METHODS: Quantitative PCR amplification was performed on four microsatellite markers and trefoil factor family genes (TFF1, TFF2, and TFF3) using a standard curve and SYBR Green I dye method. The relative copy number was determined by DNA copy number of tested locus to reference locus. The relative copy number was interpreted as deletion or amplification by comparison with normal reference range. Associations between allelic imbalance and clinicopathological parameters of CCA patients were evaluated by χ^2-tests. Kaplan-Meier method was used to analyze survival. RESULTS: The frequencies of amplification at D21S1890, D21S1893, and TFF3 were 32.5%, 30.0%, and 28.7%, respectively. Patients who had amplification at regions covering D21S1893, D21S1890, and TFF showed poor prognosis, whereas patients who had deletion showed favorable prognosis (mean: 51.7 wk vs 124.82 wk, P = 0.012). Multivariate Cox regression analysis revealed that amplification of D21S1893, D21S1890 and TFF, blood vessel invasion, and staging were associated with poor prognosis. CONCLUSION: D21S1893-D21S1890 region may harbor candidate genes especially TFF and serine protease family, which might be involved in tumor invasion and metastasis contributing to poor survival. The amplification in this region may be used as a prognostic marker in the treatment of CCA patients.展开更多

In this...The heterotrimeric guanine nucleotide-binding protein (G-protein) has been demonstrated to mediate various signaling pathways in plants. However, its role in phytochrome A (phyA) signaling remains elusive. In this study, we discover a new phyA-mediated phenotype designated far-red irradiation (FR) preconditioned cell death, which occurs only in the hypocotyls of FR-grown seedlings following exposure to white light (WL). The cell death is mitigated in the Gα mutant gpal but aggravated in the Gβ mutant agbl in comparison with the wild type (WT), indicative of antagonistic roles of GPA1 and AGBI in the phyA-mediated cell-death pathway. Further investigation indicates that FR-induced accumulation of nonphotoconvertible protochlorophyllide (Pchlide^633), which generates reactive oxygen species (ROS) on exposure to WL, is required for FR-preconditioned cell death. Moreover, ROS is mainly detected in chloroplasts using the fluorescent probe. Interestingly, the application of H2O2 to dark-grown seedlings results in a phenotype similar to FR-preconditioned cell death. This reveals that ROS is a critical mediator for the ceil death. In addition, we observe that agb1 is more sensitive to H2O2 than WT seedlings, indicating that the G-protein may also modify the sensitivity of the seedlings to ROS stress. Taking these results together, we infer that the G-protein may be involved in the phyA signaling pathway to regulate FR-preconditioned cell death ofArabidopsis hypocotyls. A possible mechanism underlying the involvement of the G-protein in phyA signaling is discussed in this study.展开更多

Such interactions produce an Electric Dipole Moment (EDM) for the top quark at one loop level. The dusty gas is taken to be a mixture of small solid particles and pe...A self-similar flow behind a cylindrical shock wave is studied under the action of monochromatic radiation in a rotational axisymmetric dusty gas. The dusty gas is taken to be a mixture of small solid particles and perfect gas,and solid particles are continuously distributed in the mixture. The similarity solutions are obtained and the effects of the variation of the radiation parameter, the ratio of the density of solid particles to the initial density of the gas, the mass concentration of solid particles in the mixture and the index for the time dependent energy law are investigated.It is observed that an increase in the radiation parameter has decaying effect on the shock waves; whereas the shock strength increases with an increase in the ratio of the density of solid particles to the initial density of the gas or the index for the time dependent energy law. Also, it is found that an increase in the radiation parameter has effect to decrease the flow variables except the density and the azimuthal component of fluid velocity. A comparison is also made between rotating and non-rotating cases.展开更多
The present paper reports a comparison of experimental SPL spectral data related to the tonal noise generated by axial-flow fans.A nine blade rotor has been operated at free discharge conditions and in four geometrica...The present paper reports a comparison of experimental SPL spectral data related to the tonal noise generated by axial-flow fans.A nine blade rotor has been operated at free discharge conditions and in four geometrical configurations in which different kinds of tonal noise generating mechanisms are present:large-scale inlet turbulent structures,tip-gap flow,turbulent wakes,and rotor-stator interaction.The measurements have been taken in a hemi-anechoic chamber at constant rotational speed and,in order to vary the acoustic source strength,during low angular acceleration,linear speed ramps.In order to avoid erroneous quantitative evaluations if the acoustic propagation effects are not considered,the acoustic response functions of the different test configurations have been computed by means of the spectral decomposition method.Then,the properties of the tonal noise generating mechanisms have been studied.To this aim,the constant-Strouhal number SPL,obtained by means of measurements taken during the speed ramps,have been compared with the propagation function.Finally,the analysis of the phase of the acoustic pressure has allowed to distinguish between random and deterministic tonal noise generating mechanisms and to collect information about the presence of important propagation effects.展开更多

We here report a novel bacterial artificial chromosome (BAC) transgenic mouse line that expresses Class Ⅲ β-tubulin fused to mCherry, an improved monomeric red fluorescent protein, for the visualization of microtubules during neuronal development. A BAC containing Tubb3 gene was modified by insertion of mCherry complementary DNA downstream of Tubb3 coding sequence via homologous recombination, mCherry fusion protein was expressed in the nervous system and testis of the transgenic animal, and the fluorescent signal was observed in the neurons that located in the olfactory bulb, cerebral cortex, hippocampal formation, cerebellum, as well as the retina. Besides, Tubb3-mCherry fusion protein mainly distributed in neurites and colocalized with endogenous Class Ⅲ β-tubulin The fusion protein labels Purkinje cell dendrites during cerebellar circuit formation. Therefore, this transgenic line might be a novel tool for scientific community to study neuronal development both in vitro and in vivo.展开更多
