目的:通过观察鹰嘴豆异黄酮对糖尿病小鼠血糖及氧化-抗氧化效应的影响,探讨其降糖机制。方法:实验于2006-08/10在中国药科大学药理教研室动物实验中心完成。①选用清洁级昆明种小白鼠50只进行造模:小鼠以高糖高脂饲料喂养4周后,按25mg/k... 目的:通过观察鹰嘴豆异黄酮对糖尿病小鼠血糖及氧化-抗氧化效应的影响,探讨其降糖机制。方法:实验于2006-08/10在中国药科大学药理教研室动物实验中心完成。①选用清洁级昆明种小白鼠50只进行造模:小鼠以高糖高脂饲料喂养4周后,按25mg/kg剂量一次尾静脉注射链脲佐菌素,注射7d后血糖升高稳定、且随机血糖达16.7mmol/L以上者为造模成功。②随机将造模成功的小鼠50只均分为5组:高血糖模型对照组、二甲双胍治疗组、鹰嘴豆异黄酮提取物低、中、高剂量组,每组10只。在各组给予高糖高脂饲料的基础上,鹰嘴豆异黄酮提取物3个剂量组分别给予25mg/kg、50mg/kg、100mg/kg的受试物,高血糖模型对照组和二甲双胍治疗组分别给予150mg/kg的蒸馏水和二甲双胍,1次/d,连续4周。③给药2周末和4周末监测空腹血糖,实验结束时于空腹12h后将小鼠处死,取血和肝脏组织,测定血清及肝组织中丙二醛含量及超氧化物歧化酶、过氧化氢酶、谷胱甘肽过氧化物酶的活性。结果:纳入50只小鼠均进入结果分析。①空腹血糖:给予鹰嘴豆异黄酮提取物2周后,高剂量组与二甲双胍治疗组小鼠血糖值显著低于高血糖模型对照组(P<0.05),且二者间差异无显著性;4周后,高、中剂量组均显著低于高血糖模型对照组(P<0.05),二甲双胍治疗组极显著低于高血糖模型对照组(P<0.01);高、中剂量组与二甲双胍治疗组间差异无显著性。②丙二醛含量:与高血糖模型对照组相比,中剂量组和二甲双胍治疗组血清和肝脏中丙二醛含量均显著降低(P<0.05),高剂量组呈极显著降低(P<0.01),且高剂量组血清中丙二醛含量显著低于二甲双胍治疗组(P<0.05)。③超氧化物歧化酶活性:中、高剂量组和二甲双胍治疗组血清和肝脏中超氧化物歧化酶活性均高于高血糖模型对照组(P<0.05)。④过氧化氢酶活性:中、高剂量组血清和高剂量组肝脏中过氧化氢酶活性高于高血糖模型对照组(P<0.05,P<0.01,P<0.05),高剂量组血清和肝脏中过氧化氢酶活性均显著高于二甲双胍治疗组(P<0.05),而二甲双胍治疗组无论血清还是肝脏中过氧化氢酶活性与高血糖模型对照组相比差异无显著性(P>0.05)。⑤谷胱甘肽过氧化物酶活性:中、高剂量组血清中谷胱甘肽过氧化物酶活性均高于高血糖模型对照组(P<0.01);各剂量组肝脏组织中谷胱甘肽过氧化物酶活性均高于高血糖模型对照组(P<0.01);二甲双胍治疗组血清和肝脏中谷胱甘肽过氧化物酶活性均显著高于高血糖模型对照组(P<0.05),但肝脏中活性仍极显著低于3个剂量组(P<0.01)。⑥给药后,模型鼠的血糖与血清超氧化物歧化酶、过氧化氢酶、谷胱甘肽过氧化物酶活性的相关系数分别为-0.9403、-0.9466和-0.9503;与肝脏超氧化物歧化酶、过氧化氢酶、谷胱甘肽过氧化物酶活性的相关系数分别为-0.9436、-0.9356和-0.9631。结论:①鹰嘴豆异黄酮提取物有显著的降血糖作用。②鹰嘴豆异黄酮提取物降血糖作用的机制可能与其提高抗氧自由基酶类的活性,保护机体组织免遭过氧化损伤有关。③鹰嘴豆异黄酮提取物在降低过氧化产物丙二醛含量、增强机体抗氧化能力方面的功能优于治疗糖尿病的常用药物二甲双胍。 展开更多

