介绍了红外光谱(IR)分析过程中常用的测试方法以及与之匹配的样品制备方法,总结了常用的样品制备方法的操作注意事项和优缺点。选取了5种常用的固相或液相的炼化原料及产品作为分析对象,比较采用不同的IR测试方法和样品制备方法所带来... 介绍了红外光谱(IR)分析过程中常用的测试方法以及与之匹配的样品制备方法,总结了常用的样品制备方法的操作注意事项和优缺点。选取了5种常用的固相或液相的炼化原料及产品作为分析对象,比较采用不同的IR测试方法和样品制备方法所带来的谱图结果上的差异,以寻求谱图质量更高、操作更简单、成本更低的样品制备方法。实验表明:衰减全反射(ATR)测得的IR谱图基线更为平整,能很好地保留高密度聚乙烯的原始结晶形态,满足对样品定性分析的需求;对于易吸湿的硬脂酸钙,石蜡油研磨法操作简便,能有效阻隔样品与水气的接触,有利于获得高质量的IR谱图;有挥发性的丙酮用液膜法制样,采用双片一次性KBr压片替代费用较高的KBr窗片,既减小了样品挥发对IR谱图质量的影响,又降低了分析成本,是很实用的液体制样改进方法。 展开更多

Nowadays,the internal structure of spacecraft has been increasingly complex.As its“lifeline”,cables require extensive manpower and resources for manual testing,and it is challenging to quickly and accurately locate quality problems and find solutions.To address this problem,a knowledge graph based method is employed to extract multi-source heterogeneous cable knowledge entities.The method utilizes the bidirectional encoder representations from transformers(BERT)network to embed word vectors into the input text,then extracts the contextual features of the input sequence through the bidirectional long short-term memory(BiLSTM)network,and finally inputs them into the conditional random field(CRF)network to predict entity categories.Simultaneously,by using the entities extracted by this model as the data layer,a knowledge graph based method has been constructed.Compared to other traditional extraction methods,the entity extraction method used in this study demonstrates significant improvements in metrics such as precision,recall and an F1 score.Ultimately,employing cable test data from a particular aerospace precision machining company,the study has constructed the knowledge graph based method in the field to achieve visualized queries and the traceability and localization of quality problems. 展开更多

MethodsHPLC-DAD method was used to determine the constituents in preparations. Diamonsil? C_(18) (200mm X 4.6 mm, 5 μm) was used as analytical column, and acetonitrile/KH_2PO_4 was used as mobilephase with gradient elu-tion. The column temperature was at 24 ℃. The HPLC profile of chemicalconstituents of control sample and preparations were analyzed using similarity software. Results Thefingerprints of different preparations from different companies were slightly different because ofthe different preparing procedures. Mean while, the fingerprints of different batches of the samepreparation from the same company were similar to each other and the technology of each preparationwas stable. Conclusion This method is accurate, reproducible , simple, and can be used as ananalytical method for the routine quality control of Ginkgo biloba preparations. 展开更多

Disulfide bond formation protein A (DsbA) is one of the important helper proteins for folding in protein synthesis in vivo. In this study, purification of recombinant DsbA was investigated by examining four important factors with Box-Behnken design method, a statistic-based design of experiments. The optimal operation conditions were obtained by adopting the effectiveness coefficient method on the multi-objective problem, which takes the protein recovery, purification efficiency and throughput of ion-exchange chromatography into account. After the optimization, protein recovery of 96.8% and purity higher than 95% DsbA was achieved, and the productivity was (377.9±1.7) mg soluble DsbA per liter broth. The purified protein was identified by peptide mass fingerprinting matching the record of gil2624856, a mutant of DsbA. The DsbA was preliminarily applied to the refolding of denatured lysozyme in vitro. 展开更多

AIM:To explore the method for early diagnosis of gastric cancer by screening the expression spectrum of saliva protein in gastric cancer patients using mass spectrometry for proteomics.METHODS:Proportional peptide mass fingerprints were obtained by analysis based on proteomics matrix-assisted laser desorption ionization time-of-flight/mass spectrometry.A diagnosis model was established using weak cation exchange magnetic beads to test saliva specimens from gastric cancer patients and healthy subjects.RESULTS:Significant differences were observed in the mass to charge ratio(m/z) peaks of four proteins(1472.78 Da,2936.49 Da,6556.81 Da and 7081.17 Da) between gastric cancer patients and healthy subjects.CONCLUSION:The finger print mass spectrum of saliva protein in patients with gastric cancer can be established using gastric cancer proteomics.A diagnostic model for distinguishing protein expression mass spectra of gastric cancer from non-gastric-cancer saliva can be established according to the different expression of proteins 1472.78 Da,2936.49 Da,6556.81 Da and 7081.17 Da.The method for early diagnosis of gastric cancer is of certain value for screening special biological markers. 展开更多

Contribution of Ground-Based Cloud Observation to Satellite-Based Cloud Discrimination

Therefore, cloud masking process is important to improve the quality of various satellite products. However, satellite-based cloud discrimination algorithm has been developing and efficient ground-based cloud observations are necessary to validate satellite-based cloud discrimination. The purpose of this study is to develop the efficient ground-based cloud observation methodology using whole sky camera. This paper deals with methods how to discriminate cloud portions on whole sky image, how to apply the ground-based cloud observation to the validations for satellite products. For the cloud discrimination on whole sky image, we propose SI (sky index) and BI (brightness index) calculated from RGB (red, green and blue) channels. SI shows the extent of the blueness and gray scale and BI indicates the extent of the brightness. Sun, cloud and blue sky portions are divided by SI and BI threshold. As an application of ground-based cloud observation for the validation of satellite products, clouds portions discriminated from whole sky image are projected onto ground surface with map coordinate. We also examine to compare with cloud portions on whole sky images and MODIS (MODerate resolution Imaging Spectroradiometer) image as one of experiments. The proposed ground-based cloud observation method and its extension to satellite-based cloud discrimination should be connected to improve the quality of satellite products. 展开更多

