The HPLC analysis was performed on a HIQ sil C18(4.6×250 mm) column with mobile phase of acetonitrilewater (3:7, V:V), flow rate 1 mLmin-1 and UV detective wavelength 266 nm. Extracting 10-hydroxycamptothecin by ultrasonic method from fruits of C. acuminata to prepare samples for analysis was systematically discussed. The optimal extraction condition was carried out by 60% alcohol solution at 60℃ for 50 minutes.展开更多

The optimization conditions of ...A reliable ultrasound-assisted extraction (UAE) method combined with HPLC-UV for quantification of eight active alkaloids in fruits of Macleaya cordata (Willd) R. Br. was developed. The optimization conditions of UAE were obtained by using Box-Behnken design of response surface methodology. Chromatography was carried out using a Kromasil C18 column by gradient elution with 0.1% phosphoric acid aqueous solution for HPLC-UV. All calibration curves showed good linear correlation coefficients (R^2〉0.999 6) and recoveries (from 97.3% to 104.9%) were acceptable. 1,1-diphenyl-2-picrylhydrazyl (DPPH) method was employed to test the antioxidant activity of the extract from the samples. The proposed method was successfully applied to quantifying eight components in nine samples of M.cordata, and significant variations of alkaloid contents and antioxidant aetivity of the samples from different habitats were demonstrated. It presents a powerful proof for the selection of the best sources to extract eight kinds of alkaloids.展开更多

The procedures of ultrasonic extraction and clean-up were optimized for the determination of polycyclic aromatic hydrocarbons (PAHs) in marine sediments. Samples were ultrasonically extracted, and the extracts were pu...The procedures of ultrasonic extraction and clean-up were optimized for the determination of polycyclic aromatic hydrocarbons (PAHs) in marine sediments. Samples were ultrasonically extracted, and the extracts were purified with a miniaturized silica gel chromatographic column and analyzed with high performance liquid chromatography (HPLC) with a fluorescence detector. Ultrasonication with methanol-dichloromethane (2:1, v/v) mixture gave higher extraction efficiency than that with dichloromethane. Among the three elution solvents used in clean-up step, dichloromethane-hexane (2:3, v/v) mixture was the most satisfactory. Under the optimized conditions, the recoveries in the range of 54.82% to 94.70% with RSDs of 3.02% to 23.22% for a spiked blank, and in the range of 61.20% to 127.08% with RSDs of 7.61% to 26.93% for a spiked matrix, were obtained for the 15 PAHs studied, while the recoveries for a NIST standard reference SRM 1941b were in the range of 50.79% to 83.78% with RSDs of 5.24% to 21.38%. The detection limits were between 0.75 ng L-1 and 10.99 ng L-1for different PAHs. A sample from the Jiaozhou Bay area was examined to test the established methods.展开更多

The data acquisition system (DAS) and the signal processing method are presented. The DAS was designed using a muhi-DSP based structure. The signal processing method adopted a well-enhanced split-spectrum processing (SSP) based on weighting algorithm according to statistical times, which was the statistical distribution of the discrete signals selected by the minimization algorithm. Furthermore, the data compressing method was discussed. It assures the mass data to be processed and compressed in real-time. The effectiveness of the method was proved by laboratory experiments. The research provides an excellent base for further development of the ultrasonic pig.展开更多

In this study, a joint spectrogram segmentation and ridge-extraction (J...Ultrasonic guided waves (GWs) can be used to evaluate long bones effectively because of the ability to provide the information of the whole bone. In this study, a joint spectrogram segmentation and ridge-extraction (JSSRE) method was proposed to separate multiple modes in long bones. First, the Gabor time-frequency transform was applied to obtain the spectrogram of multimodal signals. Then, a multi-class image segmentation algorithm was used to find the corresponding region of each mode in the spectrogram, including an improved watershed transform and a region growing procedure. Finally, the ridges were extracted and the time domain signals representing individual modes were reconstructed from these ridges in each region. The validations of this method were discussed by simulated multimodal signals with different signal-to-noise ratios (SNR). The correlation coefficients between the original signals without noise and the reconstructed signals were calculated to analyze the results quantitatively. The results showed that the extracted ridges were in good agreement with generated theoretical dispersion curves, and the reconstructed signals were highly related to the original signals, even under the SNR=3 dB situation.展开更多
