[Method] The wing venous blood of Donglan Black-bone chicken(silky feather and contour feather) was collected, and the total DNA was extracted. The 18 microsatellite loci were amplified by PCR technology, and the obtained gene fragments were scanned. The number of alleles(Na), number of effective alleles(Ne), heterozygosity(H) and polymorphism information content(PIC) were determined using Pop Gen 32 biological software to do cluster analysis of Guangxi indigenous chicken breeds. [Result] The18 microsatellite loci of Donglan black-bone chicken were found to be highly polymorphic. A total of 98 alleles were detected out from 80 Donglan black-bone chicken samples. The averages of Na, Ne, PIC and H per locus were 5.44, 2.78, 0.5382 and 0.596 3, respectively. A total of 93 and 98 alleles were detected out from silky- and contour-feather Donglan black-bone chickens, respectively. The averages of Ne per locus were 2.62 and 2.76, of PIC per locus were 0.508 8 and 0.539 8,of H per locus were 0.624 7 and 0.599 1, respectively. The results of cluster analysis showed that there was greater genetic distance between Donglan black-bone chicken and any of other Guangxi indigenous chicken breeds, including three yellow chicken, Xiayan chicken and Nandan Yao chicken, which was consistent with the breed evolution, geographical distribution and breeding history of Donglan black-bone chicken. [Conclusion] There are greater genetic variation and abundant genetic diversity in the conserved population of Donglan black-bone chicken, suggesting abundant genetic diversity and great breeding potential of Donglan black-bone chicken.展开更多
The taxonomy of Coilia has been extensively studied in China,and yet phylogenetic relationships among component taxa remain controversial.We used a PCR-based fingerprinting technique,amplified fragment length polymorp...The taxonomy of Coilia has been extensively studied in China,and yet phylogenetic relationships among component taxa remain controversial.We used a PCR-based fingerprinting technique,amplified fragment length polymorphism(AFLP) to characterize and identify all four species of Coilia in China.We examined the genetic relationships of the four species of Coilia and a subspecies of Coilia nasus with AFLP.A total of 180 AFLP loci were generated from six primer combinations,of which 76.11% were polymorphic.The mean genetic distance between pairs of taxa ranged from 0.047 to 0.596.The neighbor-joining tree and UPGMA dendrogram resolved the investigated species into three separate lineages:(1) C.mystus,(2) C.grayii and(3) C.brachygnathus,C.nasus,and C.nasus taihuensis.Phylogenetic analysis of the AFLP data is inconsistent with current morphological taxonomic systems.The AFLP data indicated a close relationship among C.brachygnathus,C.nasus taihuensis,and C.nasus.Therefore,the two species described under Coilia(C.brachygnathus and C.nasus taihuensis) are treated as synonyms of C.nasus.展开更多

[Method] The genetic diversity of 141 copies of Hu mulberry varieties was analyzed by ISSR molecular markers. [Result] 12 ISSR primers had amplified a total of 90 amplified,of which 57 bands were polymorphic,and the polymorphic rate was 63.33%. The genetic similarity coefficients of 141 Hu mulberry germplasm resources varied from 0.633 3 to 1.000 0 with the average of 0.483 35,indicating that there was difference on genetic diversity among different varieties of Hu mulberries. A dendrogram of all 141 Hu mulberry varieties based on the genetic similarity coefficients using ISSR molecular markers was generated by UPGMA cluster method. Clustering of the 141 Hu mulberry varieties did not correspond with the conventional classification involving differences in style,leaf,branch,fruit and other morphological or agronomical characters. [Conclusion] Four subgroups clearly represented the genetic relationships in the 141 accessions which were benefit for the variety improvement and germplasm resource conservation.展开更多
