In order to improve the production of human-like collagen III(HLC III)by fed-batch culture of recombinant Escherichia coli BL21,the Plackett-Burman and Box-Behnken design were applied to optimize the fermentation proc...In order to improve the production of human-like collagen III(HLC III)by fed-batch culture of recombinant Escherichia coli BL21,the Plackett-Burman and Box-Behnken design were applied to optimize the fermentation process parameters.Three variables(induction time,inoculum age and pH),which have significant effects on HLC III production,were selected from eight variables by Plackett-Burman design.With the regression coefficient analysis in the Box-Behnken design,a relationship between HLC III production and three significant factors was obtained,and the optimum levels of the three variables were as follows:induction time 3.2h,inoculum age 12.6 h and pH 6.7.The 3D response surface plots and 2D contour plots created by the Box-Behnken design showed that the interaction between induction time and pH and that between innoculum age and pH were significant.An average 9.68 g·L1HLC III production was attained in the validation experiment under optimized condition,which was 80%higher than the yield of 5.36 g·L1before optimization.展开更多

Microstructure evolution of ECAE-formed ZK...The two-pass equal channel angular extrusion (ECAE) process was introduced into strain-induced melt activation (SIMA) to predeform a ZK60 alloy with rare earth (RE) addition. Microstructure evolution of ECAE-formed ZK60+RE alloy during reheating was investigated. Furthermore, tensile properties of thixoforged components were determined. The results show that the SIMA process can produce ideal microstructures, and spheroidized solid particles with little entrapped liquid can be obtained. With prolonging holding time, the size of solid particles increases and the degree of spheroidization is improved. The tensile properties of the thixoforged ZK60+RE samples are close to those of two-pass ECAE-formed samples.展开更多
The Wulipo landslide, triggered by heavy rainfall on July 10, 2013, transformed into debris flow,resulted in the destruction of 12 houses, 44 deaths, and 117 missing. Our systematic investigation has led to the follow...The Wulipo landslide, triggered by heavy rainfall on July 10, 2013, transformed into debris flow,resulted in the destruction of 12 houses, 44 deaths, and 117 missing. Our systematic investigation has led to the following results and to a new understanding about the formation and evolution process of this hazard. The fundamental factors of the formation of the landslide are a high-steep free surface at the front of the slide mass and the sandstone-mudstone mixed stratum structure of the slope. The inducing factor of the landslide is hydrostatic and hydrodynamic pressure change caused by heavy continuous rainfall. The geological mechanical model of the landslide can be summarized as "instability-translational slide-tension fracture-collapse" and the formation mechanism as "translational landslide induced by heavy rainfall". The total volume of the landslide is 124.6×104 m3, and 16.3% of the sliding mass was dropped down from the cliff and transformed into debris flow during the sliding process, which enlarged 46.7% of the original sliding deposit area. The final accumulation area is found to be 9.2×104 m2. The hazard is a typical example of a disaster chain involving landslide and its induced debris flow. The concealment and disaster chain effect is the main reason for the heavy damage. In future risk assessment, it is suggested to enhance the research onpotential landslide identification for weakly intercalated slopes. By considering the influence of the behaviors of landslide-induced debris flow, the disaster area could be determined more reasonably.展开更多

The specimen was first...In order to investigate the springback performance of a cold-rolled transformation-induced plasticity (TRIP) steel sheet during cyclic bending, three-points bending experiments were performed. The specimen was firstly forward bended and then reversed bended according to three basic punch strokes, 10 mm, 20 mm and 30 mm, respectively. The outlines of the deformed specimens, punch load and crosshead displacement were obtained. The experimental procedures and the method of determining the springback displacement were reported in detail. The contributi^m of the strain hardening and decrease of elastic modulus to the overall springback was explained. The results demonstrated that springback increases both with the cycles and with the punch stroke during the cyclic bending process. Furthermore, springback in forward bending is larger than that in the corresponding reverse bending. This may be helpful to understand the complex springback behavior of the complicated sheet metal pancls.展开更多

In this article, we employ a Support Vector Machine (SVM) and Gaussian Process Regression (GPR) for prediction of reservoir induced earthquake M based on reservoir parameters. Comprehensive parameter (E) and maximum reservoir depth] (H) are considered as inputs to the SVM and GPR. We give an equation for determination oil reservoir induced earthquake M. The developed SVM and GPR have been compared with] the Artificial Neural Network (ANN) method.
