研究了用HPLC分析抗氧剂1520(2,4-二(正辛硫基亚甲基)-6-甲基苯酚)含量的方法。采用Waters 2695-2996型高效液相色谱仪、Waters SunFireTMC18色谱柱,以甲醇和水混合溶剂(体积比95∶5)做流动相,流速1.0mL/min,紫外检测波长285nm的条件时... 研究了用HPLC分析抗氧剂1520(2,4-二(正辛硫基亚甲基)-6-甲基苯酚)含量的方法。采用Waters 2695-2996型高效液相色谱仪、Waters SunFireTMC18色谱柱,以甲醇和水混合溶剂(体积比95∶5)做流动相,流速1.0mL/min,紫外检测波长285nm的条件时,样品中的各组分达到较好的分离效果。2,4-二(正辛硫基亚甲基)-6-甲基苯酚质量浓度在0.2~2.0mg/mL范围内与色谱峰面积(A)呈线性关系,线性回归方程为A=140.17c-28.74,相关系数R2=0.995 4。采用外标法单点校正峰面积定量,对抗氧剂1520标准溶液测定的回收率为99.96%~100.24%。 展开更多

The new indicator arises from the temperature dependence of kinetic constant. The new indicator was applied to data of observation based on differential optical absorption spectroscopy system on the outskirts of Hefei, China. Effect of Quercetin on CYP1A2, CYP2E1, CYP3A2 Activities and its Inhibitory Mechanism Studies in Rat Liver Microsomes 被引量:6

Aim To assess the potential effect of quercetin (QU), an natural plant estrogen, on CYP1A2, CYP2E1, and CYP3A2 activities in rat liver microsomes; and to identify the magnitude of inhibitory effect and the probable inhibitory mechanism of QU. Methods QU and specific substrate were concurrently incubated, with HPLC detection of the substrate metabolites for data analysis. The magnitude of inhibitory effect of QU on CYP3A2 was compared with those of ketoconazole (Ket) and erythromycin (Ery). The mechanism of its inhibitory effect on CYP3A2 and CYP2E1 was derived from Lineweaver-Burk plots. Results HPLC methods were in good linear relationship with r〉0.999 1. Relative standard deviations for intra-day and inter-day were〈8.4%. Recovery of each analyte in the concentrations studied was between 91.1% and 107.6 %. QU (up to 8 μmol·L^-1) showed potent induction to CYP1A2 (338.1% of the negative control)while inhibited CYP2E1 (49.2% of the negative control) and CYP3A2 (60.3% of the negative control) activity. The magnitude of inhibitory effect for QU on CYP3A2 was between those for Ket and Ery (Ket〉QU〉Ery). QU exhibited competitive inhibition of CYP3A2 dextromethorphan N-demethylation reaction and expressed noncompetitive inhibition of CYP2E1 chlorzoxazone-6-hydroxylation reaction. Conclusion HPLC assay has been validated with precision and accuracy. QU is an effective inhibitor of several CYP isoforms. It may cause relevant drug-drug interactions with CYP3A substrates. As a plant flavonoid, QU has potential not only in molecular advantage but also in CYP450 module capability for further application in cancer chemotherapy. 展开更多

For the production of reactive polyurethane cross-linkinger and curing agents, 2, 4-diisocyanate toluene (TDI) terpolymer, which possesses the rigid structures of hexatomic ring and three reactive functional groups, was synthesized and characterized by the Fourier transform infrared (FFIR), the gel permeation chromatography (GPC) and the chemical analysis methods. The reaction conditions were studied and optimized. A tracking research on the polymerization process of TDI was taken by using the GPC. The formation processes of the terpolymer, oligomers and higher-polymers were also dealt with. Results show that the TDI terpolymer can be prepared in the presence of Cat-3 catalyst and at the reaction temperature of (60 ±2)℃. The reaction time is short, its outcomes have narrow molecular weights distribution, namely molecular weights from 530 to 550, Mw/Mn =1.10, and the mass fraction of NCO is (25. 0 ± 0. 5)%. With the reaction time prolonging, however, TDI can be further higher-polymedzed to form higher-polymers. Benzoyl chloride (0. 4%, mass fraction), as the stabilizing agent, can effectively inhibit the occurrence of higher-polymerization. Aim To establish a reversed-phase liquid chromatographic (LC) method forsimultaneous determination of tetramethylpyrazine (TMP) and aspirin in a new compound formulation.Methods Chromatographic separation of the two drugs was achieved on a Diamonsil C_(18) column, usinga binary mixture of methanol-1.5% acetic acid (35:65, V/V, pH = 3.1) as mobile phase at a flow rateof 1.0 mL·min^(-1). Results Separation was completed in less than 12 min. Benzoic acid was used asthe internal standard. Recoveries at levels corresponding to 80 % to 120 % of the label claim ofthe formulation ranged from 99.6 to 100.3 % for aspirin and from 99.9 to 101.3% for TMP. The linearrange was 12.6 - 150.9 μg·mL^(-1)(r= 0.9997, n = 5) for aspirin and 25.0- 300.0 μg·mL^(-1) (r =0.9999, n = 5) for TMP. Conclusion The method developed can be used for the simultaneousdetermination of TMP and aspirin in pharmaceutical preparations. 展开更多