目的:探讨大豆异黄酮对膳食诱导的肥胖大鼠低度炎症介质水平的影响,以初步阐明大豆异黄酮改善肥胖性胰岛素抵抗状态的可能机制。方法:选用高脂饲料诱导的雄性SD肥胖大鼠,随机分为模型对照组和3个不同剂量大豆异黄酮组,同时另设基础饲料... 目的:探讨大豆异黄酮对膳食诱导的肥胖大鼠低度炎症介质水平的影响,以初步阐明大豆异黄酮改善肥胖性胰岛素抵抗状态的可能机制。方法:选用高脂饲料诱导的雄性SD肥胖大鼠,随机分为模型对照组和3个不同剂量大豆异黄酮组,同时另设基础饲料对照组。各组给予相应受试物1月,禁食过夜后股动脉采血处死各大鼠,分离肾周及睾周白色脂肪。以酶法检测各组动物空腹血糖,放射免疫法检测空腹血胰岛素、白介素-6及肿瘤坏死因子,酶联免疫法检测血清C-反应蛋白、抵抗素与脂联素含量。结果:与模型对照组比较:150mg/kg与450mg/kg组能明显降低大鼠空腹体质量、内脏脂肪沉积、血清胰岛素、白介素-6、肿瘤坏死因子-α含量,提高脂联素水平;450mg/kg组能明显降低血清抵抗素的含量;不同剂量大豆异黄酮组的C-反应蛋白水平与模型组比较无明显差异。结论:大豆异黄酮可能是通过减少大鼠体内脂肪沉积、调整脂肪源的低度炎症介质而改善肥胖大鼠胰岛素抵抗状态。 展开更多

采用酶动力学方法研究了鹰嘴豆异黄酮对酪氨酸酶单酚酶和二酚酶活力的抑制动力学效应。结果表明,鹰嘴豆异黄酮对酪氨酸酶单酚酶和二酚酶活性均有抑制作用,使单酚酶和二酚酶活力下降50%的鹰嘴豆异黄酮质量浓度(IC_(50))分别为0.07 mg/mL... 采用酶动力学方法研究了鹰嘴豆异黄酮对酪氨酸酶单酚酶和二酚酶活力的抑制动力学效应。结果表明,鹰嘴豆异黄酮对酪氨酸酶单酚酶和二酚酶活性均有抑制作用,使单酚酶和二酚酶活力下降50%的鹰嘴豆异黄酮质量浓度(IC_(50))分别为0.07 mg/mL和0.03 mg/mL。在降低酪氨酸酶酶活的同时,鹰嘴豆异黄酮能明显延长单酚酶的迟滞时间,质量浓度为0.08 mg/mL的鹰嘴豆异黄酮能使其迟滞时间由1.26 min延长至2.62 min。Lineweaver-Burk图显示鹰嘴豆异黄酮对二酚酶的抑制作用表现为可逆的竞争性抑制。 展开更多

以鹰嘴豆为原料,采用超声波联合酶解法提取鹰嘴豆异黄酮。通过单因素实验(因素包括纤维素酶添加量、超声温度、超声时间、酶解料液比、乙醇浓度和超声功率)确定以酶添加量、超声温度、超声时间为自变量,采用响应面法分析各因素对鹰嘴豆... 以鹰嘴豆为原料,采用超声波联合酶解法提取鹰嘴豆异黄酮。通过单因素实验(因素包括纤维素酶添加量、超声温度、超声时间、酶解料液比、乙醇浓度和超声功率)确定以酶添加量、超声温度、超声时间为自变量,采用响应面法分析各因素对鹰嘴豆异黄酮提取量的影响,确定最佳提取工艺。结果表明:酶添加量306 U/g,乙醇浓度35%,液料比30∶1,55℃超声(功率为80 W)处理45 min为最佳条件,在此条件下每克鹰嘴豆异黄酮的提取量为1.32 mg,和预测理论值一致。与传统方法相比,采用超声波联合酶解法显著提高了鹰嘴豆异黄酮的提取效率,且该提取工艺操作简便、安全环保。 展开更多

