It comprises two parts: (1) ace I gene encoding copper-responsive transcription factor under the control of a constitutive or organ-specific promoter, and (2) a gene of interest under the control of a chimeric promoter consisting of the CaMV 35S (-90 to +8) promoter linked to the metal responsive element (MRE) carrying activating copper-metallothionein expression (ACE1)-binding sites. Here, the effectiveness of two different ACE1-binding cis -elements which derive from 5'-regulatory region of yeast metallothionein gene was investigated in transgenic tobacco (Nicotiana tabacum L. cv. W38). The results revealed that the MRE (-210 to -126) could increase the system inducibility by 50% - 100% compared with the previously reported MRE (-148 to -105). It is potential to use the copper-inducible system to control valuable gene traits in plant biotechnology.展开更多

At lower oxygen coverage, it was found th...The reconstructed structures of Cu(100) surface induced by O2 dissociative adsorption were investigated by low energy electron diffraction and scanning tunneling microscopy. At lower oxygen coverage, it was found that two reconstructed structures, i.e. c(2×2)-O and (√2×2√2)R45°-O are coexistent. The domain size of the c(2×2)-O structure decreased with the increasing of O2 exposure. The reconstructed structure at very small coverage was also investigated and a “zigzag” structure was observed at this stage. The “zigzag” structure was identified as boundaries of local c(2×2) domains. It was found that the strip region shows much stronger molecule-substrate interaction than that of oxygen covered regions, making it a proper template for patterned organic films. The sequence of the thermal stability was found as zigzag structure〉c(2×2)〉(√2×2√2)R45°-O.展开更多
A new rhodamine derivative,N-(3-carboxy)acryloyl rhodamine B hydrazide(CARB),has been synthesized,and its unusual spectroscopic reaction with Cu2+ has been investigated.The derivative exhibits a rapid and reversible n...A new rhodamine derivative,N-(3-carboxy)acryloyl rhodamine B hydrazide(CARB),has been synthesized,and its unusual spectroscopic reaction with Cu2+ has been investigated.The derivative exhibits a rapid and reversible non-fluorescent absorption upon coordination to Cu2+,which is a rather unusual phenomenon for rhodamine B derivatives.Stoichiometric measurements using the Job's method and the molar ratio method reveal that one CARB molecule combines two Cu2+ ions,and the two Cu2+ ions play different roles:one opens the spirocyclic structure and the other quenches the fluorescence of the xanthene moiety.This reaction mechanism is supported by a comparative study on the model compound N-acryloyl rhodamine B hydrazide as well as by the density functional theory calculations.Furthermore,the absorption response of CARB is highly selective for Cu2+ over other common ions,which implies that CARB may be used as a colorimetric probe for the rapid visual detection of Cu2+.展开更多
A high non-photochemical quenching(NPQ) appeared below the phase transition temperature when Microcystis aeruginosa PCC7806 cells were exposed to saturated light for a short time.This suggested that a component of NPQ...A high non-photochemical quenching(NPQ) appeared below the phase transition temperature when Microcystis aeruginosa PCC7806 cells were exposed to saturated light for a short time.This suggested that a component of NPQ,independent from state transition or photo-inhibition,had been generated in the PSII complex;this was a fast component responding to high intensity light.Glutaraldehyde(GA),commonly used to stabilize membrane protein conformations,resulted in more energy transfer to PSII reaction centers,affecting the energy absorption and dissipation process rather than the transfer process of phycobilisome(PBS).In comparison experiments with and without GA,the rapid light curves(RLCs) and fluorescence induction dynamics of the fast phase showed that excess excitation energy was dissipated by conformational change in the photosynthetic pigment proteins on the thylakoid membrane(PPPTM).Based on deconvolution of NPQ relaxation kinetics,we concluded that the fast quenching component(NPQ f) was closely related to PPPTM conformational change,as it accounted for as much as 39.42% of the total NPQ.We hypothesize therefore,that NPQ f induced by PPPTM conformation is an important adaptation mechanism for Microcystis blooms under high-intensity light during summer and autumn.展开更多
The production of thermostable laccases from a native strain of the white-rot fungus Pycnoporus sanguineus isolated in Mexico was enhanced by testing different media and a combination of inducers including copper sulf...The production of thermostable laccases from a native strain of the white-rot fungus Pycnoporus sanguineus isolated in Mexico was enhanced by testing different media and a combination of inducers including copper sulfate(CuSO4).The best conditions obtained from screening experiments in shaken flasks using tomato juice,CuSO4,and soybean oil were integrated in an experimental design.Enhanced levels of tomato juice as the medium,CuSO4and soybean oil as inducers(36.8%(v/v),3 mmol/L,and 1%(v/v),respectively) were determined for 10 L stirred tank bioreactor runs.This combination resulted in laccase titer of 143000 IU/L(2,2'-azino-bis(3-ethylbenzthiazoline-6- sulfonic acid),pH 3.0),which represents the highest activity so far reported for P.sanguineus in a 10-L fermentor.Other interesting media resulting from the screening included glucose-bactopeptone which increased laccase activity up to 20000 IU/L,whereas the inducers Acid Blue 62 and Reactive Blue 19 enhanced enzyme production in this medium 10 times.Based on a partial characterization,the laccases of this strain are especially promising in terms of thermostability(half-life of 6.1 h at 60 °C) and activity titers.展开更多
