素的类群。 展开更多

魔芋种芋的机械损伤敏感性因素分析 被引量:2

【目的】分析不同种魔芋(Amorphophallus konjac)球茎的机械损伤敏感性因素,为魔芋种芋调种运输提供参考依据。【方法】测定花魔芋(A. konjac)、白魔芋(A. albus)、珠芽魔芋(A. bulbifer)、西盟魔芋(A. krausei)、疣柄魔芋(A. paeoniifo... 【目的】分析不同种魔芋(Amorphophallus konjac)球茎的机械损伤敏感性因素,为魔芋种芋调种运输提供参考依据。【方法】测定花魔芋(A. konjac)、白魔芋(A. albus)、珠芽魔芋(A. bulbifer)、西盟魔芋(A. krausei)、疣柄魔芋(A. paeoniifolius)及红薯(Sweet potato)、马铃薯(Potato)和芋(Taro)等8种薯芋类作物球茎的耐撞击性、水含量、硬度及观察其表皮细胞形态,比较分析不同种魔芋种芋与马铃薯、红薯和芋的机械损伤敏感性因素差异。【结果】各薯芋类作物的球茎撞击损伤后27℃培养3 d,其中,花魔芋、白魔芋、珠芽魔芋和西盟魔芋球茎发病腐烂,而疣柄魔芋、红薯、马铃薯和芋的球茎无任何发病特征。花魔芋、白魔芋、珠芽魔芋和西盟魔芋球茎的水含量分别为86.73%、82.34%、80.01%和80.32%,均不同程度高于疣柄魔芋、马铃薯、红薯和芋;花魔芋、白魔芋、珠芽魔芋和西盟魔芋球茎的硬度较接近,相互间差异不显著(P>0.05),其中花魔芋球茎的硬度最低,为14.40 kg/cm^2,显著低于红薯(37.70 kg/cm^2)和芋(34.17 kg/cm^2);花魔芋、白魔芋和珠芽魔芋球茎的细胞壁较薄,容易破裂,细胞排列不规则;西盟魔芋球茎的细胞较小,但细胞壁松弛,边缘间隙不明显,支持力较弱;疣柄魔芋和芋的细胞壁较松弛且粗糙,细胞排列不规则但相对密集;红薯和马铃薯的细胞壁平滑坚挺,支持力较强。相关性分析结果表明,薯芋类作物球茎的机械损伤差异与其水含量呈显著正相关(P<0.05,下同),与细胞大小呈正相关,与硬度呈显著负相关;水含量分别与硬度和细胞大小呈负相关和正相关;硬度与细胞大小呈负相关。【结论】水含量高、硬度小、表皮细胞壁薄且体积大是不同种魔芋球茎机械伤敏感性高的关键因素,因此在调种环节应采用有效的缓冲包装材料对种芋进行包装后再运输,以减轻魔芋种芋的机械损伤。 展开更多

To identify suitable grass species for growth in the Jianghan Plain,the micromorphologies of the upper and lower epidermis of the leaves of 14 bermudagrass varieties of Cynodon dactylon were investigated,and the number,length,and width of long and short cells and stomata were measured.The results showed that the anticlinal wall of epidermal cells looked like waves and the stoma was distributed in the upper and lower epidermis.However,the various indicators of the leaf epidermis of different species of Cd were significantly different.The length and width of long cells and the length of short cells of Cd Mohawk,Cd Suncity,and wild type Cd were relatively large,which may improve their ability to adapt to the pastoral soil and make them suitable for the rainy environment of the Jianghan Plain.On the contrary,the results of all indicators of Cd Xinnong 1,Cd hulled,and Cd unhulled ranked at the bottom,which was not suitable for pastoral soil habitats,while the wild type Cd could adapt to both the pastoral soil and the vegetation concrete habitat.Moreover,the long cells and short cells of Cd Kashi were tightly arranged,and the stoma in Cd Pyramid and Cd hulled were more dense,which may improve their ability to adapt to an arid environment. 展开更多

Reliable Analytic Strategy to Correlate the Morphological and Cytological Parameters on Lupinus termis L, against Fusarium oxysporum Infection

Many analytic strategies have emerged to estimate plant responses to Fusarium wilt. The demand for fast and reliable method (diagnosis, prediction) to determine isolate strength accurately is not established yet. Early determination of pathogen strength helps in plant medication. The aim of this study was to develop a faster strategy and method for early determination of fungal isolates strength in correlation to plant response. Till now, the scientists have no consensus on the most correlated parameters that could express wilt precisely. In this study, 30 isolates of Fusarium oxysporum isolated from Lupinus termis L. were used to provide an explicit image about the real strength of Fusarium isolates and its impact on the plant. Wilting percentage ranged from 26.67% to 93.33% of the infected plants depending on isolate virulence. Some of cellular, morphological and physical measurements were conducted on 8 out of 30 isolates, including root (length, fresh weight (FW) and dry weight (DW)), nodules (water content (WC), FW, DW), stem (height, WC, FW, DW), total leaves/plant (WC, FW, DW) and the fourth leaf (WC, FW, DW, leaf area, epidermal cell area, epidermal cell number, succulence). Hierarchical clustering was used to determine the variance between the isolates. Detrended correspondence analysis (DCA) and canonical correspondence analysis (CCA) were used to determine the most important growth parameters that could express wilting accurately. The CCA results showed that most of the measured parameters on the fourth leaf, except for leaf epidermal cell number, were highly and positively correlated to wilt. That makes these specific parameters valuable and sensitive for any changes in isolates strength. Accordingly, a mathematical model was created to be helpful in the quick determination of isolate strength and precise medication. 展开更多
