目的建立一种基于表面等离子体共振传感器技术检测人类甲型流感病毒的方法,并进行初步诊断应用。方法分别设计基于甲型流感M基因的引物和探针,制备RT-PCR扩增产物,优化并确定基于传感技术的实验条件,建立高灵敏度和高特异度的表面等离... 目的建立一种基于表面等离子体共振传感器技术检测人类甲型流感病毒的方法,并进行初步诊断应用。方法分别设计基于甲型流感M基因的引物和探针,制备RT-PCR扩增产物,优化并确定基于传感技术的实验条件,建立高灵敏度和高特异度的表面等离子体共振传感器方法。结果缓冲液的盐离子强度和pH值是影响杂交效果的关键因素,对其优化后标准曲线参数良好,甲型流感病毒标准品的Ru值与其相应梯度稀释标准品浓度呈良好的线性关系(R2>0.99),同时,该传感器在200 mmol/L氯化钠的磷酸盐缓冲液中可以有效区分DNA的完全匹配与单碱基错配,以乙型流感探针作为质控,该探针并不与甲型流感扩增产物发生特异性杂交。结论成功建立了高灵敏度和高特异度的甲型流感病毒表面等离子体共振传感器检测方法,为临床诊断或感染病例提供了技术平台。 展开更多

光纤包层型表面等离子体共振(surface plasmon resonance,SPR)传感器不需去除包层,可用于生化传感,但该传感器存在结构种类较少的问题。为此该文提出一种基于S型光纤的新型包层SPR传感器结构,利用电熔技术制作S型光纤,在S型结构后面的... 光纤包层型表面等离子体共振(surface plasmon resonance,SPR)传感器不需去除包层,可用于生化传感,但该传感器存在结构种类较少的问题。为此该文提出一种基于S型光纤的新型包层SPR传感器结构,利用电熔技术制作S型光纤,在S型结构后面的包层表面镀制50 nm金膜,实现纤芯中的光耦合至包层,然后与包层表面金膜接触,构成光纤包层型SPR传感器,最后实现了Hg^(2+)浓度检测。在5 pmol/L~5 mmol/L的浓度检测范围内,SPR共振谷波长由633.03 nm移动至645.09 nm,检测灵敏度达1.382 nm/lg(mmol/L)。S型光纤包层SPR传感器制作简单,可用于单模、多模光纤以实现包层SPR传感,尤其适用于生化检测领域。 展开更多

A fast and facile method of fabricating fiber-optic localized surface plasmon resonance sensors baseff on spherical gold nanoparticles was introduced in this study. The gold nanoparticles with an average diameter of 55 nm were synthesized via the Turkevich method and were then immobilized onto the surface of an uncladded sensor probe using a polydopamine layer. To obtain a sensor probe with high sensitivity to changes in the refractive index, a set of key optimization parameters, including the sensing length, coating time of the potydopamine layer, and coating time of the gold nanoparticles, were investigated. The sensitivity of the optimized sensor probe was 522.80 nm per refractive index unit, and the probe showed distinctive wavelength shifts when the refractive index was changed from 1.328 6 to 1.398 7. When stored in deionized water at 4 ℃, the sensor probe proved to be stable over a period of two weeks. The sensor also exhibited advantages, such as low cost, fast fabrication, and simple optical setup, which indicated its potential application in remote sensing and real-time detection. 展开更多

To obtain the influence rules of the coating parameters of a long-period fiber grating(LPFG)with respect to temperature,strain and refractive index sensing properties,based on the mode coupling theory,a strict four-layer theorietical model of a metal film coated LPFG is established,and these parameters that affect the spectral characteristics of the metal film coated LPFG are studied.The simulation results show that there is an optimal metal film thickness on the surface of the LPFG that will induce the surface-plasmon resonance(SP R)effect,which results in higher sensitivity to the environmental temperature and refractive index but has little influence on the strain There is theoretical evidence that when the silver thickness is between0.8and1.2nm,the refractive index sensitivity will reach the peak point of42.4026,at which the refractive index sensor sensitivity is increased by4.S%.The theoretical results of coating a long-period fiber grating provide a good theoretical basis and guidance for LPFG design and parameters optimization 展开更多

