METHODS: A 63-nucleotide (nt) oligonucleotide encoding K-rasval12 and specific siRNA were introduced into pSilencer 3.1-H1, then the H1-RNA promoter and siRNA coding insert were subcloned into pAdTrack to get plasmid pAdTrackH1-Avasval12. After homologous recombination in bacteria and transfections of such plasmids into a mammalian packaging cell line 293, siRNA expressing adenovirus Adh1-K-rasval12 was obtained. Stable suppression of K-rasval12 was detected by Northern blot and Western blot. Apoptosis in Panc-1 cells was detected by flow cytometry. RESULTS: We obtained adenovirus AdHl-K-rasval12 carrying the pSilencer 3.1-H1 cassette, which could mediate gene silencing. Through siRNA targeted K-rasval12, the oncogenic phenotype of cancer cells was reversed. Flow cytometry showed that apoptotic index of Panc-1 cells was significantly higher in the AdH1-K-rasval12-treatment group (18.70% at 72 h post-infection, 49.55% at 96 h post-infection) compared to the control groups (3.47%, 3.98% at 72 and 96 h post-infection of AdH1-empty, respectively; 4.21%, 3.78% at 72 and 96 h post-infection of AdHl-p53, respectively) (P<0.05). CONCLUSION: These results demonstrate that adenoviral vectors can be used to mediate RNA interference (RNAi) to induce persistent loss of functional phenotypes. In gene therapy, the selective down-regulation of only the mutant version of a gene allows for highly specific effects on tumor cells, while leaving the normal cells untouched. In addition, the apoptosis of pancreatic cancer cell line Panc-1 can be induced after AdH1-K-rasval12 infection. This kind of adenovirus based on RNAi might be a promising vector for cancer therapy.展开更多

The effects of the DM interaction on specific heat, susceptibility, and magnetization are simulated. The critical temperature of transitions is determined by the so-called Binder cumulant and the susceptibility of finite-size scaling. We find that the chiral Z2 symmetry reduced by the DM interactions plays an important role in a two-dimensional XY spin system, typically, the critical temperature is sensitive to weak DM spin couplings.展开更多

Despite this, sporadic efforts have been made to develop regeneration systems in cowpea during the past 40 years. This review presents the considerable progress on cowpea regeneration (organogenesis and embryogenesis) and especially focuses on the regeneration mode of organogenesis, including highlights of the effect of genotypes, explants, medium and plant hormones used in tissue culture. We introduce a completely different method to calculate the evolution of a spin interacting with a sufficient large spin bath,especially suitable for treating the central spin model in a quantum dot(QD).With only an a...We introduce a completely different method to calculate the evolution of a spin interacting with a sufficient large spin bath,especially suitable for treating the central spin model in a quantum dot(QD).With only an approximation on the envelope of central spin,the symmetry can be exploited to reduce a huge Hilbert space which cannot be calculated with computers to many small ones which can be solved exactly.This method can be used to calculate spin-bath evolution for a spin bath containing many(say,1000)spins,without a perturbative limit such as strong magnetic field condition,and works for long-time regime with sufficient accuracy.As the spin-bath evolution can be calculated for a wide range of time and magnetic field,an optimal dynamic of spin flip-flop can be found,and more sophisticated approaches to achieve extremely high polarization of nuclear spins in a QD could be developed.展开更多

Although Type Ia supernovae (SNe Ia) play very important roles in many astrophysical fields, the exact nature of the progenitors of SNe Ia is still unclear. At present, the single degenerate (SD) model is a very likel...Although Type Ia supernovae (SNe Ia) play very important roles in many astrophysical fields, the exact nature of the progenitors of SNe Ia is still unclear. At present, the single degenerate (SD) model is a very likely one. Following the comprehensive SD model developed by Meng & Yang (2010), we show the properties of SNe Ia companions at the moment of the supernova explosion. The results may provide help in searching for companion stars in supernova remnants. We compared our results with the companion candidate Tycho G of Tycho's supernova and found that integral properties of the star (mass, space velocity, radius, luminosity and effective temperature) are all consistent with those predicted from our SD model with the exception of the rotational velocity. If Tycho G was the companion star of Tycho's supernova, an interaction between supernova ejecta and the rotational companion might be a key factor to solve the confliction, and then it could be encouraged to do a detailed numerical simulation about the interaction.展开更多
