The current study comprehensively evaluates four different protein extraction methods based on urea,sodium dodecyl sulfate(SDS),anionic surfactants(BT),and total RNA extractor(Trizol),aiming to optimize the sample pre...The current study comprehensively evaluates four different protein extraction methods based on urea,sodium dodecyl sulfate(SDS),anionic surfactants(BT),and total RNA extractor(Trizol),aiming to optimize the sample preparation workflow for mass spectrometry-based proteomics.Using HeLa cells as an example,we found that the method employing the mass spectrometry-compatible surfactant BT reagent significantly reduces the total time consumed for protein extraction and minimizes protein losses during the sample preparation process.Further integrating the four protein extraction methods,we identified over 7000 proteins from HeLa cells without relying on pre-fractionation techniques,and 2990 of them were quantified using label-free quantification.It is worth noting that the BT and SDS methods demonstrate higher efficiency in extracting membrane proteins,while the Urea and Trizol methods are more effective in extracting proteins from nuclear and cytoplasmic fractions.In summary,this study provides a novel solution for deep proteome coverage,particularly in the context of cellular protein extraction,by integrating mass spectrometry-compatible surfactants with traditional extraction methods to effectively enhance protein identification numbers.展开更多

Based on the characteristics of the lignite sample, effects of assistant anthraquinone (AQ) on extraction yield of humic substances (HS) and the action mechanisms of AQ in alkaline condition were studied by Fouvie...Based on the characteristics of the lignite sample, effects of assistant anthraquinone (AQ) on extraction yield of humic substances (HS) and the action mechanisms of AQ in alkaline condition were studied by Fouvier transform infrared (FT-IR) spectroscopy. The results indicate that assistant AQ can not only increase the extraction yield of HS but also reduce the alkali dosage (NaOH) as well as the extraction temperature and extraction time. Under the optimal conditions of alkali dosage of 9%, AQ dosage of 0.75%, extraction temperature of 80 ℃, extraction time of 30 min, stirring speed of 600 r/min and solid-to-liquid ratio of 1:3, the extraction yield of HS reaches 80.08%, which is increased by more than 20% compared with the conventional extraction. FT-IR spectra show that AQ is able to prevent dissolved HS from being destroyed into undissolved substance by alkali and 1-IS obtained in the presence of AQ possesses more groups of COOR and --COOH than that obtained without AQ.展开更多

The compounds isolated from the methanol extract of Euphorbia retusa Forssk were identified as kaempferol-3-beta-D glucopyranosyl (1), 3-O-alpha-L-xylopyranosyl (1→3)-alpha-L xylopyranosyl (1→2), beta-D glucopyranosyllup 1,12-diene-3,21-diol (2) and 5,7,8,3',4'-pentahydroxy-3-methoxyflavone (3) and the compounds isolated from Euphorbia paralais L. identified as 3,5,3'-trihydroxy-6,7-d-methoxy-4' (7"-hydroxygeranyl-1"-ether) flavones (4), 3-O-syringic acid (1→4)-alpha-L- xylopyranosyl (1→3), beta-D-rahminopyranosyl hederagenin (5) and 3-O-syringic acid (1→4), beta-D-rahminopyranosyl (1→3),beta-D-rahminopyranosyl hederagenin (6). Then its extraction residue ...To investigate the structural features and provide an alternative method for high value-added utilization of coal, Lingwu coal was first extracted with organic solvent at room temperature. Then its extraction residue was oxidized in aqueous sodium hypochlorite(ASHC) under mild conditions. The effects of oxidation conditions, such as temperature, reaction time, the ratio of Lingwu coal extraction residue(LCER, g) to ASHC(m L) and p H value, on the product distributions and compositions were investigated. The results of gas chromatography/mass spectrometry(GC/MS) suggested that 53 kinds of methyl esterified products were detected in total, and benzene carboxylic acids were the main oxidation products, while chloro-substituted benzene carboxylic acids were the main by-products. Higher yield and fewer kinds of organic acids could be obtained at lower p H value, especially for the main objective product, benzene carboxylic acids.展开更多

The acearylene fractions including acenaphthylene, aceanthrylene and cyclopentapyrene were enriched together and named E, which was then transferred to a car- tridge in Isolera-One flash chromatography. Three groups of compounds were eluted out with ethyl acetate/PE mixed solvent (volume ratio 1:9) and named E1, E2 and E3 according to their main components. Acenaphthylene accounted for 78.2% in El, aceanthrylene 71.6% in E2 and cyclopentapyrene 75.9% in E3, respectively. The three groups of acearylenes were purified by Sephadex LH-20 column chromatography with ethanol/cyclohexane mixed solvent (vol- ume ratio 1:4), and then confirmed with nuclear magnetic resonance spectrometer. This method indicates that flash chromatography has a good effect on separating the compounds with a similar structnre after extraction under ultrasonic irradiation.展开更多