大豆异黄酮对大鼠局灶性脑缺血再灌注损伤的保护作用及其机制研究 被引量:7

目的:研究大豆异黄酮(Soybean Isoflavone,SI)对大鼠局灶性脑缺血再灌注损伤的保护作用,并探讨其作用机制。方法:采用线栓法制备局灶性脑缺血再灌注大鼠模型,选取120只随机分为6组:局灶性脑缺血再灌注模型组、大豆异黄酮(20、40、80和16... 目的:研究大豆异黄酮(Soybean Isoflavone,SI)对大鼠局灶性脑缺血再灌注损伤的保护作用,并探讨其作用机制。方法:采用线栓法制备局灶性脑缺血再灌注大鼠模型,选取120只随机分为6组:局灶性脑缺血再灌注模型组、大豆异黄酮(20、40、80和160 mg/kg)腹腔注射预处理组和舒血宁注射液(4mg/kg)阳性对照组,并设假手术组。再灌注6h后,采用盲法进行神经功能评分,测定脑梗死体积(IV%)及脑组织含水量;通过HE染色法观察脑组织病理形态学改变,通过TUNEL染色观察神经细胞凋亡状况并计算凋亡指数(Apoptosis Index,AI);通过生化分析仪测定血清中肌酸激酶(CK)、乳酸脱氢酶(LDH)含量;检测脑组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量。结果:与假手术组相比,局灶性脑缺血再灌注模型组大鼠出现明显的行为障碍,脑梗死体积(IV%)和脑组织含水量显著升高,脑组织出现明显的病理性改变,神经细胞凋亡状况加重,凋亡指数显著升高;血清中CK,LDH含量显著升高,脑组织中SOD、GSH-Px、CAT活性显著降低且MDA含量显著升高,差异均具有统计学意义。与局灶性脑缺血再灌注模型组相比,大豆异黄酮(40、80和160 mg/kg)组大鼠行为障碍明显好转,大豆异黄酮(80和160 mg/kg)组脑梗死体积和含水量均显著降低;大豆异黄酮预处理组大鼠脑组织病变和神经细胞凋亡均明显减轻,以160 mg/kg组效果最为显著,大豆异黄酮(80和160 mg/kg)组凋亡指数显著降低;大豆异黄酮(40、80和160 mg/kg)组大鼠血清中CK,LDH含量和脑组织中MDA含量均显著降低;大豆异黄酮(80和160 mg/kg)组脑组织中SOD、CAT活性显著降低,且160mg/kg组GSH-Px活性显著降低。结论:大豆异黄酮能够有效改善局灶性脑缺血大鼠神经功能障碍、降低脑梗死体积和脑组织含水量、改善脑组织病变和神经细胞凋亡状况,提示大豆异黄酮对大鼠局灶性脑缺血再灌注损伤具有剂量依赖性的保护作用,其作用机制可能与大豆异黄酮能够有效改善抗氧化酶活性、提高机体清除自由基能力、抑制氧化应激损伤有关。 展开更多

目的探讨鹰嘴豆异黄酮提取物(CIE)对阿尔茨海默病(AD)炎症模型大鼠记忆能力的影响,并探讨其可能的作用机制。方法采用脂多糖右侧海马注射制备AD模型,40只Wistar大鼠随机分为CIE高、中、低剂量组(100,50,25 mg·kg-1)、模型组和对照... 目的探讨鹰嘴豆异黄酮提取物(CIE)对阿尔茨海默病(AD)炎症模型大鼠记忆能力的影响,并探讨其可能的作用机制。方法采用脂多糖右侧海马注射制备AD模型,40只Wistar大鼠随机分为CIE高、中、低剂量组(100,50,25 mg·kg-1)、模型组和对照组(生理盐水),灌胃给药28 d后,采用Y-迷宫测试大鼠的学习记忆能力,采用免疫组化法检测大鼠海马内肿瘤坏死因子-α(TNF-α)和细胞白介素-6(IL-6)的表达。结果与对照组比较,模型组大鼠学习记忆能力明显下降,尝试次数均明显增加(P<0.01),TNF-α和IL-6的表达水平有显著提高(P<0.01);经过灌胃给药不同剂量的CIE后,与模型组相比,大鼠学习记忆的尝试次数和TNF-α、IL-6的表达水平有不同程度的降低,并呈现一定的剂量依赖性,特别在中、高剂量组差异具有统计学意义(P<0.05或P<0.01)。结论 CIE对AD模型大鼠的学习记忆能力有明显的改善作用,其作用机制可能与抑制模型大鼠海马组织内TNF-α和IL-6的表达有关,进而抑制海马神经元的炎症反应。 展开更多

