转移生长因子基因单核苷酸多态性与肺癌易感性的关系研究

目的研究转移生长因子(TGF)基因上rsl800469(TGFl-509C/T)和rsl982073(TGFl+869T/C)位点基因单核苷酸多态性与肺癌易感性之间的关系。方法选择原发性肺癌患者193例(病例组)和211例同期就诊的非肿瘤患者(对照组)。应用实时... 目的研究转移生长因子(TGF)基因上rsl800469(TGFl-509C/T)和rsl982073(TGFl+869T/C)位点基因单核苷酸多态性与肺癌易感性之间的关系。方法选择原发性肺癌患者193例(病例组)和211例同期就诊的非肿瘤患者(对照组)。应用实时荧光定量逆转录聚合酶链反应方法进行TGF基因单核苷酸多态性的基因分型,比较两组人口学特征、相关危险因素暴露及基因单核苷酸多态性的差异。结果对照组在TGFl+869T/C和TGFl-509C/T的基因型分布频率经拟合优度r检验,符合Hardy-Weinberg遗传平衡定律(P〉0.05)。TGFl+869T/C的基因型TT、TC、CC分布频率在病例组分别是16.1%(31/193)、55.4%(107/193)、28.5%(55/193),在对照组的分布频率分别是22.3%(47/211)、50.2%(106/211)、27.5%(58/211),差异无统计学意义(P〉0.05)。TGFl-509C/T的基因型TT、TC、CC分布频率在病例组分别是28.0%(54/193)、56.0%(108/193)、16.1%(31/193),在对照组的分布频率分别是27.0%(57/211)、51.2%(108/211)、21.8%(46/211),差异无统计学意义(P〉0.05)。T等位基因携带者患肺癌的风险与c等位基因携带者患肺癌的风险相比差异无统计学意义(P〉0.05)。结论北方汉族人群中TGF基因单核苷酸多态性与肺癌易感性无关。 展开更多

AIM: To elucidate the possible difference in two promoter polymorphisms of the transforming growth factor-β1 (TGF-β1) gene (-800G > A, -509C > T) between ulcerative colitis (UC) patients and normal subjects.METHODS: A total of 155 patients with established ulcerative colitis and 139 normal subjects were selected as controls. Two single nucleotide polymorphisms within the promoter region of TGF-β1 gene (-509C > T and -800G > A) were genotyped using PCR-RFLP. RESULTS: There was a statistically significant difference in genotype and allele frequency distributions between UC patients and controls for the -800G > A polymorphism of the TGF-β1 gene (P < 0.05). The frequency of the TGF-β1 gene polymorphism at position -800 showed that the AA genotype and the allele A frequencies significantly differed between the patients and healthy controls (P < 0.05). At position -509, there was no statically significant difference in genotype and allele frequency between the patients and control subjects.CONCLUSION: The results of our study indicate that there is a significant difference in both allele and genotype frequency at position -800G > A of TGF-β1 gene promoter between Iranian patients with UC and normal subjects. 展开更多

Despite of the relatively simple signal transduction pathway, many modulators have been found to contribute to a tight regulation of this pathway in a variety of mechanisms.
