甲基汞对大鼠生长发育的影响及脂质过氧化作用的研究 被引量:3

目的 :通过对SD大鼠进行腹腔注射氯化甲基汞的暴露方式 ,研究亚急性条件下氯化甲基汞对大鼠生长发育的影响和对大鼠脑组织造成的氧化损伤。方法 :将SD大鼠暴露于不同剂量甲基汞下 ,暴露时间 7天 ,记录大鼠的体重变化情况。 7天后处死 ,... 目的 :通过对SD大鼠进行腹腔注射氯化甲基汞的暴露方式 ,研究亚急性条件下氯化甲基汞对大鼠生长发育的影响和对大鼠脑组织造成的氧化损伤。方法 :将SD大鼠暴露于不同剂量甲基汞下 ,暴露时间 7天 ,记录大鼠的体重变化情况。 7天后处死 ,测定谷胱甘肽过氧化物酶(GSH -px)和超氧化物歧化酶 (SOD)活性及丙二醛 (MDA)含量。结果 :各指标在暴露组与对照组均表现出极显著的差异 (P <0 0 1 )。大鼠的体重呈现不同的变化趋势 ,其中暴露组体重明显受到抑制。结论 :大鼠暴露 7天后产生了明显的脂质过氧化 ,表明自由基尤其是活性氧自由基(ROS)在体内有过量积累。ROS的过量产生会降低脑中神经生长因子 (NGF)的水平 ,从而抑制大鼠的生长发育。 展开更多

Obesity promotes oxidative stress and exacerbates blood-brain barrier disruption after high-intensity exercise 被引量:4

Purpose: The purpose of this study was to investigate the effects of obesity and high-intensity acute exercise on oxidant-antioxidant status,neurotrophic factor expression, and blood-brain barrier(BBB) disruption.Methods: Twenty-four healthy, untrained men(12 non-obese(mean 14.9% body fat) and 12 obese subjects(mean 29.8% body fat)) performed20 min of continuous submaximal aerobic exercise at 85% maximal oxygen consumption. Blood sampling was performed to examine the oxidant-antioxidant status(reactive oxygen species(ROS) and superoxide dismutase(SOD)), neurotrophic factors(brain-derived neurotrophic factor(BDNF) and nerve growth factor(NGF)), and BBB disruption(S100β and neuron-specific enolase) before and after acute exercise.Results: The obese group showed significantly higher pre-exercise serum ROS levels and significantly lower pre-exercise serum SOD levels than the non-obese group(p < 0.05). Serum ROS, SOD, BDNF, NGF, and S100β levels were significantly increased post-exercise compared with pre-exercise levels in both the non-obese and the obese groups(p < 0.05). The obese group showed significantly higher serum ROS, BDNF, NGF,and S100β levels post-exercise compared to the non-obese group(p < 0.05).Conclusion: Our study suggests that episodic vigorous exercise can increase oxidative stress and blood neurotrophic factor levels and induce disruption of the BBB. Moreover, high levels of neurotrophic factor in the blood after exercise in the obese group may be due to BBB disruption,and it is assumed that oxidative stress was the main cause of this BBB disruption. 展开更多

Ethanol extract of mixed algae--spirogyra and ulothrix was investigated to assess the effect on some biochemical parameters in UVB irradiated mice. The dorsal-thoracic region of mice were shaved and exposed to UVB radiation for 24 h for two consecutive weeks. Five groups of ten mice each were conducted: non-irradiated control and irradiated control. Mice treated with commercial cream (S.M.) at a dose of 0.1 mg/cm2 and mice treated with ethanol extract of mixed spirogyra and ulothrix at a dose of 0.1 mg/cm2 or 0.2 mg/cm2. Topical application was performed to all treated mice groups once a day for four consecutive weeks. Results showed significant differences between treated and control groups during the whole period of experiment. The clear potentiality of the ethanol extract was detected through body and skin weight, level of total protein, Superoxide Dismutase (SOD) and Catalase (CAT) activities in skin. However, UVB irradiated mice treated with 0.2 mg/cm2 exhibited the most significant effect when compared to control groups. That may be attributed to anti-inflammatory and anti-oxidant effects of algal extract. 展开更多

OBJEVTIVE: To investigate the antioxidant and hepatoprotective properties of the different extracts Guizhencao (Herba Bidentis Bipinnatae) against liver injury in hyperlipidemia rats. METHODS: The rats were divided into 7 groups, with 10 rats in each. Rats were treated with high-fat diet for 18 weeks besides the normal control group, then rats in both normal control and model groups were received 5 mL/kg -1 · day -1 of saline and those in the positive control group with 2 mg/kg -1 · day -1 of lovastatin. Rats in the positive control group and different Guizhencao (Herba Bidentis Bipinnatae) extracts treatment groups (ethyl acetate extract group, n-hexane extract group, ethanol extract group, and aqueous extract group) were treated with corresponding extract at a concentration of 5 mL/kg -1 · day -1 . After 8 weeks treatment, all rats were sacrificed and total blood samples were collected. Histological analysis of liver was underdone by hematoxylin and eosin. The levels of serum ala-nine aminotransferase (ALT), aspartate aminotransferase (AST), glouse (GLU), cholesterol (TC), triglycerides (TG), low-density lipoprotein-cholesterol (LDL-C) and high-density lipoprotein-cholesterol were measured according to standard procedure using auto-analyzer. The superoxide dismutase (SOD) and malondialdehyde (MDA) levels in liver were ananlyzed by procedure instruction. RESULTS: The histopathological analysis implied that the administration of Guizhencao (HerbaBidentis Bipinnatae) extracts resulted in hepatoprotective role compared with that of the model group. In addition, the high-fat diet caused a remarkable increase of ALT, AST, GLU, TC, TG, LDL-C and MDA levels. A decline in HDL-C and SOD concentrations and a reversal of effects were observed in different Guizhencao (Herba Bidentis Bipinnatae) extracts groups, especially in the aqueous extract and ethanol extract groups. CONCLUSION: The different extracts of Guizhencao (Herba Bidentis Bipinnatae) can play a protecting role against liver injury in hyperlipidemia rats maybe through decreasing ALT, AST, GLU, TC, TG, LDL-C and MDA levels and enhancing the liver anti-oxidative ability. 展开更多
