Results A series of systematic tests show that the new type of ER fluids with volume fraction of 30% possesses obvious ER effect. METHODS: The mechanosensory response of mese...AIM: To assess the role of hyperpolarization-activated cyclic nucleotide-gated cation (HCN) channels in regu- lating the excitability of vagal and spinal gut afferents. METHODS: The mechanosensory response of mesen- teric afferent activity was measured in an ex vivo murine jejunum preparation. HCN channel activity was recorded through voltage and current clamp in acutely dissoci- ated dorsal root ganglia (DRG) and nodose ganglia (NG) neurons retrogradely labeled from the small intestine through injection of a fluorescent marker (DiI). The isoforms of HCN channels expressed in DRG and NG neurons were examined by immunohistochemistry. RESULTS: Ramp distension of the small intestine evok- ed biphasic increases in the afferent nerve activity, re- flecting the activation of low- and high-threshold fibers.HCN blocker CsCl (5 mmol/L) preferentially inhibited the responses of low-threshold fibers to distension and showed no significant effects on the high-threshold re- sponses. The effect of CsCI was mimicked by the more selective HCN blocker ZD7288 (10 ~mol/L). In 71.4% of DiI labeled DRG neurons (/7 = 20) and 90.9% of DiI labeled NG neurons (n = 10), an inward current (Ih current) was evoked by hyperpolarization pulses which was fully eliminated by extracellular CsCI. In neurons expressing Ih current, a typical "sag" was observed upon injection of hyperpolarizing current pulses in cur- rent-clamp recordings. CsCI abolished the sag entirely. In some DiI labeled DRG neurons, the Ih current was potentiated by 8-Br-cAMP, which had no effect on the Ih current of DiI labeled NG neurons. Immunohistochem- istry revealed differential expression of HCN isoforms in vagal and spinal afferents, and HCN2 and HCN3 seemed to be the dominant isoform in DRG and NG, respec- tively.CONCLUSION: HCNs differentially regulate the excit- ability of vagal and spinal afferent of murine small in- testine.展开更多
Sum frequency generation(SFG) vibrational spectroscopy has been proven an excellent tool to measure the molecular structures, symmetries and orientations at surfaces/interfaces because of its strong polarization depen...Sum frequency generation(SFG) vibrational spectroscopy has been proven an excellent tool to measure the molecular structures, symmetries and orientations at surfaces/interfaces because of its strong polarization dependence. However, a precise quantitative analysis of SFG spectral intensity and molecular orientation at interfaces must be carefully performed. In this work, we summarized the parameters and factors that are often ignored and illustrated them by evaluating studies of CO adsorption on the(111) facet of platinum(Pt) and palladium(Pd) single crystals at the gas(ultra-high vacuum, UHV)/solid interfaces and methanol(water) adsorption at the air/liquid(solid/liquid) interfaces in the presence of sodium iodide(chloride) salts. To intuitively estimate the influence of incidence angles and refractive indices on the SFG intensity, solely a defined factor of|Fyyz| was discussed, which can be individually separated from the macroscopic second-order non-linear susceptibility χ yyz^(2) term and represents the SSP intensity. Moreover, effects of refractive indices and the molecular hyperpolarizability ratio(R) were discussed in the orientational analysis of interfacial CO and methanol molecules. When IPPP/ISSP was identical, molecules with a larger R had smaller tilting angles(q) on Pt(assuming q < 51°), and CO molecules on Pd would tilt much closer to the surface than they did on Pt. A total internal reflection(TIR) geometry enhanced the SFG intensity, but it also amplified the influence of refractive index on SFG intensity at the solid(silica)/liquid interface. The refractive index and R-value had similar influence on the methanol orientation in the presence of sodium iodide salts at air/liquid and solid/liquid interfaces. This work should provide a guideline for analyzing the orientation of molecules with different R, which are adsorbed on catalysts or located at liquid interfaces involving changes of refractive indices.展开更多
The knowledge of the hyperpolarizabilities of atoms and ions is helpful for the analysis of the high order effects of the frequency shifts in precision spectroscopy experiments. Liu et al. [Phys. Rev. Lett. 114, 2230...The knowledge of the hyperpolarizabilities of atoms and ions is helpful for the analysis of the high order effects of the frequency shifts in precision spectroscopy experiments. Liu et al. [Phys. Rev. Lett. 114, 223001 (2015)] proposed to establish all-optical trapped ion clocks using laser at the magic wavelength for clock transition. To evaluate the high-order frequency shifts in this new scheme of optical clocks, hyperpolarizabilities are needed, but absent. Using the finite field method based on the B-spline basis set and model potentials, we calculated the electric-field-dependent energy shifts of the ground and low-lying excited states in Be+, Mg+, and Ca+ in the field strength range of 0.0-6×10-5 a.u.. The scalar and tensor polarizabilities (α0, α2) and hyperpolarizabilities (Y0,Y2, 74) were deduced. The results of the hyperpolarizabilities for Be+ showed good agreement with the values in literature, implying that the present method can be applied for the effective estimation of the atomic hyperpolarizabilities, which are rarely reported but needed in experiments. The feasibility of optical trapping of Ca+ is discussed, and the contributions of hyperpolarizabilities to the transition frequency shift for Ca+ in the optical dipole trap are estimated using quasi-electrostatic approximation.展开更多

Their physiological functions in invertebrate re...Hyperpolarization-activated, cyclic nucleotide-gated and cation-nonselective ion channels (lh channels, or HCN channels) are known to play important roles in mammals. Their physiological functions in invertebrate remain largely unclear. Here, we re- port our studies with lh channel in Drosophila melanogaster. Drosophila lh channel mutants are found with several defects by behavioral analyses. Their lifespan is reduced, and their chemical sensitivity is shifted. In addition, their length of sleep at light-dark condition is mildly reduced. We generated transgenic flies of lh promoter-driven Gal4 and examined its expression pattern in both larvae and adult flies. Our results suggest that Ih channel may play diverse roles in Drosophila and provide a basis to further expand our understanding of Drosophila Ih channel function in vivo.展开更多

The polarizability average value as is a maximum for the singlet state, while that for the closed-shell is a minimum. The trend in second hyperpolarizability average value yis in good agreement with that for as The yvalues of the singlet and triplet states are, respectively, about 3 and 2 times larger than that of the closed-shell. The order of the first hyperpolarizability total effective value βtot is βot (closed shell) βtot (singlet) 〉 βtot (triplet). The as, βtot, and 7 values of different electronic states obtained using the B3LYP and MP4SDQ methods are close to those obtained using the reliable CCSD method. The nonlinear optical (NLO) properties of two systems isoelectronic with the tetrahydropyrrole diradical-cyclopentane and tetrahydrofuran diradicalsshow that the polarizabilities and hyperpolarizabilities of these systems are all smaller than those of the tetrahydropyrrole diradical in the three electronic states.展开更多
