We report the finding of a retrograde apoptotic signal in axons that is suppressed by local NGF signaling. NGF withdrawal from distal axons alone was sufficient to activate the pro-apoptotic transcription factor, c-jun, in the cell bodies. Providing NGF directly to cell bodies, thereby restoring a source of NGF-induced survival signals, could not prevent c-jun activation caused by NGF withdrawal from the distal axons. This is evidence that c-jun is not activated due to loss of survival signals at the cell bodies. Moreover, blocking axonal transport with colchicine inhibited c-jun activation caused by NGF deprivation suggesting that a retrogradely transported pro-apoptotic signal, rather than loss of a retrogradely transported survival signal, caused c-jun activation. Additional experiments showed that activation of c-jun, pro-caspase-3 cleavage, and apoptosis were blocked by the protein kinase C inhibitors, rottlerin and chelerythrine, only when applied to distal axons suggesting that they block the axon-specific pro-apoptotic signal. The rottlerin-sensitive mechanism was found to regulate glyco- gen synthase kinase 3 (GSK3) activity. The effect of siRNA knockdown, and pharmacological inhibition of GSK3 suggests that GSK3 is required for apoptosis caused by NGF deprivation and may function as a retrograde carrier of the axon apoptotic signal. The existence of a retrograde death signaling system in axons that is suppressed by neurotro- phins has broad implications for neurodevelopment and for discovering treatments for neurodegenerative diseases and neurotrauma.展开更多

Peak amplitude of the...In this study, dynamic characteristics of the small base isolation system using new friction bearings are investigated by excitation experiment, and compared to other one using previous bearings. Peak amplitude of the acceleration response waves on the small base isolation system is decreased to about 10%-25% compared to the input waves. Also root mean square amplitude is decreased to about 10%-40%. In case of the ball embedded a cylindrical sponge, the new bearing, the damping ratio increases with increasing width of the cylindrical sponge. The natural frequency does not change. On the other hand, in case of the marble plate that is previous bearing, the damping ratio increases with increasing curvature radius of the marble plate, the natural frequency also increases. Therefore, the small base isolation system using new friction bearing provides better performance. The responses of the base isolation system indicate nonlinier effects by friction force.展开更多

Using wavelet operations of multi-scale discrete analysis t...Through the 5-channel SWAES digital full waveform AE detector, the paper dealt with the fracture process of coal and rock samples under uniaxial compression. Using wavelet operations of multi-scale discrete analysis the pulses of a particular time period (points) and the space domain signal by numerical method were gotten, and the paper concluded that the signal singularity in load rupture had closely relations with fracture and uniaxial compression. The detected position and the actual breaking point only differed at one sample point, the relative error was 6.82%, and there was no accumulative error. Thus it provided an effective method to solve the problem of instability analysis of the signal singularity detection and coal-rock compression failure in the whole process.展开更多
Growth is a polygenic trait that is under the influence of multiple physiological pathways regulating energy metabolism and muscle growth.Among the possible growth-regulating pathways in vertebrates,components of the ...Growth is a polygenic trait that is under the influence of multiple physiological pathways regulating energy metabolism and muscle growth.Among the possible growth-regulating pathways in vertebrates,components of the somatotropic axis are thought to have the greatest influence.There is growing body of literature focusing on the somatotropic axis and its role regulating growth in fish.This includes research into growth hormone,upstream hypothalamic hormones,insulin-like growth factors,and downstream signaling molecules.Many of these signals have both somatic effects stimulating the growth of tissues and metabolic effects that play a role in nutrient metabolism.Signals of other endocrine axes exhibit profound effects on the function of the somatotropic axis in vivo.In this review we highlight recent advances in our understanding of the teleost fish endocrine somatotropic axis,including emerging research using genetic modified models.These studies have revealed new aspects and challenges associated with regulation of the important steps of somatic growth.展开更多
High performance force sensors often encounter the conflicting requirements of high resolution and large measurement range.To address this problem,this paper presents a conceptual design of a novel uniaxial force sens...High performance force sensors often encounter the conflicting requirements of high resolution and large measurement range.To address this problem,this paper presents a conceptual design of a novel uniaxial force sensor with large range and dual-stage force resolutions which enables us to measure forces within a wide range with satisfied resolutions.The newly developed force sensor features an aluminum alloy body with a probe to transfer external forces into the sensing element.It employs an optical linear encoder to detect the displacement of the sensing body.This sensing scheme may immunize outside electromagnetic noises and therefore enhance the performance of the sensor thanks to its digital signal output.In this paper,an accurate,analytical model for calculating the static stiffness and dynamics of the system was developed by using pseudo-rigid-body-model(PRBM)methodology.To optimize the design,finite element simulations were conducted.After a prototype sensor was fabricated,preliminary characterization tests were carried out to verify the accuracy of the theoretical model and demonstrate the effectiveness of the design.The experiment results indicate that the structure of the new sensor is compact,and it has the ability to measure both micro range and macro range forces within one setup,meanwhile keeps very fine resolutions.展开更多
