The survival thermal maximum for L. crocea was 33.0℃, the 50% critical thermal maximum (50% CTMax) was 35.5℃, and the critical thermal maximum (CTMax) was 36.0℃. Three microsatellite markers (LYC0148, LYC0200 and LYC0435), associated with thermal tolerance were screened and identified using a Bulked Segregation Analysis (BSA) method. These markers have six amplified fragments in which four are related to thermal tolerance. These fragments were cloned and sequenced, and the results showed the core motif were all "AC" repeats. For LYC0148 and LYC0200, the lengths of fragments are 18l bp and 197 bp, respectively. For LYC0435, which has two fragments, the fragment lengths are 112 bp and 100 bp. The results provide useful molecular markers for thermal-tolerance breeding of large yellow croaker in the near future.展开更多
In this present work, the best conditions for production of peptidases under solid state fermentation by the fungi Penicillium corylophilum and Penicillium waksmanii, partial purification using Sephadex G-75 gel filtr...In this present work, the best conditions for production of peptidases under solid state fermentation by the fungi Penicillium corylophilum and Penicillium waksmanii, partial purification using Sephadex G-75 gel filtration column, as well as the biochemical characterization of the partial purified enzymes were investigated. P. corylophilum showed the best production in medium containing wheat bran, agro-industrial residue, without additives (egg albumin or casein), in which peptidase activity reached 520 U mL^-1 and the enzyme displayed the optimum activity between pH range from 7 to 8 and 60 ℃. It also showed high stability in wide pH range and temperature until 45 ℃ for 60 min of incubation. On the other hand, P. waksmanii, the best production was noted in a medium containing wheat bran (95%) and casein (5%), reaching 545 U mL^-1, with proteolytic optimum activity at pH 7.5 and 55 ℃. The enzyme was mainly stable in pH range from 8 to 9 and at temperatures until 45 ℃ for 60 rain of incubation. The peptidases secreted by both fungi were inhibited in the presence of phenylmethane sulfonyl fluoride, showing that they belong to the subclass of serine peptidases.展开更多

The seeds of Badami-e-Zarand and Sarakhs cultivars were treated with the f...This experiment was carried out in order to investigate the effect of diurnal temperature fluctuations on seed germination of Pistacia vera L. The seeds of Badami-e-Zarand and Sarakhs cultivars were treated with the following temperatures: T1 = 24 hr at 25℃, T2= 16 hr at 25 ℃ + 8 hr at 10 ℃, T3= 12 hr at 25 ℃ + 12 hr at 10℃ and T4= 8 hr at 25℃ + 16 hr at 10℃. The germination percentage was increased by diurnal temperature fluctuations, however all of treatments extended MTG compared to control (T1). Temperature affected the quality of roots, in a matter that high constant temperature resulted in dichotomous (forked) roots during seed germination of pistachio. The response of cultivars was different in this case, so that, Badami-e-Zarand had higher dichotomous root percentage. This disorder can be decreased by diurnal temperature fluctuation.展开更多
