小体积肝移植后再生肝脏中受体来源骨髓干细胞跨分化的实验研究

目的研究小体积肝移植后受体来源的造血干细胞跨分化现象及影响因素。方法采用性别错配的大鼠不同体积肝脏移植,动态观察移植肝体积的大小和缺血损伤对术后骨髓干细胞向移植物内肝细胞跨分化促肝脏再生过程的影响。结果全肝移植组Y-染... 目的研究小体积肝移植后受体来源的造血干细胞跨分化现象及影响因素。方法采用性别错配的大鼠不同体积肝脏移植,动态观察移植肝体积的大小和缺血损伤对术后骨髓干细胞向移植物内肝细胞跨分化促肝脏再生过程的影响。结果全肝移植组Y-染色体肝细胞阳性率为0.66%,而60%和30%肝移植组Y-染色体肝细胞阳性率分别增加到1.89%和3.78%,3组间差异均有显著性(P<0.05)。延长冷缺血时间使得肝内Y-染色体阳性肝细胞率减少到0.73%,与1h缺血时间组差异有显著性(P<0.05)。结论减体肝移植能增强HSCs在肝脏内向肝细胞跨分化。而延长冷缺血时间明显降低HSCs在肝内的跨分化。 展开更多

目的:研究Sox2对于人骨髓间充质干细胞(hBMSCs)向神经元跨分化能力的影响。方法:利用Western Blot检测hBMSCs中Sox2、Oct4和c-Myc的表达,将携带Sox2和rhGFP编码序列的Tet-On慢病毒感染hBMSCs,通过多西环素(Dox)处理10 d诱导Sox2和rhGFP... 目的:研究Sox2对于人骨髓间充质干细胞(hBMSCs)向神经元跨分化能力的影响。方法:利用Western Blot检测hBMSCs中Sox2、Oct4和c-Myc的表达,将携带Sox2和rhGFP编码序列的Tet-On慢病毒感染hBMSCs,通过多西环素(Dox)处理10 d诱导Sox2和rhGFP表达;利用神经诱导培养基诱导感染慢病毒的hBMSCs分化,通过免疫荧光染色技术检测神经元标记物Tuj1的表达;利用Western Blot技术检测Smad 2/3信号通路蛋白在hBMSCs向神经元分化过程中的表达水平。结果:Western Blot结果显示hBMSCs不仅表达Sox2和Oct4,同时还表达c-Myc;光镜下发现Sox2的过表达促进hBMSCs的形态向神经元样细胞转化,同时通过免疫荧光染色发现Tuj1的表达水平增加; Western Blot证实Sox2基因过表达引起hBMSCs中磷酸化Smad 2/3的表达水平降低。结论:Sox2可通过Smad 2/3信号通路增强人骨髓间充质干细胞向神经元分化。 展开更多

Activation ofNotch inhibits cellular differentiation, and abnormality ofthe Notch pathway leads to premature neuronal differenti-ation, the lack of some cell types, and severe defects of tis-sue morphogenesis. Recent data demonstrate that Notchfails to inhibit cellular differentiation in the absence of thebHLH genes Hes1 and He5, which functionally afitago-nize the neuronal bHLH genes such as theh1- These re-sults indicate that the two Hes genes are essential effecorsfor the Notch pathway and that neuronal differentiation iscontrolled by the pathway "Notch-+He1/Hs5,Mish1". 展开更多

This paper discusses the interconnection between the Daoism and vagueness. On one hand, it summarizes the viewpoints of vagueness proposed by numerous scholars, displaying the function of such expressions, stressing the affirmative role; on the other hand, it launches the term into the Chinese first philosophy Daoism, revealing the links of these two good brothers. Then, the paper argues about the insights of the logics put forward by the Chinese and the Western professionals, drawing on the conclusion that the Daoism is concerned with the Dao of vagueness. It is a fuzzy set as well as logics of dialectics with reversion reasoning. The two high context terms resemble the cultures of the East and West. The integration can contribute to the success in business communication. No universal way can guide the business interaction; our heart and soul can soften all the discrepancy and conflict. Harmony can be achieved through a variety of Daoes, each cultural group bearing their own vagueness, silence, rhetoric, and reasoning. The logics of the Dao in business communication are in your eyes, your mouth, your hands, and your heart and soul in particular. 展开更多

In this article, I seek to stre... Constitutional Patriotism is a new form of identity. It addresses the national component of identity formations in order to transform them in light of universal human rights principles. In this article, I seek to strengthen this theory left underdeveloped by Habermas. To do so, I use the idea of moral development which Habermas borrowed from Kohlberg. I argue that Constitutional Patriotism is the missing link in Habermas's reading of Kohlberg. I complement Kohlberg's reading of moral consciousness with the psychoanalytic idea of individuation. Communication, language, and autonomy all fall into their places in this interdisciplinary puzzle of Constitutional Patriotism spanning over the cultural terrain. This article takes part of the broader project of Constitutional Patriotism here only focusing on the notion of the selfhood. 展开更多

Economic Analysis of CO2 Emission Abatement Applying a Dynamic CGE Model with Endogenous Technological Change: Impacts of the Time Horizon

We evaluate economic and environmental impacts of climate change mitigation in a country scale considering various time horizons in the analysis applying a single-country dynamic computable general equilibrium model with endogenous technological change. Although there is a possibility that Gross Domestic Product (GDP) becomes larger for the abatement cases than the baseline case in the earlier years, it tends to be lower than that in the later years. The longer the time horizon and/or the more severe the abatement, the larger the negative impacts will be. When subsidizing R&D investment, increase in GDP compared to the baseline case is realized in the middle of the time horizon, and the larger increase tends to be observed for the longer-term cases. These results would be due to technological change induced by the subsidies and emission abatement. Environmental indicators are also improved. We showed that the results were influenced by the target time horizon when using an intertemporal dynamic model. 展开更多

In this paper,exergy analysis method is developed to assess a Rankine cycle system,by using supercritical CO2 as working fluid and powered by solar energy.The proposed system consists of evacuated solar collectors,throttling valve,high-temperature heat exchanger,low-temperature heat exchanger,and feed pump.The system is designed for utilize evacuated solar collectors to convert solar energy into mechanical energy and hence electricity.In order to investigate and estimate exergy performance of this system,the energy,entropy,exergy balances are developed for the components.The exergy destructions and exergy efficiency values of the system components are also determined.The results indicate that solar collector and high temperature heat exchanger which have low exergy efficiencies contribute the largest share to system irreversibility and should be the optimization design focus to improve system exergy effectiveness.Further,exergy analysis is a useful tool in this regard as it permits the performance of each process to be assessed and losses to be quantified.Exergy analysis results can be used in design,optimization,and improvement efforts. 展开更多
