循环轴向压缩应力增强基质依赖型组织工程骨骨再生能力的研究

目的研究循环轴向压缩应力(CACS)对基质依赖型组织工程骨(M-TEB)骨再生能力的影响。方法首先,在构建静态M-TEB过程中加载CACS,得到动态M-TEB;然后,对各组M-TEB进行表征,并构建动物模型来评价其骨再生能力;最后,通过转录组测序探究CACS... 目的研究循环轴向压缩应力(CACS)对基质依赖型组织工程骨(M-TEB)骨再生能力的影响。方法首先,在构建静态M-TEB过程中加载CACS,得到动态M-TEB;然后,对各组M-TEB进行表征,并构建动物模型来评价其骨再生能力;最后,通过转录组测序探究CACS对骨髓间充质干细胞(BMSCs)基因表达的影响,并研究M-TEB来源条件培养基对内皮祖细胞(EPCs)迁移、增殖和对BMSCs成骨分化的影响。结果①动态M-TEB组在骨缺损区有更多新骨生成,且骨体积分数和骨密度均显著优于假手术组和静态M-TEB组(P均<0.01);②前30个有显著差异(q<0.01)的基因本体论(GO)术语主要涉及MAPK通路、细胞凋亡和血管生成等,且差异基因VEGFA在这些GO术语中出现频次最高(28次);③相比静态M-TEB来源条件培养基,动态M-TEB来源条件培养基具有更强的促进EPCs迁移、增殖和BMSCs成骨分化的作用(P均<0.05),阻断实验证实VEGFA在其中发挥重要功能。结论CACS促进BMSCs表达和分泌VEGFA,提高动态M-TEB中VEGFA浓度,最终增强其在大鼠股骨缺损模型中的骨再生能力。 展开更多

耗能型防落梁装置是由普通拉索式防落梁装置改进而来的,其主要吸能部件为吸能控制管,由扩径管和铝蜂窝缓冲器组成。为了给铝蜂窝缓冲器的设计与优化提供可靠的理论依据,建立了铝蜂窝轴向压缩简化模型,对其轴向压缩应力进行了分析、推导... 耗能型防落梁装置是由普通拉索式防落梁装置改进而来的,其主要吸能部件为吸能控制管,由扩径管和铝蜂窝缓冲器组成。为了给铝蜂窝缓冲器的设计与优化提供可靠的理论依据,建立了铝蜂窝轴向压缩简化模型,对其轴向压缩应力进行了分析、推导并采用有限元模拟和借鉴相关文献试验数据进行了验证,同时分析了加载速率对铝蜂窝材料吸能特性的影响。此外分析了铝蜂窝芯铝箔厚度和蜂窝边长对其吸能特性的影响,并通过桥梁工程实例来分析和评价铝蜂窝缓冲器在吸能控制管中的吸能作用。研究结果表明:铝蜂窝轴向压缩简化模型精度较高,理论计算值相对于有限元计算值的偏差平均值为4.53%,相对于文献试验值的偏差平均值为6.47%;随着加载速率的提高,铝蜂窝材料的单位体积吸能值、初始峰值应力和屈服平均应力均有所提高;在铝蜂窝芯铝箔厚度或蜂窝边长一定的情况下,两者之比β值越大,铝蜂窝缓冲器吸能性能越好;配置了铝蜂窝缓冲器的吸能控制管,其总吸能值平均增大率为18.56%,最大增大率为29.51%,吸能性能提升最大可达近30%,在消耗地震能量方面将发挥显著的作用。 展开更多

In order to investigate the size effect and other effects on the stress-strain relationship of confined concrete, 42 specimens with different sizes and section shapes were placed under axial compression loading. Effects of key parameters such as size of specimens, tie configuration, transverse reinforcement ratio, and concrete cover were studied. The results show that for specimens with the same configuration and the same volumetric ratio of the transverse reinforcement, along with the increasing specimen size, the peak stress, peak strain and deformation of the post-peak show a down trend, however, the volumetric ratio of the transverse reinforcement is lowered, the decreasing of the peak stress is accelerated, but the decreasing of the deformation is slow down. For specimens with the same volumetric ratio but different configurations of transverse reinforcement, though the transverse reinforcement configuration becomes more complicated, the peak stress of the large size specimen does not improve more than that of the small size. However, the deformation occurs before the stress declines to 85% of peak stress, and the improvement with the grid pattern tie configuration is much greater due to size effect. 展开更多

Using an MTS816.03 test system and self-designed seepage apparatus, seepage tests of saturated broken rocks were conducted, and the influence of lithology, axial stress, grain size distribution and loading rate on seepage characteristics was analyzed. The results show that: (1) Under the same axial stress (12 MPa), the permeability of different lithologic samples increases in the order: gangue 〈 mudstone 〈 sandstone 〈 limestone. The permeability of gangue is 3 magnitudes lower than that of limestone. The absolute value of the non-Darcy coefficient β increases in the order: limestone 〈 sandstone 〈 mudstone 〈 gangue. The non-Darcy coefficient β of limestone, which is positive, is 5 magnitudes lower than that of gangue. (2) With increasing axial stress, the permeability of saturated broken sandstone decreases, and the absolute value of the non-Darcy coefficient β increases. After the axial stress exceeds 12 MPa, the curves of permeability and non-Darcy coefficient β all tend to be stable. (3) With increasing Talbol power exponent, the permeability increases, and the absolute value of the non-Darcy coefficient β decreases. (4) With increasing loading, the permeability increases, and the absolute value of the non-Darcy coefficient β decreases. When the loading rate is 0.5 kN/s, the non-Darcy coefficient β is positive. 展开更多

Experimental investigation was conducted to characterize the responses of high performance concrete(HPC) subjected to multiaxial compressive stresses. The HPC specimens were prepared with three different mix proportions, which corresponds to three different uniaxial compressive strengths. The cubic specimens with size of 100 mm for each edge were tested with servo-hydraulic actuators at different stress ratios. The principal stresses and strains of the specimens were recorded, and the failure of the cubic specimens under various stress states was examined. The experimental results indicated that the stress states and stress ratios had significant influence on the strength and deformation of HPC under biaxial and triaxial compression, especially under triaxial compression. Failure criteria were proposed for the HPC specimens under biaxial and triaxial compressive loading. The test results provided a valuable reference for obtaining multi-axial constitutive law for HPC. 展开更多
