[Method] The aseptic seedlings of Guisangyou 12 were used as the materials and the comparative experiment on subculture multi-plication and root induction used different kinds,concentrations and combinations of exogenous hormones. [Result] The hormone combination of 0.1 mg/L IBA+1.5 mg/L 6-BA+0.03 mg/L TDZ showed the best effects to bud multiplication, and the multiple of bud multiplication was 5.28. The effects of NAA was better than IBA and IAA on root induction. The hormone combination of 2.0 mg/L NAA+1.0 mg/L PP333 was most suitable to the root induction, in which the rooting rate arrived to 100%, the root number was 7.01 and the root length was 1.38 cm on average. [Conclusion] The results wil provide some technical reference for large-scale propagation of mul-berry seedlings in vitro.展开更多
The artemisinin accumulation in the hairy root cultures of Artemisia annua L. was enhanced via a treatment of three fungal elicitors separately ( Verticillium dahliae Kleb., Rhizopus stolonifer (Ehrenb. ex ...The artemisinin accumulation in the hairy root cultures of Artemisia annua L. was enhanced via a treatment of three fungal elicitors separately ( Verticillium dahliae Kleb., Rhizopus stolonifer (Ehrenb. ex Fr.) Vuill and Colletotrichum dematium (Pers.) Grove). Among these three elicitors, V. dahliae had the highest inducing efficiency, but none of them manifests any noticeable effects on the cell growth of the hairy root cultures. The artemisinin content of the hairy root cultures treated with V. dahliae elicitor was 1.12 mg/g DW, which was 45% higher than the control (0.77 mg/g DW). The results showed that elicitation was dependent on the elicitor concentration, the incubation period and the physiological stage at which the hairy root cultures were treated. In addition, the authors found that for V. dahliae , the optimum concentration was 0.4 mg carbohydrate per millilitre medium, the strongest response of A. annua hairy root cultures to the elicitation was at the late exponential growth stage, and the highest artemisinin content of the hairy root cultures was on the 4th day post treatment.展开更多

When hairy roots of 22-day-old cultures (la...The oligosaccharide elicitor from the mycelial wall of an endophytic Colletotrichum sp. B501 promoted the production of artemisinin in Artemisia annua L. hairy root culture. When hairy roots of 22-day-old cultures (later growth phase) were exposed to the elicitor (20 mg/L) for 4 d, the maximum content of artemisinin reached 1.15 mg/g, a 64.29% increment over the control. The electron X-ray microanalysis disclosed the rapid accumulation of Ca 2+ in the elicited cortical cells of hairy root. The electronic microscope observation revealed the high electron density area in vacuole of elicited cells. During the first day of elicitation the peroxidase activity of hairy roots was improved sharply. Some cellular morphological changes including cell shrinkage, condensation of cytoplasm and nuclear fragmentation, coincident with the appearance of DNA ladders, were observed after the third day of elicitation. It was suggested that the oligosaccharide elicitor triggered the programmed cell death, which may provide the substance or chemical signal for artemisinin biosynthesis.展开更多

The results showed that the effects of CrO3 concentration on the mitotic indexes were complicated. CrO3 increases the micronucleus rate of Vicia faba root tip cells. It was found that within certain range of CrO3 concentration the micronucleus rate increased systematically with increased concentration of CrO3, but that the micronucleus rate decreased at higher level of CrO3 and that CrO3 also caused various types of chromosome aberration at a rate which increased systematically with increased concentration of CrO3. We concluded that CrO3 has significant mutagenic effect on Vicia faba root tip cells.展开更多
It is increasingly recognized that metal bioavailability is a better indicator of the potential for phytoremediation than the total metal concentration in soils; therefore, an understanding of the influence of phytore...It is increasingly recognized that metal bioavailability is a better indicator of the potential for phytoremediation than the total metal concentration in soils; therefore, an understanding of the influence of phytoremediation plants on metal dynamics at the soil-root interface is increasingly vital for the successful implementation of this remediation technique. In this study, we investigated the heavy metal and soil solution chemical changes at field moisture, after growth of either Indian mustard (Brassica juncea) or sunflower (Helianthus annuus L.), in long-term contaminated soils and the subsequent metal uptake by the selected plants. In addition, the fractions of free metal ions in soil solution were determined using the Donnan membrane technique. After plant growth soil solution pH increased by 0.2-1.4 units and dissolved organic carbon (DOC) increased by 1-99 mg L^-1 in all soils examined. Soluble Cd and Zn decreased after Indian mustard growth in all soils examined, and this was attributed to increases in soil solution pH (by 0.9 units) after plant growth. Concentrations of soluble Cu and Pb decreased in acidic soils but increased in alkaline soils. This discrepancy was likely due to a competitive effect between plant-induced pH and DOC changes on the magnitude of metal solubility. The fractions of free Cd and Zn ranged from 7.2% to 32% and 6.4% to 73%, respectively, and they generally decreased as pH and DOC increased after plant growth. Metal uptake by plants was dependant on the soil solution metal concentration, which was governed by changes in pH and DOC induced by plant exudates, rather than on the total metal concentrations. Although plant uptake also varied with metal and soil types, overall soluble metal concentrations in the rhizosphere were mainly influenced by root-induced changes in pH and DOC which subsequently affected the metal uptake by plants.展开更多