To establish two-dimensional electrophoresis profiles with high resolution and reproducibility from murine brain tissues by human cytomegalovirus(HCMV) infection and paired murine brain tissues and to identify the differential expression proteins. Methods: Forty Kunming mice were randomly divided into infection group (20) injected with HCMVAD169 and control group (20) injected with saline into their brain. After 30 days, the murine brain tissues by HCMV infection and paired murine brain tissues were separated by two-dimensional gel electrophoresis(2-DE), analyzed by Image Master 2D software, and identified by peptide mass fingerprint(PMF) and database searching, and make Western blotting analyses the differential expression of individual proteins. Results: Well resolved, reproducible 2-D maps of the above tissues were obtained. Some of the different proteins identified by mass spectrometry(MS) were matched in the SWISS-2D PAGE database, Western blotting analyses were further carried out to verify the differential expression of individual proteins. Conclusion: These data will be valuable for studying the diagnosis of disease at an early stage, mechanisms of pathogenic and the key to the development of anti-HCMV medicine. 展开更多

A simple, sensible and reliable HPLC-DAD fingerprint analysis method for the raw materials of Oxytropisfalcata and Oxytropis chiliophylla, both of which were used as "Er-Da-Xia" in Tibetan medicines, was developed and then subsequently applied to analyze samples collected from different locations or times. 19 common fingerprint peaks for O. falcata, 24 for O. chiliophylla, and 11 for the two herbs were designated respectively, including 7 identified characteristic peaks existing in both herbs and 1 uniquely presenting in O. chiliophylla. Although there were some slight differences in the chemicals of O. falcata and O. chiliophylla, the main components of both herbs were consistent generally. The results provided scientific basis, at least from the chemical point of view, for the reasonablity of two herbs being used as the same drug in Tibetan medicines and for the necessary of further investigation on their detailed chemical and pharmacological differences. 展开更多

A simple and accurate high-performance liquid chromatography(HPLC)method to generate the fingerprints of crude Pu-erh tea(CPT)and ripened Pu-erh tea(RPT)is described.A suitable chromatographic system was established using a linear gradient elution with acetonitrile and water containing 0.6% formic acid as the mobile phase and a detection wavelength of 300 nm.HPLC analysis identified 24 common peaks among RPT and 21 common peaks among CPT.This study revealed that crude Pu-erh tea and ripened Pu-erh tea contained some similar major constituents,but with distinctive peaks,which may be caused by the difference in the fermentation process.This HPLC fingerprint method could be used to evaluate and authenticate crude Pu-erh tea and ripened Pu-erh tea. 展开更多

Homoisoflavonoids and steroidal saponins were determined simultaneously in a single run. A total of 27 Ophiopogonis Radix samples were analyzed, and 18 reference substances were used for the identification of the common peaks. The fingerprint was further analyzed by chemometrics methods including similarity analysis (SA), hierarchical clustering analysis (HCA) and principal component analysis (PCA). The results indicated that the combination of chromatographic fingerprint and chemometrics analysis could be used for the geographical differentiation and quality evaluation of Ophiopognnis Radix. 展开更多

The root of Hedysarum multijugum(RHM) is recorded as a folk herbal medicine in China and is sometimes used as a substitute for Hedysari Radix, which is a famous traditional Chinese medicine derived from the roots of Hedysarum polybotrys. In the present study, a sensible, reliable, and reproducible HPLC-DAD fingerprint analysis method for RHM was developed and then subsequently applied to analyze RHM samples from different origins. The chemical constituents of the RHM samples were generally consistent, although it was slightly affected by the local environment of the plant. In addition, the chemical constituency of RHM was shown to be significantly different from that of Hedysari Radix, suggesting that RHM is not suitable as a substitute for Hedysari Radix, at least from the chemical point of view. 展开更多

A suitable ehro- matographic system was established using a linear gradient elution with acetonitrile and 0.1% phosphoric acid (H3PO4) as the mobile phase and a detection wavelength of 210 nm. HPLC analysis identified 12 common peaks and 8 common fingerprint peaks among the different parts of the herb. We found that the constituents varied at different harvesting times and in different parts of the plant. The HPLC fingerprint method could be used to evaluate the quality ofM. suaveolens Ledeb and Melilotus genus herbs. The same strategy can also be applied for the development of fingerprints for the quality control of other herbal medicines. 展开更多

The rhizome of Alpinia officinarum Hance is a well-known traditional Chinese medicine(TCM)and has been widely used for the remedy of gastrointestinal diseases.In the present study,a simple and rapid HPLC-DAD was developed for the quality control of the rhizomes of A.officinarum.Its chemical fingerprint was established using raw material of 15 different origins in China.Similarity analysis(SA)and hierarchical clusting analysis(HCA)were applied to select the qualitative markers.Principal components analysis(PCA)was conducted to select the quantitative markers of the rhizomes of A.officinarum samples from different origins.The constituents were confirmed by(+)electrospray ionization LC-MS.12 constituents were selected as common peaks and 10 of them were confirmed by(+)electrospray ionization LC-MS.Six bioactive constituents including DPHA,galangin flavanone,galangin,galangin 3-methylether,DPHB and DPHC were simultaneous determination by using the HPLC-DAD analysis.The developed method was able to determine the bioactive components with excellent resolution,precision and recovery.The results indicated that chromatographic fingerprint combination with multi-components determination method is suitable for quality assessment of the rhizomes of A.officinarum. 展开更多
