随着生物技术的发展,蛋白多肽类药物日益增多,对此类药物的定量检测及药代动力学研究变得越来越重要。传统生物技术药物分析方法主要是以配体结合分析(ligand-binding assay,LBA)为主,而液相色谱串联质谱(liquid chromatography tandem ...随着生物技术的发展,蛋白多肽类药物日益增多,对此类药物的定量检测及药代动力学研究变得越来越重要。传统生物技术药物分析方法主要是以配体结合分析(ligand-binding assay,LBA)为主,而液相色谱串联质谱(liquid chromatography tandem mass spectrometry,LC-MS/MS)技术由于兼具色谱的高分离能力和质谱的高选择性等优点,也已成为生物大分子药物定量分析的有力手段。综合上述2种方法的各自优势,LCMS/MS技术联用LBA技术进行样品前处理,不仅能降低背景的复杂性,还能避免交叉反应,可以大大提高方法的选择性和灵敏度。另外,使用肽水平的免疫捕获可进一步提高分析灵敏度。联合使用配体结合分析-液相色谱串联质谱(hybird LBA with LC-MS/MS,LBA-LC-MS/MS)技术的关键影响因素包括(:1)捕获试剂的亲和力;(2)酶切位点和效率;(3)指纹肽的选择;(4)内标的选择;(5)蛋白质和肽段的纯化;(6)色谱分离;(7)质谱条件的优化。本文综述了近年来研究蛋白多肽类药物药代动力学的各种分析方法,其中LC-MS/MS和毛细管电泳(capillary electrophoresis,CE)-MS联用技术可望成为药代动力学研究中较有前途的分析方法。展开更多
[ Objective] This study was to reveal the differences in crude fat and glucosinolates between self pollinated seeds and naturally pollinated seeds in Brassica napus in sichuan ecological region.. [ Method] Near-infrar...[ Objective] This study was to reveal the differences in crude fat and glucosinolates between self pollinated seeds and naturally pollinated seeds in Brassica napus in sichuan ecological region.. [ Method] Near-infrared spectroscopy method (NIRS) was employed to measure the quality components in self pollinated seeds and naturally pollinated seeds of 861 shares of Brassica napus from Sichuan ecological region. And correlation analysis and regression analysis were conducted based on the experimental data via SPSS (statistics package for social science). [ Result] The contents of crude fat in the self pollinated seeds were commonly a higher than that in the naturally pollinated seeds at 0.01 significant level; while the contents of glucosinolates in the self pollinated seeds and the naturally pollinated seeds were insignificantly different. Both the correlation relationship and linear regression for the crude fat between the self pollinated seeds and naturally pollinated seeds reached the significant level. The regression equations for the contents of crude fat(y1 ) and glucosinolates( y2 ) in the naturally pollinated seeds and of crude fat( x1 ) and glucosinolates( x2 ) in self pollinated seeds were respectively determined to be y1 = 16.844 +0.614x1 and y2 = -0.620 + 1.017 x2. [ Conclusion] In Brassica napus breeding, crude fat in naturally pollinated seeds should be emphatically taken into account, meanwhile concurrently considering that in self pollinated seeds; while glucosinolates in both the self pollinated seeds and the naturally pollinated seeds must be simultaneously concerned.展开更多
It is a common issue in the processing industry of crayfish that flavored crayfish stored at room temperature is perishable.In order to establish an effective putrid prediction mechanism,high-throughput sequencing and...It is a common issue in the processing industry of crayfish that flavored crayfish stored at room temperature is perishable.In order to establish an effective putrid prediction mechanism,high-throughput sequencing and solid phase microextraction gas chromatography-mass spectrometry(SPME-GCMS)were used to analyse the microbial community structure and volatile flavor compounds of normal and putrid crayfish.The results showed that Aeromonas(57%),Macrococcus(7.7%),Vibrio sp.(6.6%),Acinetobacter(5%),Citrobacter(4.9%)and Enterobacter(1.49%)were the main bacterial genus in the refrigerated fresh crayfish(HNA).And Staphylococcus(17.04%),Aeromonas(4.46%),Xanthomonas(4.16%),Streptococcus(4.62%)and Enterococcus(2.77%)were the main bacterial genus in the marinated and refrigerated crayfish(HND).With the spoilage of samples(HNE and HNC),the diversity of bacteria decreased,and the specific spoilage bacteria grew rapidly,mainly Enterococcus,Bacillus,Lactobacillus,Leuconostoc,Weissella.Meanwhile,the volatile compounds in non-spoilage sample(HNA and HND)were mainly alkane compounds,aldehydes compound and esters compounds;and the volatile compounds in spoilage samples were mainly alcohols,acids,benzene compounds,terpenoids,N-containing compounds,S-containing compounds and ethers.This indicated that the contents and types of volatile compounds changed with the sample spoilage and deterioration.Correlation analysis results showed that Enterococcus,Lactobacillus and Bacillus were significantly positively correlated with alcohols,acids,benzene,terpenoids,N-containing compounds,S-containing compounds and ether compounds,while Aeromonas,Megasphaera,Acinetobacter,Citrobacter and Vibrio were significantly positively correlated with alkane compounds and esters compounds,and Leuconostoc were significantly positively correlated with alcohol compounds.These results can provide a theoretical guidance for the storage of cooked flavor crayfish at room temperature.展开更多