[Objective]The aim of this study was to set up a high performance liquid chromatography for rapid determination of isoflavones from soybean and analyze the correlation between isofalvone content and protein or fat content. [Method]The isoflavones were firstly extracted by 80% methanol and then hydrolyzed at 100 ℃. The chromatographic separation adopted a reversed-phase C18 analytical column with binary high-pressure gradient elution,while its analysis time was 25 min and column temperature was 40 ℃. The diode array detector was used for monitoring with wavelength of 260 nm. The correlation between isofalvone content and protein or fat content was analyzed by data processing system Origin 6.0. [Result]The high performance liquid chromatograph for determination of isoflavones from soybean was verified to be accurate and reliable by methodology. The isoflavones of 85 soybean lines cultivated in Jilin Province were determined,and the results primarily showed the characters and ranges of isoflavones from soybean lines cultivated in Jilin Province,while the isoflavone content of soybeans ranged from 2.29 to 4.89 mg/g,and the average content was 3.36 mg/g. The isoflavone content of 5 soybean lines exceeded 4 mg/g,while there was a remarkably negative correlation between isoflavone content and protein content,and there was no significant positive correlation between isoflavone content and fat content. [Conclusion]The isoflavone content of soybean lines cultivated in Jilin Province is higher,so it is feasible for breeding the soybean lines with high isoflavone content and fat contetnt. 展开更多

An 8-week feeding trial was conducted to investigate the effects of dietary soy isoflavones on feeding intake,growth performance,and digestion of juvenile Japanese flounder(Paralichthys olivaceus).Four isonitrogenous(49% crude protein) and isoenergetic(20.1 MJ kg-1) diets were formulated to contain four graded levels of soy isoflavones,namely,0,1,4 and 8 g soy isoflavones in 1 kg of diet.Each diet was randomly fed to triplicate tanks of fish(Initial average weight:2.58 g ± 0.01 g),and each tank was stocked with 35 fish.No significant difference was observed among diets with levels of 0,1 and 4 g kg-1 soy isoflavones in feed intake,weight gain,feed efficiency ratio(FER),proximate composition of fish whole body and apparent digestibility coefficients(ADC) of nutrients and energy(P>0.05).However,high dietary soy isoflavones level(8 g kg-1) significantly depressed weight gain,FER,whole-body crude lipid content of fish and ADC of nutrients(P<0.05).These results indicate that high level of dietary soy isoflavones(above 4 g kg-1) significantly depresses growth responses and FER of Japanese flounder.However,as the content of soy isoflavones in soybean meal is around 1 to 3 g kg-1,the adverse effects might be neglected when soybean products are used as a fish feed ingredient. 展开更多

Objective: To investigate the urinary soy isoflavone metabolites from women, female piglets and rats fed with diet containing soy protein. Methods: Urinary samples from human and animals were collected after soy diet consumption. Identification for soy isoflavone metabolites in urine samples was processed using an Agilent Bruker LC Esquire ion trap system. Quantification of aglycone and conjugated soy isoflavone metabolites were also analyzed using a method published previously. Results: Identification studies showed that aglycones and conjugates of soy isoflavone metabolites were found in women and porcine samples. Interestingly, glucuronide conjugate of equol, besides glucuronide conjugates of genistein and daidzein, were found in rat urine. Glucuronide conjugate of equol was the major metabolite found in rat urine. A quantitative study showed that conjugated forms of isoflavones were more than 90% in woman urine, were between 80.5% and 84.5% in female porcine urine, and were less than 50% in female rat urine. Conclusion: Equol is the major metabolite found in female rat urine, but it is not found in woman or female porcine urine. Urinary profiles show that porcine model is more appropriate for mimicking human soy diet consuming studies. 展开更多