MethodsHPLC-DAD method was used to determine the constituents in preparations. Diamonsil? C_(18) (200mm X 4.6 mm, 5 μm) was used as analytical column, and acetonitrile/KH_2PO_4 was used as mobilephase with gradient elu-tion. The column temperature was at 24 ℃. The HPLC profile of chemicalconstituents of control sample and preparations were analyzed using similarity software. Results Thefingerprints of different preparations from different companies were slightly different because ofthe different preparing procedures. Mean while, the fingerprints of different batches of the samepreparation from the same company were similar to each other and the technology of each preparationwas stable. Conclusion This method is accurate, reproducible , simple, and can be used as ananalytical method for the routine quality control of Ginkgo biloba preparations. 展开更多

为观察牦牛主要寄生虫病高效低残留防治技术防治示范应用效果,选择1.5岁感染线虫和部分外寄生虫的牦牛40头,应用埃谱利诺菌素注射剂,按0.2 mg/kg体重剂量进行牦牛线虫病及外寄生虫病规模防治技术示范,设埃谱利诺菌素注射剂防治组和未防... 为观察牦牛主要寄生虫病高效低残留防治技术防治示范应用效果,选择1.5岁感染线虫和部分外寄生虫的牦牛40头,应用埃谱利诺菌素注射剂,按0.2 mg/kg体重剂量进行牦牛线虫病及外寄生虫病规模防治技术示范,设埃谱利诺菌素注射剂防治组和未防治的对照组,进行防治效果检查。结果:技术示范群牦牛防治后7、14、21、28、35d对牦牛消化道线虫虫卵转阴率、减少率均达100.0%,近中期防治效果佳;对牦牛原圆科线虫幼虫的转阴率分别为61.5%、92.3%、100.0%、100.0%和100.0%,减少率分别为67.2%、96.2%、100.0%、100.0%和100.0%,近期防治效果随时间的变化而逐步提高,在21d后均达100.0%。防治后146d对牦牛消化道线虫的虫卵转阴率、减少率分别达75.0%和88.7%;对牦牛原圆科线虫幼虫转阴率、减少率分别达90.0%和87.4%,远期防治效果显著。防治后第7d牛颚虱全部死亡干瘪,转阴率达100%,对牛颚虱有高效驱杀作用。结果表明应用本项技术防治牦牛主要寄生虫病高效安全低残留,能有效降低牦牛寄生虫病的危害,在冷季保膘、保重和提高牦牛成活率等方面效益显著,牧民乐于接受使用。 展开更多

Ultraviolet (UV) aging is one of the main factors which cause premature damage of asphalt pavements in the Tibetan Plateau, China. According to the measured levels of UV radiation, aging tests of styrene-butadiene rubber (SBR) asphalts with different contents of three anti-UV-aging agents including nano-TiO2, CeO2 and carbon black are performed. Common indices, which include retained penetration after thin film oven tests (TFOT) and softening point, and strategic highway research program (SHRP) indices of aged asphalts are evaluated. Infrared absorption spectral analysis is performed on asphalt specimens with 0. 8% carbon black which have been aged for different aging times (500, 1000 and 1 500 h). By grey incidence analysis, the optimal contents of anti-UV-aging agents are determined. The results show that TiO2 and CeO2 are not only good UV absorbing or shielding agents, but also strong oxidants. Carbon black is a good anti-UV-aging agent, and its optimal content is about 0. 8% of asphalt weight. UV aging of asphalt mainly occurs in the early stages of aging. The longer the aging time, the more severe the aging of asphalt. 展开更多
