Compared with other biological nanopores and solid-state nanopores, the glass capillary nanopore has low cost, easy availability and stable mechanical characteristics, thus it has been widely used in the nanopore technology for single molecule detection. In this review, we will focus on the studies of the glass nanopore sensors. The popular glass nanopore fabrication methods would be introduced, and the applications of glass nanopores in the detection of nanoparticles, proteins and DNA molecules would be presented. Gene replacement therapy for ra- re genetic disorders is perhaps the most extreme form of personalized medicine, in that th...Personalized medicine aims to utilize genomic information about patients to tailor treatment. Gene replacement therapy for ra- re genetic disorders is perhaps the most extreme form of personalized medicine, in that the patients' genome wholly determines their treatment regimen. Gene therapy for retinal disorders is poised to become a clinical reality. The eye is an optimal site for gene therapy due to the relative ease of precise vector delivery, immune system isolation, and availability for monitoring of any potential damage or side effects. Due to these advantages, clinical trials for gene therapy of retinal diseases are currently underway. A necessary precursor to such gene therapies is accurate molecular diagnosis of the mutation(s) underlying disease. In this review, we discuss the application of Next Generation Sequencing (NGS) to obtain such a diagnosis and identify disease causing genes, using retinal disorders as a case study. After reviewing ocular gene therapy, we discuss the application of NGS to the identification of novel Mendelian disease genes. We then compare current, array based mutation detection methods against next NGS-based methods in three retinal diseases: Leber's Congenital Amaurosis, Retinitis Pigmentosa, and Stargardt's disease. We conclude that next-generation sequencing based diagnosis offers several advantages over array based methods, including a higher rate of successful diagnosis and the ability to more deeply and efficiently assay a broad spectrum of mutations. However, the relative difficulty of interpreting sequence results and the development of standardized, reliable bioinforrnatic tools remain outstanding concerns. In this review, recent advances NGS based molecular diagnoses are discussed, as well as their implications for the development of personalized medicine.展开更多

METHODS: Eighty-four subjects recruited from sub- stance abuse cl...OBJECTIVES: To explore the distribution of TCM patterns in ketamine users and the inter-rater reli- ability of TCM diagnosis among TCM practitioners. METHODS: Eighty-four subjects recruited from sub- stance abuse clinics and non-governmental coun- seling services catering for ketamine abusers were examined by one or two TCM practitioners. The dis- tribution and inter-rater reliability of the TCM diag- noses between the two TCM practitioners were de- scribed and measured. RESULTS: Seven TCM patterns were differentiated. The four most common patterns were the Heart Yin deficiency (29.8%), Kidney Yang deficiency (26.2%), Kidney Yin deficiency (19.0%), and Spleen Yang defi-ciency (14.3%). The agreement between the two TCM practitioners was 59%, the kappa for the three most common patterns was 0.472 (P=O.O03). The proportion of men was lower in the Kidney Yin defi- ciency group (18.8%), in comparison to the Heart Yin deficiency (60.0%, P=0.009) and Kidney Yang de- ficiency group (59.1%, P=0.013). CONCLUSION: The differences between the TCM groups in terms of the source of referral and num- ber of drug uses other than ketamine were of bor- derline significance. Further research should focus on validating diagnostic tools and improving in- ter-rater reliability in TCM to enhance the quality of clinical trials of TCM treatments for ketamine abuse.展开更多

Ten sessions of classic massage of the lower limbs were performed on the subjects. The massage sessions were conducted every day over ten consecutive days. Throughout the treatment, the subjects' blood pressure was measured using a manometer with a stethoscope TRO-CARDIO KIT 2 MONO. Their blood pressure was taken 1 min before the massage, as well as 1 min and5 min after each session. The study took place between March and June 2013 in Szpital Specjalistyczny im. J?drzeja ?niadeckiego(J?drzej ?niadecki Specialist Hospital) in Nowy S?cz, at the Department of Internal Diseases.RESULTS: For ten consecutive days, the blood pressure values in the examined women were decreasing, with the exception of the diastolic blood pressure measured 5 min after the massage.CONCLUSION: The drops in blood pressure by several percent, noted in the majority of the female subjects over the time of the study. Classic massage might provide a safe supportive measure in pharmacologic treatment of hypertension.展开更多