These flavonoids are thought to be haptens that can induce allergic reactions. The interaction mechanism of these haptens with bovine serum albumin (BSA) was investigated by surface plasmon resonance (SPR) and molecular modeling method. The SPR study indicated that these compounds could specifically bind to the BSA with one binding site and equilibrium dissociation constant (KD) values were determined. Molecular modeling explored the mechanism of interaction under simulated physiological condition. The result of molecular modeling indicated that flavonoids could bind with BSA in the hydrophobic pocket of sub-domain II with hydrogen bonding as the main acting force. 展开更多

An overview of recent researches of surface plasmon resonance (SPR) sensing technology in Laboratory of Science and Technology of Micro-Nano Optics (LMNO), University of Science and Technology of China, is presented. Some novel SPR sensors, such as sensors based on metallic grating, metal-insulator-metal (MIM) nanoring and optical fiber, are designed or fabricated and tested. The sensor based on localized surface plasmon resonance (LSPR) of metallic nanoparticles is also be summarized. Because of the coupling of propagating surface plasmons and localized surface plasmons, the localized electromagnetic field is extremely enhanced, which is applied to surface-enhanced Raman scattering (SERS) and fluorenscence enhancement. We proposed a new bilayer surface plasmon resonance-based fiber-optic refractive index sensor with silver and an over-layer of TiO2. We numerically investigated the optimal thickness of TiO2 over-layer in the proposed sensor and compared its performance to that based on typical bimetallic layers of silver-and-gold in the aqueous media using finite-difference time domain approach. We show that the use of TiO2 over-layer greatly improves the sensor performance in terms of sensitivity and signal-to-noise ratio compared to that with gold as the over-layer. Not only does the TiO2 over-layer offer a cost-effective alternative to gold for overcoming the oxidation problem, but also it allows resonance wavelength-tunability. 展开更多

Optical fiber based SPR sensors have attracted more and more attention due to their unique advantages over the prism-based SPR sensors. A novel fiber-optic SPR sensor with multi-alternating metal layers for biochemical analysis is presented in this paper. Based on the fundamental SPR theory of the fiber optic sensing technology, we theoretically investigated the effects of the existence of alternating layers deposited on sensing region SPR wavelength changes. The emphasis was placed on the numerical simulation of the fiber-optic SPR sensor's sensitivity which could be affected by its technical parameters such as the metal thickness, number of alternating layers. Results showed that, compared to the normal SPR sensor with the single metal layer, the proposed sensor had a wider detecting range of the refractive index and higher sensitivity, which can find applications in biological analysis. 展开更多

Silica-based fiber tips are used in a variety of spectroscopic, micro- or nano-scopic optical sensor applications and photonic micro-devices. The miniaturization of optical sensor systems and the technical implementation using optical fibers can provide new sensor designs with improved properties and functionality for new applications. The selective-etching of specifically doped silica fibers is a promising method in order to form complex photonic micro structures at the end or within fibers such as tips and cavities in various shapes useful for the all-fiber sensor and imaging applications. In the present study, we investigated the preparation of geometrically predefined, nanoscaled fiber tips by taking advantage of the dopant concentration profiles of highly doped step-index fibers. For this purpose, a gas phase etching process using hydrofluoric acid (HF) vapor was applied. The shaping of the fiber tips was based on very different etching rates as a result of the doping characteristics of specific optical fibers. Technological studies on the influence of the etching gas atmosphere on the temporal tip shaping and the final geometry were performed using undoped and doped silica fibers. The influence of the doping characteristics was investigated in phosphorus-, germanium-, fluorine- and boron-doped glass fibers. Narrow exposed as well as protected internal fiber tips in various shapes and tip radiuses down to less than 15 nm were achieved and characterized geometrically and topologically. For investigations into surface plasmon resonance effects, the fiber tips were coated with nanometer-sized silver layers by means of vapour deposition and finally subjected to an annealing treatment. 展开更多