In this paper, we proposed a novel resolution criterion(improved calibrated normalized resolution product, r*') to evaluate separation quality of fingerprints. By comparing with the calibrated normalized resolution product(r*) and the hierarchical chromatographic response function(HCRF), the validity of this criterion was demonstrated by experimental chromatograms. The soy isoflavone extract was selected as the analytical object. The initial and end percentages of methanol and elution time affecting gradient elution were tested by orthogonal design. The final optimized conditions were as follows. It was detected by UV absorbance at 254 nm, column temperature was maintained at 36 oC, solvent A was 0.1%(v/v) acetic acid, solvent B was methanol, gradient elution was from 34% to 65% B in a linear gradient in 25 min, and the flow-rate was set at 1.0 m L/min. In addition, the main ingredients of the soy isoflavone extract were confirmed by LC-ESI/MS. 展开更多

Soybean isoflavones have been one of the potential preventive candidates for antitumor research in recent years. In this paper, we first studied the transformation of soybean isoflavones with the homogenized slurry of Ganoderma lucidum. The resultant transformed products(TSI) contained(703.21±4.35) mg/g of genistein, with transformed rates of 96.63% and 87.82% of daidzein and genistein, respectively, and TSI also could enrich the bioactive metabolites of G. lucidum. The antitumor effects of TSI on human colorectal cancer cell line HTL-9, human breast cancer cell line MCF-7, and human immortalized gastric epithelial cell line GES-1 were also studied. The 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide(MTT) assay showed that TSI could dramatically reduce the viability rates of HTL-9 cells and MCF-7 cells without detectable cytotoxicity on GES-1 normal cells when the TSI concentration was lower than 100 μg/ml. With 100 μg/ml of TSI, HTL-9 cells were arrested in the G1 phase, and late-apoptosis was primarily induced, accompanied with partial early-apoptosis. TSI could induce primarily earlyapoptosis by arresting cells in the G1 phase of MCF-7 cells. For HTL-9 cells, Western-blot and reverse-transcriptase polymerase chain reaction(RT-PCR) analysis showed that TSI(100 μg/ml) can up-regulate the expression of Bax, Caspase-3, Caspase-8, and cytochrome c(Cyto-c), indicating that TSI could induce cell apoptosis mainly through the mitochondrial pathway. In addition, the expression of p53 was up-regulated, while the expression of Survivin and nuclear factor κB(NF-κB) was down-regulated. All these results showed that TSI could induce apoptosis of HTL-9 cells by the regulation of multiple apoptosis-related genes. 展开更多

To investigate the effect of glycitein, a synthetic soybean isoflavone(ISF), on the intestinal antioxidant capacity, morphology, and cytokine content in young piglets fed oxidized fish oil, 72 4-d-old male piglets were assigned to three treatments. The control group was fed a basal diet containing fresh fish oil, and the other two groups received the same diet except for the substitution with the same dosage of oxidized fish oil alone or with ISF(oxidized fish oil plus ISF). After 21 d of feeding, supplementation of oxidized fish oil increased the levels of malondialdehyde(MDA), oxidized glutathione(GSSG), interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α), interleukin-2(IL-2), nuclear factor κ B(NF-κB), inducible nitric oxide synthase(iN OS), NO, and Caspase-3 in jejunal mucosa, and decreased the villous height in duodenum and the levels of secretory immunoglobulin A(sI gA) and IL-4 in the jejunal mucosa compared with supplementation with fresh oil. The addition of oxidized fish oil plus ISF partially alleviated this negative effect. The addition of oxidized fish oil plus ISF increased the villous height and levels of sI gA and IL-4 in jejunal mucosa, but decreased the levels of IL-1β and IL-2 in jejunal mucosa(P0.05) compared with oxidized fish oil. Collectively, these results show that dietary supplementation of ISF could partly alleviate the negative effect of oxidized fish oil by improving the intestinal morphology as well as the antioxidant capacity and immune function in young piglets. 展开更多
