AIM: To investigate the correlation between uridine diphosphate glucuronosyl transferase 1A1 (UGT1A1) gene polymorphisms and irinotecan-associated side effects and parameters of drug efficacy in patients with metastat...AIM: To investigate the correlation between uridine diphosphate glucuronosyl transferase 1A1 (UGT1A1) gene polymorphisms and irinotecan-associated side effects and parameters of drug efficacy in patients with metastatic colorectal cancer (mCRC) receiving a low-dose weekly irinotecan chemotherapeutic regimen. METHODS: Genotypes were retrospectively evaluated by gene scan analysis on the ABI 310 sequencer of the TATAA box in the promoter region of the UGT1A1 gene in blood samples from 105 patients who had received 1st line irinotecan-based chemotherapy for mCRC. RESULTS: The distribution of the genotypes was as follows: wild type genotype (WT) (6/6 ) 39.0%, heterozygous genotype (6/7) 49.5%, and homozygous genotype (7/7) 9.5%. The overall response rate (OR) was similar between patients carrying the (6/7, 7/7) or the WT genotype (6/6) (44.3% vs 43.2%, P = 0.75). Neither time to progression [(TTP) 8.1 vs 8.2 mo, P = 0.97] nor overall survival [(OS) 21.2 vs 18.9 mo, P = 0.73] differed significantly in patients who carried the(6/6) when compared to the (6/7, 7/7) genotype. No significant differences in toxicity were observed: Grade 3 and 4 delayed diarrhoea [(6/7, 7/7) vs (6/6); 13.0% vs 6.2%, P =0.08], treatment delays [(6/7, 7/7) vs (6/6); 25.1% vs 19.3%, P = 0.24] or dose reductions [(6/7, 7/7) vs (6/6); 21.5% vs 27.2%, P = 0.07].CONCLUSION: This analysis demonstrates the non-significant influence of the UGT1A1 gene polymorphism on efficacy and rate of irinotecan-associated toxicity in mCRC patients receiving low-dose irinotecan based chemotherapy.展开更多

Important features of the leader such and ability to assist in developing and accommodating the implementation of knowledge management programs, learning organization concepts and innovation protocols are discussed in this paper. This paper demonstrates that shifting the organization to become a knowledge based and then to be learning organization and finally to become innovative company could involve some unique attributes of a transformation leadership. In that regards, the paper also demonstrates that organizations need first to create, capture, transfer, and mobilize knowledge before it can be used for learning and then for innovation. The paper will present a method of a studying how successful innovation leaders of companies could found themselves acting in three roles namely: knowledge leader, learning leader and then innovation leader.展开更多

The concepts and quantitative expressions of the steering road feel, steering sensibility, and steering operation stability are introduced. Based on quality engineering theory, the optimization algorithm is proposed by integrating the Monte Carlo descriptive sampling, elitist non-dominated sorting genetic algorithm (NSGA-II) and 6-sigma design method. With the steering road feel and the steering portability as optimization targets, the system parameters are optimized by the proposed optimization algorithm. The simulation results show that the system optimized based on quality engineering theory can improve the steering road feel, guarantee steering stability and steering portability and thus provide a theoretical basis for the design and optimization of the novel electric power steering system.展开更多

Unlike other researches, this paper analyzes the conversion process of carbon element in the ocean from the qualitative viewpoint. There are many complex roles in the ocean carbon cycle, and it is hard to represent the case that an entity plays different role in different environment. An ontology technology Hozo role theory developed by Osaka University Mizoguchi Laboratory is proposed as a solution. The basic concepts and representation mode of Hozo role theory is introduced. The conversion process of ocean carbon cycle is abstracted and an ontology model using Hozo role theory is proposed. Instead of comprehensive common ontology construction method, we propose our own ontology development steps. Then an ontology about ocean carbon cycle is built in order to describe and share the basic knowledge of ocean carbon cycle. A knowledge base of material circulation is proposed based on the ontology. Its construction framework is described and some knowledge base query examples are also illustrated. Conclusions show that the role theory can effectively solve the problem of multirole description in ocean carbon cycle, and knowledge reasoning based on ontology is also effective.展开更多

Although codified knowledge and its capture is commonplace, tacit knowledge has up until recent years proved elusive in its inclusion within the organization's knowledge base. In this paper, the conversion process has been dealt with the help of protocols. It is learned that while structure may significantly improve the tacit to explicit conversion process, it also may matter how the structure is employed. It is anticipated that eventual successful modeling of such knowledge will ultimately be undertaken in several organizations with a view to improving information capture and transference.展开更多