Methods: TIP was dissolved in 0.1 M NaOH and added to the culture medium at a final concentrations of 50 mg/L, 100 mg/L and 200 mg/L. Some cells (control) were exposed to 0.001 M NaOH. TIP was added to PC12 cells 30 min prior to the administration of MPP^+. TIP and MPP^+ remained in the culture medium for 96 h. D2 dopamine receptor mRNA, preproenkephalin mRNA and prodynorphin mRNA expressions were assayed by real-time quantitative reverse transcription-PCR. Results: The D2 dopamine receptor mRNA and preproenkephalin mRNA expressions were up-regulated in MPP^+ group compared with the control group, and prodynorphin mRNA expression was down-regulated in that. The D2 dopamine receptor mRNA expression being down-regulated and prodynorphin mRNA expression being up-regulated in TIP group compared with the MPP^+ group. And there was no effect of TIP on preproenkephalin gene expression in PC12 cells induced by MPP^+. Conclusion: The results suggest that TIP down-regulates the D2 dopamine receptor mRNA expression, up-regulates prodynorphin mRNA expression and not affects preproenkephalin gene expression in PC12 cells induced by MPP^+.展开更多

[Method] A field experiment was carried out to determine the changes in yielc components, yield, contents of anthocyanin and starch, activities of phenylalanine ammonia-lyase (PAL) and adenosine 5-diphosphate glucose pyrophosphorylase (AG- Pase) in two genotypes of PFS (Ipomoea batatas L., var. 'Ayamurasaki' and 'Jishu18'). [Result] The application of COR significantly increased starch and antho- cyanin content in storage roots of Jishu18 across developmental stages by inducing the activities of PAL and AGPase, and finally enhanced yield by promoting fresh weight of storage roots. Ayamurasaki was insensitive to treatment with COR al- though its PAL activity temporally increased. The starch and anthocyanin content of Aya, and the anthocyanin content of Jishu18 increased progressively across devel- opmental stages with or without COR application, but the starch content of Jishu18 increased initially, then decreased before increasing again without application of COR. Treatment with COR reduced downward trend of starch accumulation in Jishu18. Thus, the effect of COR on accumulation of anthocyanin and starch in storage roots of PFS differs according to genotypes. [Conclusion] The application of 0.05 μmol/L COR may increase starch and anthocyanin content in PFS genotypes with lower starch and anthocyanin content in storage roots.展开更多

Formation of nodular outgrowth on the roots of wheat seedlings treated with 2, 4-D commenced in 8-10 days. The maximum Para-nodule formation was found in 1 ppm of 2, 4-D when incubated with bradyrhizobium P-132. Inoculation of the same has helped to increase the number of Para-nodule, but not essential for Para-nodulation. Histological study shows that, these induced Para-nodules originated from the pericycle and these are appeared to be modified lateral roots and Para-nodule structure formations however, it enhanced by bradyrhizobial inoculation.展开更多

METHODS: The colonic myeloperoxidase (MPO) activity and tumor necro...AIM: To investigate proteomic changes in spinal cord and dorsal root ganglia (DRG) of rats with trinitrobenzene sulfonic acid (TNBS)-induced colitis. METHODS: The colonic myeloperoxidase (MPO) activity and tumor necrosis factor-(TNF- ) level were determined. A two-dimensional electrophoresis (2-DE)-based proteomic technique was used to profile the global protein expression changes in the DRG and spinal cord of the rats with acute colitis induced by intracolonic injection of TNBS. RESULTS: TNBS group showed significantly elevated colonic MPO activity and increased TNF-level. The proteins derived from lumbosacral enlargement of the spinal cord and DRG were resolved by 2-DE; and 26 and 19 proteins that displayed significantly different expression levels in the DRG and spinal cord were identified respectively. Altered proteins were found to be involved in a number of biological functions, such as inflammation/immunity, cell signaling, redox regulation, sulfate transport and cellular metabolism. The over-expression of the protein similar to potassium channel tetramerisation domain containing protein 12 (Kctd 12) and low expression of proteasome subunit type-1 (psma) were validated by Western blotting analysis. CONCLUSION: TNBS-induced colitis has a profound impact on protein profiling in the nervous system. This result helps understand the neurological pathogenesis of inflammatory bowel disease.展开更多
