[Method] The aseptic seedl...Objective The purpose of this experiment was to investigate the effects of exogenous hormone factors on subculture multiplication and root induction of tis-suecultured seedlings in mulberry. [Method] The aseptic seedlings of Guisangyou 12 were used as the materials and the comparative experiment on subculture multi-plication and root induction used different kinds,concentrations and combinations of exogenous hormones. [Result] The hormone combination of 0.1 mg/L IBA+1.5 mg/L 6-BA+0.03 mg/L TDZ showed the best effects to bud multiplication, and the multiple of bud multiplication was 5.28. The effects of NAA was better than IBA and IAA on root induction. The hormone combination of 2.0 mg/L NAA+1.0 mg/L PP333 was most suitable to the root induction, in which the rooting rate arrived to 100%, the root number was 7.01 and the root length was 1.38 cm on average. [Conclusion] The results wil provide some technical reference for large-scale propagation of mul-berry seedlings in vitro.展开更多
The ability of triadimefon (TDM), a triazolic fungicide, to alter the biochemical constituents and thereby minimizing the days required for sprouting in white yam (Dioscorea rotundata Poir.) tubers during storage ...The ability of triadimefon (TDM), a triazolic fungicide, to alter the biochemical constituents and thereby minimizing the days required for sprouting in white yam (Dioscorea rotundata Poir.) tubers during storage under (30±2) ℃ in the dark, was studied. TDM at 20 mg/L was given to tubers by dipping the tubers in treatment solution containing 20 mg/L TDM on 10, 25 and 40 d after storage (DAS). Starch, sugars, protein, amino acid contents as well as protease and a-amylase activities were estimated on 15, 30 and 45 DAS from two physiological regions viz., apical and basal regions of the tubers. In normal conditions (control) sprouting occurred on 70 to 80 DAS. The starch content decreased, while protein, amino acid, sugar contents and protease and a-amylase activities were increased due to TDM treatment and led to early sprouting.展开更多

We have previously identified an Arabidopsis mutant bud2 that displays altered...The ratio of auxin and cytokinin plays a crucial role in regulating aerial architecture by promoting or repressing axillary bud outgrowth. We have previously identified an Arabidopsis mutant bud2 that displays altered root and shoot architecture, which results from the loss-of-function of S-adenosylmethionine decarboxylase 4 (SAMDC4). In this study, we demonstrate that BUD2 could be induced by auxin, and the induction is dependent on auxin signaling. The mutation of BUD2 results in hyposensitivity to auxin and hypersensitivity to cytokinin, which is confirmed by callus induction assays. Our study suggests that polyamines may play their roles in regulating the plant architecture through affecting the homeostasis of cytokinins and sensitivities to auxin and cytokinin.展开更多
The effects of sterilization pretreatment, sampling time and growth regulators on callus formation, explant contamination and adventitious bud induction from stems of balsam pear (Momordica charantia L.) were invest...The effects of sterilization pretreatment, sampling time and growth regulators on callus formation, explant contamination and adventitious bud induction from stems of balsam pear (Momordica charantia L.) were investigated. Sterilized for 6 rain with 0.1% (w/v) mercuric chloride solution is an essential sterilization method for stems. June and July proved to be better for material taking, followed by May. The callus formation rate of balsam pears reached the highest when medium added with IBA 1.0mg/L and BA 2.0 mg/L, while the optimum medium for adventitious bud induction was MS added with IBA 0.5mg/L and BA 4.0 mg/L. And it will provide theoretical and technical basis for rapid propagation and breeding.展开更多
